<div><div><div><div><p>A very concise total synthesis of (+)-brefeldin C starting from 2-furanylcyclopentene is described. This approach is based on an unprecedented enantioselective radical hydroalkynylation process to introduce the two cyclopentane stereocenters in a single step. The use of a furan substituent allows to achieve a high trans diastereoselectivity during the radical process and it contains the four carbon atoms C1–C4 of the natural product in an oxidation state closely related to the one of the target molecule. The eight-step synthesis require six product purifications and it provides (+)-brefeldin C in 18% overall yield.</p></div></div></div></div>

The resistance to N-methyl-N'-nitro-N-nitrosoguanidine (NG) of five single-step mutants of a lysogenic group A Streptococcus after several cycles of growth in NG-free media was within the range of 1.5 to 3.7 times that of the parent. After ultraviolet irradiation (UV) these mutants had a higher chance to survive than the wild type, giving dose reduction factors of 1.4 to 2.4.In prophage induction experiments using UV and NG as inducing agents, the dose induction functions of the NG-resistant strains were either shifted toward higher doses or the efficiency of induction was decreased. In the strains examined caffeine and acriflavine (which are already known to block host cell reactivation) greatly enhanced both the killing and the inducing effect of UV, indicating the ability of Streptococcus pyogenes to perform dark repair of UV damaged DNA. The extra resistance to UV conferred on the strains carrying the NG-resistance mutation was not decreased by UV enhancers, moreover, their action on dark reactivation seemed to be less efficient in these strains.

OBJECTIVE: The aim of this study was to assess - using scanning electron microscopy (SEM) - the effectiveness of two abrasive discs, one made from silicon and one from aluminum oxide, in removing adhesive remnants (AR) after debonding orthodontic brackets. METHODS: Ten randomly selected bovine teeth were used, i.e., 2 in the control group, and the other 8 divided into two groups, which had orthodontic brackets bonded to their surface with Concise Orthodontic Adhesive (3M). The following methods were employed - in one single step - to remove AR after debracketing: Group A, Optimize discs (TDV) and Group B, Onegloss discs (Shofu), used at low speed. After removing the AR with the aforementioned methods, the teeth were prepared to undergo SEM analysis, and photographs were taken of the enamel surface with 50x magnification. Six examiners evaluated the photographs applying the Zachrisson and Årtun enamel surface index (ESI) system (1979). RESULTS: Group A exhibited minor scratches on the enamel surface as well as some AR in some of the photographs, while Group B showed a smoother surface, little or no AR and some abrasion marks in the photographs. No statistically significant differences were found between the two methods and the control group. CONCLUSIONS: The two abrasive discs were effective in removing the AR after bracket debonding in one single step.

ABSTRACT Degradation of lignin-related aromatic compounds is an important ecological process in the highly productive salt marshes of the southeastern United States, yet little is known about the mediating organisms or their catabolic pathways. Here we report the diversity of a gene encoding a key ring-cleaving enzyme of the β-ketoadipate pathway, pcaH, amplified from bacterial communities associated with decaying Spartina alterniflora, the salt marsh grass that dominates these coastal systems, as well as from enrichment cultures with aromatic substrates (p-hydroxybenzoate, anthranilate, vanillate, and dehydroabietate). Sequence analysis of 149 pcaH clones revealed 85 unique sequences. Thirteen of the 53 amino acid residues compared were invariant in the PcaH proteins, suggesting that these residues have a required catalytic or structural function. Fifty-eight percent of the clones matched sequences amplified from a collection of 36 bacterial isolates obtained from seawater, marine sediments, or senescent Spartina. Fifty-two percent of thepcaH clones could be assigned to the roseobacter group, a marine lineage of the class α-Proteobacteria abundant in coastal ecosystems. Another 6% of the clones matched genes retrieved from isolates belonging to the genera Acinetobacter, Bacillus, and Stappia, and 42% of the clones could not be assigned to a cultured bacterium based on sequence identity. These results suggest that the diversity of the genes encoding a single step in aromatic compound degradation in the coastal marsh examined is high.

Background: Traumatic avulsion injury poses severe risk as the overlying protective covering is lost and the raw tissue is exposed to the environment. Avulsion injuries involving the scalp are even more complicated to treat because of significant cosmetic concern involved. Aim of the study was to find a better solution than the existing method, we conducted a prospective study involving 13 patients with isolated traumatic scalp avulsion injury.Methods: This prospective study was conducted in Motilal Nehru Medical College and associated Swaroop Rani Nehru Hospital, Prayagraj,  after taking written and informed consents from the patients, between June 2017 and June 2019.These were divided into two groups (A and B) based on whether the underlying periosteum was intact or not.Results: Patients with intact periosteum (Group A) underwent primary thin thickness skin grafting within a few hours of their admission while the other group (Group B) was treated with a traditional conservative approach. We compared the results of both the groups and found that Group A patients not only had satisfactory graft uptake (≥85 TBSA) but also had significant decreased risk of infection, lesser hospital stay, overall decreased healthcare cost, better cosmoses and early return to routine activity.Conclusions: For the surgeon, this single step procedure is safe and technically easy. Thus this approach was found to be superior than the current traditional approach.

Streptococcal cell membrane (SCM) antiserum injected mice show a significant thickening of glomerular basement membrane (GBM) and an increase in mesangial matrix within 4 to 24 hours of antiserum administration (1,2,3). This study was undertaken to evaluate the incorporation of 3H proline into glomerular cells and GBM under normal and anti-SCM induced conditions. Mice were administered, intraperitoneally, 0.1 ml of normal or anti-SCM serum followed by a 10 µC/g body weight injection of 3H proline. Details of the preparation of anti-SCM (Group A type 12 streptococcal pyogenes) and other sera and injection protocol have been described elsewhere (2). After 15 minutes of isotope injection a chase of cold proline was given and animal sacrificed at 20 minutes, 1,2,4,8,24 and 48 hours. One of the removed kidneys was processed for immunofluorescence, light and electron microscopic radioautographic studies; second kidney was used for GBM isolation and aminoacid analysis.

We have recently carried out a study of spermiogenic differentiation in two geographically isolated populations of Aphanius dispar (freshwater teleost), with a view to ascertaining variation at the ultrastructural level. The sampling areas were the Jebel Al Akhdar in the north (Group A) and the Dhofar region (Group B) in the south. Specimens from each group were collected, the testes removed, fixed in Karnovsky solution, post fixed in OsO, en bloc stained with uranyl acetate and then routinely processed to Agar 100 resin, semi and ultrathin sections were prepared for study.
