Toxicity and gastric tolerance of essential oils from Cymbopogon citratus, Ocimum gratissimum and Ocimum basilicum in Wistar rats

Aim: The aim of this study was to evaluate the sanitary quality of smoked fish by determining their mycotoxin content, and also the chemical composition, antifungal activity of Cymbopogon citratus and Ocimum basilicum essential oils from Cameroon against some mycotoxigenic fungal strains responsible of the smoked fish biodegradation. Place and Duration of Study: Laboratory of Food Microbiology and Biotechnology, National School of Agro-Industrial Sciences, University of Ngaoundere, Cameroon, from August 2019 to April 2020. Methodology: Fifteen samples of smoked fishes have been collected in August 2019 from “petit marché” market (Ngaoundere, Cameroon). Physico-chemical parameters of smoked fishes were evaluated. Mycotoxin (AFB1, CIT, and OTA) contents have been determined with HPLC. Isolation and identification of molds were done using their macroscopic and microscopic characteristics, and the identity of the strains was done by PCR sequencing methods. For plants, 15 kg of Cymbopogon citratus leaves and 5 kg of Ocimum basilicum leaves have been harvested on 10 September 2019 in Ngaoundere and used for the essential oil’s extraction. Essential oil extraction has been done through hydrodistillation and the determination of its chemical composition done with GC/MS. Results: Major part of samples contains at least one mycotoxin, in quantities which are beyond the safe dose. Eleven species of molds have been identified: Aspergillus spp., Penicillium citrinum, and Mucor hiemalis. Amongst the isolated species, those which can produce mycotoxins are: Penicillium citrinum, Aspergillus versicolor, Aspergillus fumigatus, Aspergillus carbonarius, Aspergillus niger, and Fusarium moniliforme. Geranial (42.4%), Neral (33.5%) and Myrcene (10.8%) are major compounds found in the essential oil of C. citratus while monoterpens (60.8%), Eugenol (30.7%), Linalol (29.4%) and 1,8-Cineol (14.3%) are the major compounds found in the essential oil of O. basilicum.  Conclusion: Essential oils of C. citratus and O. basilicum are efficient against isolated toxigenic species. C. citratus being more efficient than O. basilicum.

The essential oils and methanol extracts of Ocimum basilicum L., Ocimum kilimandscharicum Guerke, Ocimum gratissimum L, Ocimum canum Sims, and Ocimum tenuiflorum L. (green type) were examined for their potential antibacterial activities. The chemical composition of essential oils of Ocimum species was analysed by GC-MS. The inhibitory effects of essential oils and methanol extracts were studied on two Gram-positive (Bacillus subtilis, Micrococcus luteus) and five Gram-negative (Pseudomonas aeruginosa, Shigella dysenteriae, Escherichia coli, Vibrio cholera, and Shigella flexneri) bacteria using disc-diffusion method. Minimum inhibitory concentration (MIC) was assessed by micro broth dilution method. The antibacterial test results showed that the essential oils of Ocimum basilicum L., Ocimum kilimandscharicum Guerke, and Ocimum gratissimum L. strongly inhibited the growth of all of the microorganisms studied, especially of the Gram-negative strains, whereas other two essential oils showed moderate activities. The result may suggest that the essential oils of Ocimum possess compounds with antibacterial activities, and therefore could be used as natural preservative ingredients in food and/or pharmaceutical industries.

Aims: The present investigations describes bactericidal potential of essential oil for management of Xanthomonas albilineans (Ashby. 1929) Dowson 1943, pathogen responsible for sugarcane Leaf scald disease in Côte d'Ivoire. Study Design: the study was conducted at the Ferké 1 sugar mill and the Laboratory of Plant Physiology of Felix Houphouët BOIGNY University. Methodology: Diseased leaves specimens showing signs of X. albilineans presence were collected from sugarcane industrial plantations in Côte d'Ivoire. Pathogenicity was confirmed by observing Koch’s postulates under semi-controlled conditions. Asymptomatic leaves, obtained after 60 days of culture, were inoculated at three points by infiltration with bacterial suspension calibrated at 108 bacteria/ml. Apparition of small tan-brown necrotic lesions on the leaf blade, parallel to the veins and prolonged to the ends by a discolored vascular bundle confirmed that disease is caused by Xanthomonas albilineans. Three essential oils of aromatic plants whose antibacterial properties are recognized were used at 100; 300; 500; 1000 and 2000 ppm in comparison with Callicuivre (56% copper oxychloride) used as reference product. Results: The aqueous solutions based on these essential oils induced an inhibition zone of bacterial growth proportional to the dose applied and the incubation period. Thus, on day 5 of incubation, the essential oil solution of Ocimum gratissimum L. at 100 ppm induced an average inhibition zone diameter not significantly different from the one induced by the Cymbopogon citratus (DC.) Stapf solution at 300 ppm and the one of copper oxychloride at 1000 ppm. At 1000 ppm, Zingiber officinale Roscoe, Cymbopogon citratus (DC.) Stapf and Ocimum gratissimum L essential oils induced equivalent inhibition rates (6 mm) that were higher than that of the control (3.5 mm). Conclusion: sugarcane varieties cultivated at the Ferké 1 sugar mill complex, despite the selection for resistance are threatened by Leaf Scald Disease, which is significantly expressed on variety R585. Cymbopogon citratus (DC.) Stapf and Ocimum gratissimum L oils have the strongest antibacterial activity and may be useful to control Leaf Scald Disease.
