Population growth of the stored product pest Tyrophagus putrescentiae (Acari: Acaridae) on environmentally and medically important fungi

Legume proteins were shown to have insecticidal activity against stored-product pests. Grain enriched by bean (<I>Phaseolus vulgaris</I>) flour inhibits the growth of stored-product mites. In this study, we tested the toxicity of bean flour to storage mites under optimal conditions for their population growth (i.e. rearing diet, temperature: 25C and humidity optimum: 85% RH). Bean flour was added&nbsp; to the diet in one of eight concentrations: 0, 0.01, 0.1, 0.5, 1, 2.5, 5, 10%). The population growth of <I>Tyrophagus putrescentiae, Acarus siro </I>and <I>Aleuroglyphus ovatus </I>initiating from a density of 50 mites per 0.2 g of diet was recorded for 21 days. The enrichment of grain with bean flour suppressed the population growth of the tested species. These differed in their sensitivity to bean flour. Population growth was decreased to 50% in comparison to the control (rC<SUB>50</SUB>) by the bean flour concentration of 0.02% in <I>T. putrescentiae, </I>0.04% in<I>&nbsp; A. siro, </I>and by<I> </I>4.87% in <I>A. ovatus. </I>&nbsp;The concentration of 5% bean flour in diets kept populations of <I>A. siro </I>and <I>T. putrescentiae</I><I> </I>at the initial level. The results are discussed in the context of applying bean flour in the integrated control of stored-product mites.

ABSTRACT Interactions among endosymbiotic bacteria inside their eukaryotic hosts are poorly understood, particularly in mites. The mite Tyrophagus putrescentiae is a common, medically important generalist species that has many intracellular and gut bacterial symbionts. In the experiments, we examined bacterial abundances and composition in mite populations obtained by controlled mixing of stock mite populations that differed in the presence/absence of the major intracellular bacteria Wolbachia and Cardinium. Changes in microbial communities were characterized using 16S ribosomal RNA high-throughput sequencing (pooled mite individuals) and quantitative PCR for key microbial taxa (individual mites). Mite fitness was estimated as a parameter of population growth. We detected that in mixed mite populations, Cardinium and Wolbachia can co-occur in the same mite individual. The presence of Cardinium was negatively correlated with the presence of Wolbachia and Bartonella, while the Bartonella and Wolbachia were positively correlated in individual level samples. Since mixed populations had lower abundances of Wolbachia, while the abundance of Cardinium did not change, we suggest that the presence of Cardinium inhibits the growth of Wolbachia. The mixed mite populations had lower population growth than parental populations. The possible effect of symbionts on the fitness of mixed population is discussed.

ABSTRACT Tyrophagus putrescentiae is inhabited by bacteria that differ among mite populations (strains) and diets. Here, we investigated how the microbiome and fitness of T. putrescentiae are altered by dietary perturbations and mite populations. Four T. putrescentiae populations, referred to as dog, Koppert, laboratory, and Phillips, underwent a perturbation, i.e., a dietary switch from a rearing diet to two experimental diets. The microbiome was investigated by sequencing the V1-V3 portion of the 16S rRNA gene, and selected bacterial taxa were quantified by quantitative PCR (qPCR) using group/taxon-specific primers. The parameters observed were the changes in mite population growth and nutritional status, i.e., the total glycogen, lipid, saccharide, and protein contents in mites. The effect of diet perturbation on the variability of the microbiome composition and population growth was lower than the effect induced by mite population. In contrast, the diet perturbation showed a greater effect on nutritional status of mites than the mite population. The endosymbionts exhibited high variations among T. putrescentiae populations, including Cardinium in the laboratory population, Blattabacterium-like bacteria in the dog population, and Wolbachia in the dog and Phillips populations. Solitalea-like and Bartonella-like bacteria were present in the dog, Koppert, and Phillips populations in different proportions. The T. putrescentiae microbiome is dynamic and varies based on both the mite population and perturbation; however, the mites remain characterized by robust bacterial communities. Bacterial endosymbionts were found in all populations but represented a dominant portion of the microbiome in only some populations. IMPORTANCE We addressed the question of whether population origin or perturbation exerts a more significant influence on the bacterial community of the stored product mite Tyrophagus putrescentiae. The microbiomes of four populations of T. putrescentiae insects subjected to diet perturbation were compared. Based on our results, the bacterial community was more affected by the mite population than by diet perturbation. This result can be interpreted as indicating high stability of the putative intracellular symbionts in response to dietary perturbation. The changes in the absolute and relative numbers of Wolbachia, Blattabacterium-like, Solitalea-like, and Cardinium bacteria in the T. putrescentiae populations can also be caused by neutral processes other than perturbation. When nutritional status is considered, the effect of population appeared less important than the perturbation. We hypothesize that differences in the proportions of the endosymbiotic bacteria result in changes in mite population growth.

The fumigant pesticide methyl bromide (MB) was used for stored products, but it is now banned for most uses in many countries as an ozone-depleting substance. MB was the only pesticide used to manage the ham mite, Tyrophagus putrescentiae, which is the most significant pest of dry cured hams. Effective alternatives to MB are needed to develop integrated pest management (IPM) programs for this pest. This study evaluated plant essential oils and food-safe compounds as repellents to directly protect hams from infestation. Experiments to assess the repellency to orientation, oviposition, and population growth of mites on pieces of aged country hams were conducted. Test compounds at different concentrations were dissolved in respective solvents and compared to the solvent control. Results showed that C8910, a mixture of three short-chain fatty acids, and the sesquiterpene ketone nootkatone had repellency indices of (RI) of 85.6% and 82.3%, respectively, at a concentration of 0.1 mg/cm2, when applied to a Petri dish arena. DEET and icaridin were also tested but performed poorly with RIs below 70% even at 0.1 mg/cm2.The monoterpene alcohol geraniol had the highest RI of 96.3% at 0.04 mg/cm2. Ham pieces dipped in C8910 and nootkatone at 150 ppm each had RIs of 89.3% and 82.8%, respectively. In general, as the concentrations of test compounds increased, the numbers of eggs that were laid on these treated ham cubes decreased after the 48 h exposure time. Ham pieces dipped in different concentrations of test compounds and then inoculated with 20 adult mites showed a significant decrease in mite population growth compared to control pieces after 14 days. The results of these experiments suggest that some plant secondary metabolites and synthetic food-safe compounds could serve as potential alternatives for managing mites on hams.

39 ilçesiyle Türkiye’nin en büyük nüfusuna sahip ili İstanbul aynı zamanda Türkiye’nin en çok iç göç alan şehridir. Özellikle kırdan kente göç bağlamında sosyo-ekonomik ve demografik özellikleriyle birçok araştırmaya konu olmuştur. Fakat İstanbul yerleşik nüfusunun Türkiye’nin diğer şehirlerine kayıtlı olma yoğunluğu da önemlidir. Bu çalışmanın amacı 2012 ve 2017 yıllarındaki nüfus değişimini göz önünde bulundurarak İstanbul ilçelerinin ayrıntılı nüfus yoğunluğu ve büyüme analizini yapmak, ilgili faktörleri değerlendirmek, hemşehri ağlarını okumak adına yerleşik nüfus kütük bilgileri bakımından inceleyerek elde edilen örüntüler doğrultusunda ilçe tipolojileri oluşturmaktır. Çalışmanın birinci bölümünde ilgili beş yıllık nüfus değişimlerine göre İstanbul ilçe nüfusları analiz edilmiştir. Ardından her bir ilçe için nüfusa kayıtlı olunan kente göre nüfus büyüme hızlarına bakılarak ilçelerin ağırlıklı olarak barındırdığı hemşehri ağları belirlenmiştir. İkinci bölümde ise ilçeler nüfus değişim özelliklerine göre belirli kategorilere ayrılmış ve bu kategoriler doğrultusunda ilçe tipolojileri oluşturulmuştur..ABSTRACT IN ENGLISHA District Level Analysis of Istanbul’s Population Change (2012-2017)Istanbul having the largest population of Turkey with its 39 districts is the most internal-migrant-receiving city in Turkey. Particularly in the context of rural-to-urban migration, Istanbul has been became a subject of various researches with its socio-economic and demographic features. However, the density of Istanbul’s settled population who registered other cities of Turkey is important. The main aim of this study is to analyse population growth of all districts considering the population change between 2012 and 2017, to evaluate the related factors and to develop a district typology by using the data of settled population according to their family registration in the name of reading the current countryman networks. In the first section of the study, district populations of Istanbul are examined regarding the related five-year change. Afterwards, most repeated countryman networks of all Istanbul’s districts are specified regarding the population growth rate of the registered cities. In the latter section of the study, districts were divided into categories regarding the specific population change features which help to create district typology.
