Objective. To evaluate the immune complex expression detected by immunofluorescence in order to establish the diagnosis and predict the course of glomerulopathies with extracapillary proliferation. Material and methods. A total of 165 kidney biopsies with extracapillary proliferation were analyzed by light and luminescent microscopy. Results. The immunofluorescence expression and its prognostic significance in glomerulopathies were described in the study. IgG deposits in IgA nephropathy increased the risk of impaired renal function by 2.68 times (95% CI 1.13-6.33). In pauci-immune glomerulonephritis, IgGs were associated with a higher segmental glomerulosclerosis percentage (p=0.020), the absence of IgM increased the risk of progression by 3.15 times (95% CI 1.43-6.94). IgM was more often detected in IgA nephropathy than in IgA vasculitis (p=0.049). Conclusion. Kidney biopsy assessment using immunofluorescence is important for the diagnosis and prognosis of glomerulopathies with extracapillary proliferation.

The Aсanthamoeba keratitis is a rare, but very dangerous disease of cornea; its development is connected with contact lens wearing, and corneal microtraumas with contamination of particles of the soil or water. The disease has no pronounced distinguishing clinical signs therefore it is difficult to distinguish them from herpetic or fungal keratitis. Nevertheless, it can be suspected on the basis of anamnestic data (use of contact lenses, corneal injury with contamination by the soil or water), uneven strong pain syndrome, ring-shaped infiltration of cornea, absence of effect of traditional therapy. Isolation of Acanthamoeba by bioculture method, morphological research of corneal bioptat (by light and luminescent microscopy), gene diagnostics, in – vivo confocal microscopy of the cornea can give additional help in the diagnosis. Also perspective diagnostic methods of Acanthamoeba keratitis are given. Nevertheless, so far there is no conventional technique of diagnosis of this disease.

The purpose of the study was to obtain luminescent serum based on highly purified anthracoid globulins to diagnose anthrax agents in animals. To date, there are plenty diagnostic agents that allow rapid and accurate diagnostics of infectious diseases of animals. One of them is the luminescent microscopy of the fluorescent antibody method, which is used as an express method and provides for diagnostics within 3-5 hours. Hyperimmune serum globulins prepared on two types of antigens – protective from strain 55 (VNIIVViM) and capsular from Lange-2 strain at five-fold scheme of introduction of these antigens – were used to make luminescent anthracoid serum. The luminescent serum made on the basis of highly purified anthracoid globulins has a coloring titer (specific activity) of 1:16. When examining the specificity of the obtained luminescent serum in smears from anthrax agent strains, clear fluorescence was observed with more intense luminescence along the periphery of microbial cells.

Aim. To determine the relationship between the level of inhibin A and biochemical factors of inflammation with the development of infertility in women suffering from chronic trichomoniasis. Methods. The object of the study was the blood serum of 167 women suffering from chronic trichomoniasis. The level of inhibin A was determined by enzyme immunoassay. As a control, the blood serum of 22 practically healthy fertile women was used who had no history of trichomoniasis. The lymphocyte phenotype was determined using luminescent microscopy with monoclonal antibodies staining. The level of immunoglobulins in serum was determined by the standard Mancini method of radial immunodiffusion. The synthesis of cytokines was determined by the method of studying the spontaneous synthesis of interleukins-1, -8, -10 and interferon . The level of cytokines and lactoferrin was evaluated by enzyme immunoassay. The concentrations of 2-macroglobulin and C-reactive protein were determined by the nephelometric method. The fibrinogen concentration was determined by the coagulometric method. Results. In the group of patients with chronic trichomoniasis, a significant variability in the serum concentration of inhibin A from 4.3 to 29.4 ng/L was found. In a group of patients who had a reduced content of inhibin A, in 63.2% of cases, the development of secondary infertility was subsequently diagnosed; in the group of patients who had a normal level of inhibin A, secondary infertility developed only in 12.9% of cases. Conclusion. It was found that among all acute phase proteins, only levels of C-reactive protein and lactoferrin have prognostic value and statistically significantly differ in groups of women with subsequently developed infertility and uncomplicated infertility trichomoniasis.

The paper presents experimental study results of the cytopathic effect of peste de petits ruminants virus on a goat gonad continuous cell line (YaDK-04). The interaction of peste de petits ruminants virus with cells at different stages of its reproduction was shown using a combination of phase-contrast and luminescent microscopy. It was found that at the initial stage of interaction (20–24 hours) the cells became rounded and de-adhered, and the monolayer was partially loosened. On day 2 post reproduction the most part of the culture monolayer affected by the virus began to destruct, and the cell nuclei were displaced to periphery. At the terminal stage (72 hours) the destruction of monolayer cells and cytoplasmic matrix, deformation and partial lysis of the nuclei and cytoplasm, aggregation of detritus occurred. At the final stage of reproduction (96 hours) the peste de petits ruminants virus diffused into the culture medium, the fluorescence in the yellow spectrum decreased significantly, but the virus titer reached 6.89 lg TCD50/cm3.

AbstractThe formation of thin layers of luminescent defects on the faces of planar lithium fluoride crystals located in the positive column and Faraday dark space of a glow gas discharge was studied by time-resolved confocal scanning luminescent microscopy and time-correlated single photon counting. The formation of aggregated color centers in the surface layers of crystals was established using the spectral and kinetic characteristics of luminescence appearing after irradiation. The role of gas discharge electrons, ions, and photons in the defect formation mechanism was considered. The defects were shown to be formed under the influence of vacuum ultraviolet (VUV) photons. The VUV radiation intensity distribution in the discharge gap was measured by the method of thermostimulated luminescence. The main source of this radiation was the anodic and cathodic voltage drop regions in a glow discharge.
