MAC5 is a component of the conserved MOS4-associated complex. It plays critical roles in development and immunity. Here we report that MAC5 is required for microRNA (miRNA) biogenesis. MAC5 interacts with Serrate (SE), which is a core component of the microprocessor that processes primary miRNA transcripts (pri-miRNAs) into miRNAs and binds the stem-loop region of pri-miRNAs. MAC5 is essential for both the efficient processing and the stability of pri-miRNAs. Interestingly, the reduction of pri-miRNA levels inmac5is partially caused by XRN2/XRN3, the nuclear-localized 5′-to-3′ exoribonucleases, and depends on SE. These results reveal that MAC5 plays a dual role in promoting pri-miRNA processing and stability through its interaction with SE and/or pri-miRNAs. This study also uncovers that pri-miRNAs need to be protected from nuclear RNA decay machinery, which is connected to the microprocessor.

AbstractNegative symptoms have long been considered a core component of schizophrenia. Modern conceptualizations of the structure of negative symptoms posit that there are at least two broad dimensions (motivation and pleasure and diminished expression) or perhaps five separable domains (avolition, anhedonia, asociality, blunted affect, alogia). The current review synthesizes a body of emerging research indicating that avolition may have a special place among these dimensions, as it is generally associated with poorer outcomes and may have distinct neurobiological mechanisms. Network analytic findings also indicate that avolition is highly central and interconnected with the other negative symptom domains in schizophrenia, and successfully remediating avolition results in global improvement in the entire constellation of negative symptoms. Avolition may therefore reflect the most critical treatment target within the negative symptom construct. Implications for targeted treatment development and clinical trial design are discussed.

Using TSG101 pre-mRNA, we previously discovered cancer-specific re-splicing of mature mRNA that generates aberrant transcripts/proteins. The fact that mRNA is aberrantly re-spliced in various cancer cells implies there must be an important mechanism to prevent deleterious re-splicing on the spliced mRNA in normal cells. We thus postulated that mRNA re-splicing is controlled by specific repressors, and we searched for repressor candidates by siRNA-based screening for mRNA re-splicing activity. We found that knock-down of EIF4A3, which is a core component of the exon junction complex (EJC), significantly promoted mRNA re-splicing. Remarkably, we could recapitulate cancer-specific mRNA re-splicing in normal cells by knock-down of any of the core EJC proteins, EIF4A3, MAGOH, or RBM8A (Y14), implicating the EJC core as the repressor of mRNA re-splicing often observed in cancer cells. We propose that the EJC core is a critical mRNA quality control factor to prevent over-splicing of mature mRNA.

Purpose – For over a 100 years, Macau’s Pataca has been tied to Macau’s identity, its independent financial existence and its links to the Lusophone world. Its role as a supporter of the Macau identity relies upon the strength and capabilities of its financial institutions, the Macau Monetary Authority and banking institutions that issue its currency (Banco Nacional Ultramarino and Bank of China). The paper aims to discuss the above issue. Design/methodology/approach – The author uses statistics and data from the Macau Monetary Authority. Findings – There are three possible scenarios that could emerge: first, retired in favor of the RMB, the HK dollar; second, maintained with its use expanding as Macau’s revenues and investment funds grow, or third, become a force for stronger economic cohesion and identity among the Lusophone nations. Practical implications – Which of these scenarios will offer Macau the highest possibility of maintaining its international network of relationships and representation, diversifying its economy into new growth markets and playing a unique role in China’s twenty-first century destiny? Originality/value – This paper studies a rarely discussed topic and focuses on a core component of Macau’s existence.

ABSTRACT Nine metal-polluted white dwarfs are observed with medium-resolution optical spectroscopy, where photospheric abundances are determined and interpreted by comparison with Solar system objects. An improved method for making such comparisons is presented, which overcomes potential weaknesses of prior analyses, with numerous sources of error considered to highlight the limitations on interpretation. The stars are inferred to be accreting rocky, volatile-poor asteroidal materials with origins in differentiated bodies, in line with the consensus model. The most heavily polluted star in the sample has 14 metals detected, and appears to be accreting material from a rocky planetesimal, whose composition is mantle-like with a small Fe–Ni core component. Some unusual abundances are present. One star is strongly depleted in Ca, while two others show Na abundances elevated above bulk-Earth abundances; it is speculated that either the latter reflect diversity in the formation conditions of the source material, or they are traces of past accretion events. Another star shows clear signs that accretion ceased around 5 Myr ago, causing Mg to dominate the photospheric abundances, as it has the longest diffusion time of the observed elements. Observing such post-accretion systems allows constraints to be placed on models of the accretion process.

AbstractThe RNA-binding protein Lin28 (Lin28a) is an important pluripotency factor that reprograms translation and promotes cancer progression. Although Lin28 blocks let-7 microRNA maturation, Lin28 also binds to a large set of cytoplasmic mRNAs directly. However, how Lin28 regulates the processing of many mRNAs to reprogram global translation remains unknown. We show here, using a structural and cellular approach, a mixing of Lin28 with YB-1 (YBX1) in the presence of mRNA owing to their cold-shock domain, a conserved β-barrel structure that binds to ssRNA cooperatively. In contrast, the other RNA binding-proteins without cold-shock domains tested, HuR, G3BP-1, FUS and LARP-6, did not mix with YB-1. Given that YB-1 is the core component of dormant mRNPs, a model in which Lin28 gains access to mRNPs through its co-association with YB-1 to mRNA may provide a means for Lin28 to reprogram translation. We anticipate that the translational plasticity provided by mRNPs may contribute to Lin28 functions in development and adaptation of cancer cells to an adverse environment.

Dairy science is a multidisciplinary area of scientific investigation and Ph.D. students aiming to do research in the field of animal and/or veterinary sciences must be aware of this. Ph.D. students often have vast spectra of research interests, and it is quite challenging to satisfy the expectation of all of them. The aim of this study was to establish an international Ph.D. training program based on research collaboration between the University of Sydney and the University of Padova. The core component of this program was a two-week Postgraduate Summer School in Dairy Science, which was held at the University of Padova, for Ph.D. students of both universities. Therefore, we designed a program that encompassed seminars, workshops, laboratory practical sessions, and farm visits. Participants were surveyed using a written questionnaire. Overall, participants have uniformly praised the Summer School calling it a rewarding and valuable learning experience. The Ph.D. Summer School in Dairy Science provided its participants a positive learning experience, provided them the opportunity to establish an international network, and facilitated the development of transferable skills.

Radial-inflow turbine is a core component in supercritical CO2 (SCO2) Brayton cycle. The leakage from the nozzle outlet towards the impeller back brings a great challenge to the efficiency and security of the power system. In this paper, the labyrinth seal (LS) and dry gas seal (DGS) are arranged on the impeller back of a SCO2 radial-inflow turbine and the influence on the comprehensive performance is investigated. Results demonstrate that both LS and DGS configurations can significantly reduce leakage of the impeller back and DGS configuration performs better. Compared with the configuration without leakage, the power and efficiency of DGS configuration are only reduced by 0.27% and 0.35% respectively. The seal clearance and the inlet width have a greater effect on LS configuration. The thermo-mechanical seal deformation values of DGS configurations are all less than 8 μm, which verifies the feasibility. Finally, a novel combined seal configuration with both LS and DGS is proposed and excellent performance is achieved, providing a potential approach for the sealing problem of SCO2 radial-inflow turbine.

This article describes how the enormous potential benefits provided by the cloud services, made enterprises to show huge interest in adopting cloud computing. As the service provider has control over the entire data of an organization stored onto the cloud, a malicious activity, whether internal or external can tamper with the data and computation. This causes enterprises to lack trust in adopting services due to privacy, security and trust issues. Despite of having such issues, the consumer has no root level access right to secure and check the integrity of procured resources. To establish a trust between the consumer and the provider, it is desirable to let the consumer to check the procured platform hosted at provider side for safety and security. This article proposes an architectural design of a trusted platform for the IaaS cloud computing by the means of which the consumer can check the integrity of a guest platform. TCG's TPM is deployed and used on the consumer side as the core component of the proposed architecture and it is distributed between the service provider and the consumer.
