Zusammenfassung. In der ärztlichen Praxis die banalen Unverträglichkeiten nach Konsum von Speisepilzen von einer sich anbahnenden Vergiftung mit potenziell tödlichem Ausgang zu unterscheiden ist nicht trivial. Trotzdem gibt es einige Kriterien, an denen man sich orientieren kann: Für eine Vergiftung mit Amatoxinen, den Giftstoffen z.B. im Grünen Knollenblätterpilz (Amanita phalloides) spricht eine Latenz von sechs Stunden oder mehr zwischen dem Konsum von nicht durch Fachpersonen kontrollierten Lamellenpilzen und dem Beginn meist heftiger Brechdurchfälle. Auch wenn die Therapieoptionen kontrovers diskutiert werden, hat sich die zeitnahe antidotale Behandlung mit Silibinin als zielführend erwiesen.

There are limited data on amperometric biosensors (ABSs) for L-arginine (Arg) determination based on oxidases that produce hydrogen peroxide (H2O2) as a byproduct of enzymatic reaction, and artificial peroxidases (POs) for decomposition of H2O2. The most frequently proposed Arg-sensitive oxidase-based ABSs contain at least two enzymes in the bioselective layer; this complicates the procedure and increases the cost of analysis. Therefore, the construction of a one-enzyme ABS for Arg analysis is a practical problem. In the current work, fabrication, and characterization of three ABS types for the direct measurement of Arg were proposed. L-arginine oxidase (ArgO) isolated from the mushroom Amanita phalloides was co-immobilized with PO-like nanozymes (NZs) on the surface of graphite electrodes. As PO mimetics, chemically synthesized NZs of CeCu (nCeCU) and NiPtPd (nNiPtPd), as well as green-synthesized hexacyanoferrate of copper (gCuHCF), were used. The novel ABSs exhibited high sensitivity and selectivity to Arg, broad linear ranges and good storage stabilities. Two ABSs were tested on real samples of products containing Arg, including the pharmaceutical preparation “Tivortine”, juices, and wine. A high correlation (R = 0.995) was demonstrated between the results of testing “Tivortine” and juice using nCeCU/GE and nNiPtPd/GE. It is worth mentioning that only a slight difference (less than 1%) was observed for “Tivortin” between the experimentally determined content of Arg and its value declared by the producer. The proposed ArgO-NZ-based ABSs may be promising for Arg analysis in different branches of science, medicine, and industry.

Розроблено скринінгові кольорові експрес-тести виявлення L-аргініноксидази (ArgО), запропоновано схему очищення ArgО з шапинкового гриба Amanita phalloides. Препарати АргО виділено в аналітичних кількостях, вивчено деякі властивості ферменту, а саме, умови зберігання та субстратну специфічність.

The consumption of mushrooms has become increasingly popular, partly due to their nutritional and medicinal properties. This has increased the risk of confusion during picking, and thus of intoxication. In France, about 1300 cases of intoxication are observed each year, with deaths being mostly attributed to Amanita phalloides poisoning. Among amatoxins, α- and β-amanitins are the most widely studied toxins. Hepatotoxicity is the hallmark of these compounds, leading to hepatocellular failure within three days of ingestion. The toxic mechanisms of action mainly include RNA polymerase II inhibition and oxidative stress generation, leading to hepatic cell apoptosis or necrosis depending on the doses ingested. Currently, there is no international consensus concerning Amanita phalloides poisoning management. However, antidotes with antioxidant properties remain the most effective therapeutics to date suggesting the predominant role of oxidative stress in the pathophysiology. The partially elucidated mechanisms of action may reveal a suitable target for the development of an antidote. The aim of this review is to present an overview of the knowledge on amanitins, including the latest advances that could allow the proposal of new innovative and effective therapeutics.

Despite the progress of diagnostic and therapy, the cancer burden is still rising worldwide. The new chemotherapeutical toxicity to somatic cells and its tolerance to tumor cells illustrates the immediate demand through recent pharmaceutical products with less harmful impacts. The use of natural anticancer products, like alpha-amanitin toxins have reached the cancer field therapy since the separation of Amanita phalloides fungi was performed. Application of Amanita phalloides affects tumor cell activity. It is thought that Amanita phalloides dilutions are recommended for a patient suffering from various cancer types and have no severe side effects resulting from amanita therapy. This review aims to explain the use of the therapeutic potential of -amanitin toxin against different cancer types.

Alpha-amanitin (α-AMA) is a cyclic peptide and one of the most lethal mushroom amatoxins found in Amanita phalloides. α-AMA is known to cause hepatotoxicity through RNA polymerase II inhibition, which acts in RNA and DNA translocation. To investigate the toxic signature of α-AMA beyond known mechanisms, we used quantitative nanoflow liquid chromatography–tandem mass spectrometry analysis coupled with tandem mass tag labeling to examine proteome dynamics in Huh-7 human hepatoma cells treated with toxic concentrations of α-AMA. Among the 1828 proteins identified, we quantified 1563 proteins, which revealed that four subunits in the T-complex protein 1-ring complex protein decreased depending on the α-AMA concentration. We conducted bioinformatics analyses of the quantified proteins to characterize the toxic signature of α-AMA in hepatoma cells. This is the first report of global changes in proteome abundance with variations in α-AMA concentration, and our findings suggest a novel molecular regulation mechanism for hepatotoxicity.
