AbstractRecent advances in computer sciences have resulted in a significant improvement in reservoir modeling, which is an important stage in studying and comprehending reservoir geometry and properties. It enables the collection of various types of activities such as seismic, geological, and geophysical aspects in a single container to facilitate the characterization of reservoir continuity and homogeneity. The main goal of this work is to build a three-dimensional reservoir model of the Abu Roash G reservoir in the Hamra oil field with enough detail to represent both vertical and lateral reservoir heterogeneity at the well, multi-well, and field scales. The Late Cenomanian Abu Roash G Member is the main reservoir in the Hamra oil field. It is composed mainly of shale, carbonate and some streaks of sandstone, these streaks are shaly in some parts. Conducting the 3D geostatic model begins with the interpretation of seismic data to detect reflectors and horizons, as well as fault picking to explain the structural framework and frequently delineate the container style with proposed limitations to construct the structural model. The lithology and physical properties of Abu Roash G reservoir rock, including total and effective porosity and fluid saturation, were determined using well log data from four wells in the Hamra field. The constructed 3D geological model of the Abu Roash G has showed that the petrophysical parameters are controlled by the facies distribution and structure elements, whereas properties are the central part to the northern side of the deltaic environment than the other sides of the same environment. The model will be useful in displaying the reservoir community and indicating prospective zones for enhancing the dynamic model to improve the behavior of the flow unit productivity, as well as, section of the best sites for the future drilling.

Glucagon-like peptide-1 receptor (GLP-1R) agonists are effective in treating type 2 diabetes and obesity with proven cardiovascular benefits. However, most of them are peptides and require subcutaneous injection except for orally available semaglutide. Boc5 was identified as the first orthosteric non-peptidic agonist of GLP-1R that mimics a broad spectrum of bioactivities of GLP-1 in vitro and in vivo. Here, we report the cryo-electron microscopy structures of Boc5 and its analog WB4-24 in complex with the human GLP-1R and Gs protein. Bound to the extracellular domain, extracellular loop 2, and transmembrane (TM) helices 1, 2, 3 and 7, one arm of both compounds inserted deeply into the bottom of the orthosteric binding pocket that is usually accessible by peptidic agonists, thereby partially overlapping with the residues A8-D15 in GLP-1. The other three arms, meanwhile, extended to the TM1-TM7, TM1-TM2, and TM2-TM3 clefts showing an interaction feature substantially similar to a previously known small molecule agonist LY3502970. Such a unique binding mode creates a distinct conformation that confers both peptidomimetic agonism and biased signaling induced by non-peptidic modulators at GLP-1R. Further, the conformational difference between Boc5 and WB4-24, two closed related compounds, provides a structural framework for fine tuning of pharmacological efficacy in the development of future small molecule therapeutics targeting GLP-1R.

AbstractThis chapter examines the development of a different form of corporate religious governance in the Atlantic in the years after the Jamestown massacre. It focuses on the denominational identity of its members and how this influenced the direction and formation of a theocratic model of governance that the company would adopt. This chapter illustrates how the leaders of the Plymouth and Massachusetts Bay companies, such as William Bradford, John Endicott and John Winthrop, established authoritarian governments by manipulating charter privileges, forming a theocratic model of governance in New England. It examines how the leaders and members of the Plymouth Company and Massachusetts Bay Company, as corporate bodies, established and nurtured a distinct form of governmental identity. By tracing the development of the Massachusetts Bay Company’s congregational theocratic governance through works such as Bradford’s Of Plimoth Plantation, the Winthrop Papers, as well as the Records of the Town of Plymouth and the Records of the Governor and Company of the Massachusetts Bay New England, it shows how the joint stock corporation offered its members the legal and structural framework that would dogmatically police the religious behaviour of its members to secure and establish a godly republic.

The SARS-CoV-2 Omicron variant of concern evades antibody mediated immunity with an unprecedented magnitude due to accumulation of numerous spike mutations. To understand the Omicron antigenic shift, we determined cryo-electron microscopy and X-ray crystal structures of the spike and RBD bound to the broadly neutralizing sarbecovirus monoclonal antibody (mAb) S309 (the parent mAb of sotrovimab) and to the human ACE2 receptor. We provide a structural framework for understanding the marked reduction of binding of all other therapeutic mAbs leading to dampened neutralizing activity. We reveal electrostatic remodeling of the interactions within the spike and those formed between the Omicron RBD and human ACE2, likely explaining enhanced affinity for the host receptor relative to the prototypic virus.

In order to promote the efficient use of reclaimed water in China and make water resources allocation better structured, this paper analyzed the factors that drive and restrain the current utilization of reclaimed water and unveiled their correlation and hierarchy in a way to develop a non-recursive structural framework of what drives and restrain reclaimed water use. By structural equation modeling (SEM), the transmission path of affecting factors was identified, the contribution of the factors quantified, and key indicators for potential prediction selected. On that basis, a random-effects model (REM) was built to predict the potential availability of the country’s reclaimed water. Meanwhile, parametric confidence intervals at 10–90% quantile levels were described, given the uncertainty of REM parameters. The results showed that four indicators for potential prediction, namely the total amount of wastewater treated, the density of water pipelines in built-up areas, investment in facilities for reclaimed water treatment, and the processing of applications for water treatment patents, are intertwined with the utilization of reclaimed water. Overall, the REM for potential prediction produced more precise fitting results, with the most significant fitting error standing at 5.9%. Going ahead, China is set to maintain the rapid growth in reclaimed water use, and up to 13.7 billion cubic meters of reclaimed water is expected to be available by 2025. This will help better structure the urban water supply and render regional water recycling more efficient.

The prime goals of this article are to classify farmers’ production structures in different regions through comparative analysis and propose a structural framework that can quantify the transformation and upgrade of the vegetable industry in Shaanxi. The study utilized New Structural Economics (NSE) theory to construct the theoretical model, and the empirical data set has been crafted from 1992 vegetable growers in the Shaanxi, Hebei, and Shandong provinces. We qualitatively evaluated the potentiality of the Shaanxi vegetable industry, facilitating an industrial upgrade by drawing on the development experience of other regions. More specifically, we utilized the tomato growers to construct a five-step analysis method to transform and upgrade the vegetable industries. This article found that the chosen three provinces have structural differences and similarities in their production sides, and, compared with Hebei and Shandong, the vegetable industry in Shaanxi is still in the catching-up stage. Hebei has similar planting characteristics to Shaanxi, but the production characteristics of growers in Shaanxi are quite different from those in Shandong. Therefore, tomato growers in Hebei are the desired learning object for growers in Shaanxi. By learning planting experience from Hebei growers, tomato growers in the Shaanxi Province can improve their yield and product quality. We found that Shaanxi farmers have 15.57 percent improvement potential in improving crop management and rational allocation of production materials. Proper information-sharing platforms should be implemented to grasp consumption trends on the demand side and guide industrial upgrading.

PARP13/ZAP acts against multiple viruses through recognizing and promoting degradation of cytoplasmic viral mRNA. PARP13 has four N-terminal Zn-finger motifs that bind CG-rich nucleotide sequences, and a C-terminal ADP ribosyltransferase fold similar to other PARPs. A central region predicted to contain a fifth Zn-finger and two tandem WWE domains is implicated in binding poly(ADP-ribose); however, there are limited insights into the structure and function of this PARP13 region (ZnF5-WWE1-WWE2). Here, we present crystal structures of ZnF5-WWE1-WWE2 from mouse PARP13 in complex with ADP-ribose and with ATP. ZnF5-WWE1-WWE2 crystallized as a dimer with major contacts formed between WWE1 and WWE2 originating from different monomers, indicative of a more compact monomeric arrangement of the tandem WWE domains. Solution scattering experiments and biophysical analysis indicated a monomer in solution, suggesting that the crystal dimer represents domain swapping that could potentially represent a PARP13 conformation assumed when signaling viral RNA detection. The crystal structure and binding studies demonstrate that WWE2 interacts with ADP-ribose and ATP, whereas WWE1 does not have a functional binding site. The shape of the WWE2 binding pocket disfavors interaction with the ribose-ribose linkage of poly(ADP-ribose). Binding studies with poly(ADP-ribose) ligands indicate that WWE2 serves as an anchor for preferential binding to the terminal end of poly(ADP-ribose), and the composite structure of ZnF5-WWE1-WWE2 forms an extended surface to engage polymer chains of ADP-ribose. This model represents a novel mode of poly(ADP-ribose) recognition and provides a structural framework for investigating poly(ADP-ribose) impact on PARP13 function.
