Effects of essential oils from Cymbopogon citratus (DC) Stapf., Lippia sidoides Cham., and Ocimum gratissimum L. on growth and ultrastructure of Leishmania chagasi promastigotes

Abstract This study verified the effects of essential oils from Lippia sidoides (EOLS) and Cymbopogon citratus (EOCC) on the anesthesia of freshwater angelfish (Pterophyllum scalare) of two different sizes (juveniles I (0.82 g) and II (2.40 g)) and on the transport (8 h) of juveniles II. Fish were exposed to different concentrations of EOLS and EOCC: 0, 10, 25, 50, 75, 100, 150, 200, and 250 mg L−1. Ventilatory rate (VR) and transport for 8 h with 0, 10, and 15 mg L−1 of each essential oil were evaluated in juveniles II. The major components found in EOLS and EOCC were carvacrol (44.50%) and a-citral (73.56%), respectively. The best sedation and anesthesia times for both essential oils were obtained with 10 and 25 mg L−1 and 200 and 250 mg L−1 for juveniles I and II, respectively. Fish sedated with EOLS had lower VR values than the other treatments. For transported fish, blood glucose levels were higher in ornamental fish transported with 10 and 15 mg EOLS L−1 and 15 mg EOCC L−1. Hepatic glycogen values were higher in the control group. In general, fish transported with 10 mg EOLS L−1 showed fewer gill histological alterations than other transported fish. When the type of lesion was evaluated, the highest gill alterations occurred in fish transported with EOCC. In conclusion, for the transport of juveniles II, 10 mg EOLS L−1 could be used because although this concentration increased blood glucose, it decreased the VR and caused only mild alterations to the gills.

Aims: The present investigations describes bactericidal potential of essential oil for management of Xanthomonas albilineans (Ashby. 1929) Dowson 1943, pathogen responsible for sugarcane Leaf scald disease in Côte d'Ivoire. Study Design: the study was conducted at the Ferké 1 sugar mill and the Laboratory of Plant Physiology of Felix Houphouët BOIGNY University. Methodology: Diseased leaves specimens showing signs of X. albilineans presence were collected from sugarcane industrial plantations in Côte d'Ivoire. Pathogenicity was confirmed by observing Koch’s postulates under semi-controlled conditions. Asymptomatic leaves, obtained after 60 days of culture, were inoculated at three points by infiltration with bacterial suspension calibrated at 108 bacteria/ml. Apparition of small tan-brown necrotic lesions on the leaf blade, parallel to the veins and prolonged to the ends by a discolored vascular bundle confirmed that disease is caused by Xanthomonas albilineans. Three essential oils of aromatic plants whose antibacterial properties are recognized were used at 100; 300; 500; 1000 and 2000 ppm in comparison with Callicuivre (56% copper oxychloride) used as reference product. Results: The aqueous solutions based on these essential oils induced an inhibition zone of bacterial growth proportional to the dose applied and the incubation period. Thus, on day 5 of incubation, the essential oil solution of Ocimum gratissimum L. at 100 ppm induced an average inhibition zone diameter not significantly different from the one induced by the Cymbopogon citratus (DC.) Stapf solution at 300 ppm and the one of copper oxychloride at 1000 ppm. At 1000 ppm, Zingiber officinale Roscoe, Cymbopogon citratus (DC.) Stapf and Ocimum gratissimum L essential oils induced equivalent inhibition rates (6 mm) that were higher than that of the control (3.5 mm). Conclusion: sugarcane varieties cultivated at the Ferké 1 sugar mill complex, despite the selection for resistance are threatened by Leaf Scald Disease, which is significantly expressed on variety R585. Cymbopogon citratus (DC.) Stapf and Ocimum gratissimum L oils have the strongest antibacterial activity and may be useful to control Leaf Scald Disease.

Objetivou-se avaliar dois métodos de inoculação de Colletotrichum gloeosporioides em maracujá, testar a patogenicidade de diferentes isolados, o efeito fungitóxico e a composição química dos óleos essenciais das espécies medicinais alecrim-pimenta (Lippia sidoides Cham.), capim-santo [Cymbopogon citratus (D. C.) Stapf.], alfavaca-cravo (Ocimum gratissimum L.), no controle da antracnose [Colletotrichum gloeosporioides (Penz.)], associado ao estádio de maturação de frutos de maracujazeiro-amarelo. Avaliaram-se três experimentos, onde se testou a patogenicidade de seis isolados do fungo em delineamento inteiramente casualizado, com seis repetições, outro com o mesmo delineamento em esquema fatorial 2x2 (suspensão de conídios e disco de micélio) e frutos (verdes e maduros), com seis repetições. No tratamento com frutos, utilizou-se o delineamento inteiramente casualizado, em esquema fatorial 5x3+1, sendo cinco concentrações (0; 2; 4; 6 e 8µL mL-1) e três espécies medicinais, mais o tebuconazol, com cinco repetições. Fez-se a caracterização química dos óleos por cromatografia gasosa, com espectrometria de massas. Todos os isolados foram patogênicos. Os frutos maduros apresentaram maior diâmetro das lesões, quando inoculados com suspensão de conídios. O óleo de C. citratus proporcionou o menor diâmetro das lesões nos frutos, até a concentração de 6 µL mL-1. Na concentração de 8 µL mL-1, todos os óleos inibiram o desenvolvimento do fungo. O timol (30,24%), o citral (77,74%) e o eugenol (92,89%) foram componentes majoritários em L. sidoides, C. citratus e O. gratissimum, respectivamente.
