This work investigated the partitioning of b-galactosidase from Kluyveromyces fragilis in aqueous two-phase systems (ATPS) by bioaffinity. PEG 4000 was chemically activated with thresyl chloride, and the biospecific ligand p-aminophenyl 1-thio-b-D-galactopyranoside (APGP) was attached to the activated PEG 4000. A new two-step method for extraction and purification of the enzyme b-galactosidase from Kluyveromyces fragilis was developed. In the first step, a system composed of 6% PEG 4000-APGP and 8% dextran 505 was used, where b-galactosidase was strongly partitioned to the top phase (K = 2,330). In the second step, a system formed of 13% PEG-APGP and 9% phosphate salt was used to revert the value of the partition coefficient of b-galactosidase (K = 2 x 10-5) in order to provide the purification and recovery of 39% of the enzyme in the bottom salt-rich phase.

A decrease in the number of binding sites for125I-transferrin, without an apparent modification of the association constant, has been observed during the maturation of reticulocytes into erythrocytes. As an experimental model, different red cell populations from phenylhydrazinic anaemic rates (95% to 12% reticulocyte-rich) have been used. The fractionation by multiple partition in two-phase systems of these red cell populations has been applied here to show the relationship between number of transferrin receptors and rate of reticulocyte maturation.

The effect of various sulphate salts on the partitioning of endo-pectinase and exo-pectinase in aqueous two-phase systems, composed of polyethylene glycol and dextran, was studied. Presence of ammonium sulphate and sodium sulphate at concentration 15 mmol/l in the system polyethylene glycol 4000/crude dextran, at tie-line length 7.44%, increased partition coefficient of endo-pectinase 1.25 and 1.2 fold, respectively. Ammonium sulphate at 15 mmol/l and sodium sulphate at 5 mmol/l enhanced partition coefficient for exo-pectinase for about 60% in comparison to the system without salts. Addition of magnesium and sodium sulphate to a final concentration of 0.3 mmol/l in the system containing polyethylene glycol 6000/dextran 500 000, at tie-line length 6.26%, increased the partition coefficient of endo activity for 95% and 32%, respectively. Both salts at the same concentration increased partition coefficient of exo activity 1.5 and 3 times, respectively, in comparison to the system without salts.

Abstract Aqueous two-phase systems (ATPS) have the advantages of environmentally friendly, high mass transfer efficiency, mild extraction conditions. However, it is difficult to recycle these components, which limits the large-scale application of ATPS in industrial production. In this study, a novel recyclable ATPS of PN/PADB4.78 was constructed with a thermo-responsive polymer PN and a pH-responsive polymer PADB4.78 for the partition study of tea saponin. The recovery of PN reached 98.71% by adjusting the temperature to above its lower critical solution temperature (LCST, 33.0 °C ) and the recovery of PADB4.78 was 97.32% by regulating the pH to its isoelectric point (pI, 4.78). Meanwhile, the phase formation mechanism of ATPS was studied by surface tension and low-field nuclear magnetic resonance (LF-NMR). The effects of polymer concentration, pH, temperature, types and concentrations of salt were investigated on tea saponin partition. In the 1.5% PN/3.5% PADB4.78 ATPS, the optimal partition coefficient (K) and extraction recovery (ERb) of crude tea saponin were 0.15 and 92.13% in the presence of 1.5 mM KCl at pH=7.6 and 25℃, respectively. The K and ERb of tea saponin from tea seeds were 0.12 and 94.50% with 7.5 mM LiBr at pH=8.0 and 25℃, respectively.

The objective of this paper was to separate curcumine from Rhizoma curcuma longae and aqueous two-phase extraction technology was employed to perform the study. Four types of PEG (400, 600, 2000 and 4000) and three types of salt ((NH4)2SO4, KH2PO4 and K2HPO4) were tested to form the aqueous two-phase systems. The results showed that the ATPS composed of PEG 400 and (NH4)2SO4 were the best. And the partition coefficient of curcumine could reach 21.79 when the concentration of PEG 400 and (NH4)2SO4 was 14.6% and 26% respectively.
