Abstract Duplex sequencing is currently the most reliable method to identify ultra-low frequency DNA variants by grouping sequence reads derived from the same DNA molecule into families with information on the forward and reverse strand. However, only a small proportion of reads are assembled into duplex consensus sequences (DCS), and reads with potentially valuable information are discarded at different steps of the bioinformatics pipeline, especially reads without a family. We developed a bioinformatics toolset that analyses the tag and family composition with the purpose to understand data loss and implement modifications to maximize the data output for the variant calling. Specifically, our tools show that tags contain polymerase chain reaction and sequencing errors that contribute to data loss and lower DCS yields. Our tools also identified chimeras, which likely reflect barcode collisions. Finally, we also developed a tool that re-examines variant calls from raw reads and provides different summary data that categorizes the confidence level of a variant call by a tier-based system. With this tool, we can include reads without a family and check the reliability of the call, that increases substantially the sequencing depth for variant calling, a particular important advantage for low-input samples or low-coverage regions.

AbstractWith ever-increasing computational power, larger computational domains are employed and thus the data output grows as well. Writing this data to disk can become a significant part of runtime if done serially. Even if the output is done in parallel, e.g., via MPI I/O, there are many user-space parameters for tuning the performance. This paper focuses on the available parameters for the Lustre file system and the Cray MPICH implementation of MPI I/O. Experiments on the Cray XC40 Hazel Hen using a Cray Sonexion 2000 Lustre file system were conducted. In the experiments, the core count, the block size and the striping configuration were varied. Based on these parameters, heuristics for striping configuration in terms of core count and block size were determined, yielding up to a 32-fold improvement in write rate compared to the default. This corresponds to 85 GB/s of the peak bandwidth of 202.5 GB/s. The heuristics are shown to be applicable to a small test program as well as a complex application.

The construction of a goniometric glove is described. Each of the sensors in the glove was calibrated over a custom built metal hand using blocks of known angles as angular references. The digital data output from each sensor of the glove were converted into angular displacements at each joint. The glove was validated for consistency of measurement and accuracy over a custom built metal jig and in the human hand. The accuracy of the glove was found to be within the limits of traditional goniometry. It is proposed that goniometric gloves could be useful in the assessment of hand function.

Balance in children is one of the most important things to notice during its development. Increased risk of loss ofbalance is often associated with a slow reaction time. The purpose of this study was to determine the relationshipbetween auditory reaction time and body balance in students at Elementary School in Baha village. This research wasconducted in March 2018 with cross sectional analytic study design. Samples were taken by simple random samplingmethod with 96 samples. The sample age range is 8 - 10 years. The independent variable is auditory reaction timemeasured using computerized reaction time test software. Dependent variable is body balance measured usingPediatric Balance Scale. The relationship between two variables was analyze using Pearson Correlation test. There isa significant correlation between auditory reaction time to body balance, based on data output from analysis withsignificance <? (p = 0,006). Furthermore, based on data output known Correlation Coefficient of -0.228 which meansthe existence of negative and linear relationship. There was a significant relationship between auditory reaction timeand body balance in elementary school children in Baha Village.Keywords: Reaction Time, Auditory, Balance, Children

Sistem data logger telah banyak dikembangkan untuk berbagai keperluan perolehan data untuk melakukan proses monitoring dan akuisisi data. Penelitian ini adalah aplikasi data logger untuk pemrosesan lima data dari sensor-sensor navigasi pada suatu kapal maritim. Peralatan sensor navigasi kapal maritim di Indonesia sudah mendukung protokol komunikasi secara digital yang mengacu pada format standar komunikasi sensor kemaritiman National Marine Electronics Association (NMEA). Terdapat lima sensor navigasi yang digunakan sebagai standar minimal yang mendukung kapal maritim untuk dapat beroperasi di lautan, yaitu; sensor GPS, Gyrocompass, Echosounder, Weather dan Velocity. Penelitian ini mencoba untuk menampung lima buah data sensor yang berbeda, kemudian menggabungkannya dalam suatu variabel dengan susunan format data yang baru sebagai data output. Data output kemudian dikirimkan pada PC untuk diterjemahkan melalui software, dan data setiap sensor ditampilkan pada software aplikasi tersebut. Sistem ini dibangun dengan Arduino ATMega328 sebagai prosesor. Pengkondisi data input dibuat dengan kombinasi rangkaian IC 74LS08 dan IC 74LS32. Hasil penelitian ini adalah prototipe data logger yang menangani sejumlah lima data sensor dengan format standar NMEA dan satu output data sensor-sensor yang telah disusun dengan format tertentu, dengan tingkat akurasi saat pengiriman data sebesar 100% dan ekstraksi data pada software aplikasi display sebesar; 96% untuk sensor GPS, 97% untuk Gyro, 98% untuk Echosounder, 97% untuk Weather, dan 98% untuk sensor Velocity.
