Mechanoenzymes are enzymes which use a chemical reaction to power directed movement along biological polymer. Such enzymes include the cytoskeletal motors (e.g., myosins, dyneins, and kinesins) as well as nucleic acid polymerases and helicases. A single catalytic turnover of a mechanoenzyme moves the enzyme molecule along the polymer a distance on the order of 10−9 m We have developed light microscope and digital image processing methods to detect and measure nanometer-scale motions driven by single mechanoenzyme molecules. These techniques enable one to monitor the occurrence of single reaction steps and to measure the lifetimes of reaction intermediates in individual enzyme molecules. This information can be used to elucidate reaction mechanisms and determine microscopic rate constants. Such an approach circumvents difficulties encountered in the use of traditional transient-state kinetics techniques to examine mechanoenzyme reaction mechanisms.

Abstract The characteristic of specialized computer programs has been presented, serving for identification of W. Budryk-S. Knothe theory parameters, used for description of asymptotic state of post-mining deformations, as well as for transient state. The software is the result of several years of authors’ work. It is a part of complete software system designed for forecasting of underground mining influences on the rock mass and land surface and graphical processing of calculations results. Apart from software description, a short example of its practical utilization has been attached.

In the field of microgrids (MGs), steady-state power imbalances and frequency/voltage fluctuations in the transient state have been gaining prominence owing to the advancing distributed energy resources (DERs) connected to MGs via grid-connected inverters. Because a stable, safe power supply and demand must be maintained, accurate analyses of power system dynamics are crucial. However, the natural frequency components present in the dynamics make analyses complex. The nonlinearity and confidentiality of grid-connected inverters also hinder controllability. The MG considered in this study consisted of a synchronous generator (the main power source) and multiple grid-connected inverters with storage batteries and virtual synchronous generator (VSG) control. Although smart inverter controls such as VSG contribute to system stabilization, they induce system nonlinearity. Therefore, Koopman mode decomposition (KMD) was utilized in this study for consideration as a future method of data-driven analysis of the measured frequencies and voltages, and a frequency response analysis of the power system dynamics was performed. The Koopman operator is a linear operator on an infinite dimensional space, whereas the original dynamics is a nonlinear map on a finite state space. In other words, the proposed method can precisely analyze all the dynamics of the power system, which involve the complex nonlinearities caused by VSGs.

The dynamic model of the front double wishbone suspension and the rear multi-link suspension of the vehicle are established. On the basis of detailed analysis of suspension kinematics, calculation method of wheel alignment angle and force calculation of suspension bushing, the influence mechanism of suspension bushing on the vehicle transient state is clarified, and the vehicle transient characteristic index is derived from the vehicle three-free dynamic model. The sensitivity analysis of the suspension bushing is carried out, and the bushing stiffness which has a great influence on the transient state of the vehicle is obtained. The bushing stiffness scale factor is used as the optimization variable, the vehicle transient characteristic index is used as the optimization target, and the NSGA-II optimization algorithm is used for multi-objective optimization. After optimization, one Pareto solution is selected to compare with the original vehicle, the comparison results show that the yaw rate gain, resonance frequency and delay time of yaw rate in the vehicle transient characteristic index are all improved, other optimization targets change less. In the steady-state comparison, the understeer tendency of the vehicle increases, and the roll angle of the vehicle increases but is within an acceptable range.

AbstractObstructive sleep apnea (OSA) is a common sleep respiratory disease. Previous studies have found that the wakefulness electroencephalogram (EEG) of OSA patients has changed, such as increased EEG power. However, whether the microstates reflecting the transient state of the brain is abnormal is unclear during obstructive hypopnea (OH). We investigated the microstates of sleep EEG in 100 OSA patients. Then correlation analysis was carried out between microstate parameters and EEG markers of sleep disturbance, such as power spectrum, sample entropy and detrended fluctuation analysis (DFA). OSA_OH patients showed that the microstate C increased presence and the microstate D decreased presence compared to OSA_withoutOH patients and controls. The fifth microstate E appeared during N1-OH, but the probability of other microstates transferring to microstate E was small. According to the correlation analysis, OSA_OH patients in N1-OH showed that the microstate D was positively correlated with delta power, and negatively correlated with beta and alpha power; the transition probability of the microstate B → C and E → C was positively correlated with alpha power. In other sleep stages, the microstate parameters were not correlated with power, sample entropy and FDA. We might interpret that the abnormal transition of brain active areas of OSA patients in N1-OH stage leads to abnormal microstates, which might be related to the change of alpha activity in the cortex.
