The research was conducted to explore the potency of pluchea leaves-black tea drink as antioxidant at various proportions. The research used a single factor randomized block design of pluchea leaves and black tea proportions, including 100:0; 75:25; 50:50; 25:75; and 0:100% (w/w). Each of it was repeated five times. The parameters observed in this study were secondary metabolites, total phenolic, total flavonoids, free radical DPPH scavenging activity, and iron reducing power. The data were statistically analyzed using Analysis of Variance (ANOVA) at α = 5%, if the analysis showed a significant effect then it was continued with Duncan's Multiple Range Test (DMRT). Data was stated as mean ± standard deviation. The results showed that the secondary metabolites containing in drink from pluchea leaves and black tea at various proportions were alkaloids, flavonoids, phenolics, saponins, tannins, and cardiac glycosides. The increasing of black tea proportion in samples added the intensity of alkaloids, phenolics, flavonoids, saponins, and cardiac glycosides compounds detected, but the tannins were decreased. These secondary metabolites were correlated with total phenolic content (TPC) and total flavonoid content (TFC). The increasing of black tea proportion in drink significantly decreased DPPH free radical scavenging activity and iron ion reducing power, except for 100% black tea proportion. Tannin compounds seems determining antioxidant activity.  Based on coefficient correlation between TPC or TFC and DPPH free radical scavenging activity or iron ion reducing power, the result showed that DPPH free radical scavenging activity was dominantly contributed by TPC and iron ion reducing power was determined by TPC and TFC. ABSTRAKPenelitian ini dilakukan untuk mengetahui potensi minuman daun beluntas teh hitam sebagai antioksidan pada berbagai proporsi. Penelitian ini menggunakan rancangan acak kelompok satu faktor, yaitu proporsi daun beluntas teh hitam meliputi 100:0; 75:25; 50:50; 25:75; dan 0:100% (b/b). Tiap faktor diulang sebanyak 5 kali. Parameter yang diamati pada penelitian ini meliputi kandungan metabolit sekunder, total fenolik (TPC), total flavonoid (TFC), kemampuan menangkal radikal bebas DPPH, dan kemampuan mereduksi ion besi. Data dianalisis secara statistik dengan Analysis of Variance (ANOVA) pada α = 5%, jika terdapat beda signifikan dilanjutkan dengan uji Duncan's Multiple Range Test (DMRT). Data dinyatakan dengan rata-rata ± standar deviasi.  Hasil menunjukkan bahwa metabolit sekunder yang terkandung dalam minuman daun beluntas teh hitam pada berbagai proporsi adalah alkaloid, flavonoid, fenolik, saponin, tannin, dan kardiak glikosida. Peningkatan proporsi teh hitam menambah intensitas senyawa alkaloid, fenolik, flavonoid, saponin, dan kardiak glikosida yang terdeteksi, tetapi kandungan senyawa tannin berkurang. Kandungan metabolit sekunder ini berkorelasi dengan total fenolik (TPC) dan total flavonoid (TFC). Peningkatan proporsi teh hitam dalam minuman menurunkan kemampuan menangkal radikal bebas DPPH dan kemampuan mereduksi ion besi, kecuali pada proporsi teh hitam 100%. Kandungan senyawa tannin dalam minuman menentukan aktivitas antioksidan. Berdasarkan koefisien korelasi antara TPC atau TFC dan kemampuan menangkal radikal bebas DPPH atau kemampuan mereduksi ion besi, hasil menunjukkan bahwa kemampuan menangkal radikal bebas DPPH dominan dikontribusi oleh TPC dan kemampuan mereduksi ion besi ditentukan oleh TPC dan TFC.

Secondary metabolites have been extensively used in the treatment of various health problems. The role of solvent polarity on the phytochemical isolation and antioxidant capacity of Isatis tinctoria (woad) is elusive. In the present study, 14 solvents with different polarity were used in the extraction and total phenolic and flavonoid content (TPC and TFC) investigation. Ferricyanide, phosphomolybdenum, and 2,2-diphenyl-1-picrylhydrazyl (DPPH) methods were used to calculate and compare the antioxidant/free radical scavenging capacity. Our results showed that solvent polarity greatly affects TPC and TFC yield, which is mainly increasing with increasing solvent polarity index and suddenly decreasing at very high polarity. The comparative results showed that TPC is directly correlated with reducing power, antioxidant, and free radical scavenging capacity. Taken together, we conclude that different woad plant parts contain different level of secondary metabolites with a specific polarity that requires a particular solvent with an appropriate polarity index for the extraction. The identification of these biologically active crude extracts and fractions are very important for the basic biological sciences, pharmaceutical applications, and future research for HPLC based active compounds isolation.

Fungal endophytes are the microbes residing in internal tissues of the plant forming symbiotic, mutualistic, communalistic and trophobiotic relationship. Endophytes from medicinal plants are considered as essential source of secondary metabolites accompanied by interesting biological/pharmacological activities. In this study, an effort was made to isolate, characterize endophytic fungi from leaves of Andrographis paniculata and to screen the fungal fractions for in-vitro antioxidant and hepatoprotective activity. Two fungal endophytes (APLF-1 and APLF-2) from Andrographis paniculata were isolated and fermented to get chloroform (A1C, A2C), ethyl acetate (A1EA, A2EA) and n butanol (A1nB, A2nB) extracts. All the endophytic fractions of APLF-1 and APLF-2 were assayed for free radical scavenging properties against 2, 2-diphenyl-1-picrylhydrazyl (DPPH), hydroxyl radical and reducing power. Fractions of APLF-2 showed good scavenging activity compared to APLF-1 extracts. Further, A2EA and A2nB were evaluated for hepatoprotective activity against CCl4 induced hepatotoxicity. A2EA (50 mg/kg &100 mg/kg) and A2nB (50 mg/kg &100 mg/kg) reversed the elevated biochemical parameters with respect to CCl4 treated group (p<0.001). The LPO, SOD and CAT levels were also restored by A2EA and A2nB (100 mg/kg p.o). APLF-1 and APLF-2 were studied for rDNA sequencing by PCR technique. The endophytic fungi, APLF-1 and APLF-2 were identified as Diaporthe sp. A25 and Preussia sp. PPV3.6 respectively based on their morphology and molecular characterization. The presence of polyvalent secondary metabolites in A2EA and A2nB were confirmed by HPTLC analysis. Keyword: Endophytic fungi; Hepatoprotective; Antioxidant; Andrographis paniculata; Diaporthe sp. In this research study, quantitative determination of phytoconstituents in seed coat, dehusked raw dal, cooked dal and whole seed of locally grown pulses viz. pigeonpea, lentil, desi and kabuli chickpea, and ricebean was carried out. Among the four treatments of all the pulses, highest yield in methanol was found in ricebean. Total phenolic content (mg GAEg-1) varied from 1.72-103.62 in all of the extracts. Similarly, flavonoids (mg CAEg-1) ranged from 1.13 to 33.66 which also found to be reduced in dehulled and cooked extracts. o-Dihydric phenols, hydrophobic and hydrophilic phenols followed order as: seed coat> whole seed> raw dal> cooked dal. Results obtained by DPPH method showed that free radical scavenging capacities ranged from 53.80 to 92.00% while antioxidant activity ranged from 51.00 to 78.60% by FTC method. A highly significant correlation (plessthan0.05) between antioxidant activities/free radical scavenging efficiencies and phenolic contents as well as flavonoids was observed in most of the extracts under study. The results concluded that the pulse seed extracts may be valuable natural source of secondary metabolites for nutraceutical industry.

Free radical scavenging and cell aggregation inhibitory activities of 36 secondary metabolites isolated from the methanolic extracts of Verbascum cilicicum Boiss., V. lasianthum Boiss. ex Bentham, V. pterocalycinum var. mutense Hub.-Mor., and V. salviifolium Boiss. (Scrophulariaceae) were investigated. The isolated compounds, 6-O-vaniloyl ajugol (1), ilwensisaponin A (2), ilwensisaponin C (3), verbascoside (4), β-hydroxyacteoside (5), martynoside (6), poliumoside (7), forsythoside B (8), angoroside A (9), dehydrodiconiferyl alcohol-9-O-β-D-glucopyranoside (10), dehydrodiconiferyl alcohol-9′-O-β-D-glucopyranoside (11), apigenin 7-O-β- glucopyranoside (12), luteolin 7-O-β-glucopyranoside (13), luteolin 3′-O-β-glucopyranoside (14) and chrysoeriol 7-O-β-glucopyranoside (15), exhibited a dose-dependent inhibition of bioautographic and spectrophotometric DPPH activities. Verbascoside (4) was the most active (IC50 4.0 μg/ml) comparing it to vitamin C (IC50 4.4 μg/ml) to inhibit phorbol 12-myristate 13-acetate (PMA)-induced peroxide-catalyzed oxidation of 2′,7′-dichlorofluorescein (DCFH) by reactive oxygen species (ROS) within human promyelocytic HL-60 cells. Ilwensisaponin A (2) (MIC 6.9 μg/ml) showed moderate in vitro activity on lymphocyte-associated antigen-1 (LFA-1)/intercellular adhesion molecule-1 (ICAM-1)-mediated aggregation using the HL-60 cell line [positive control was cytochalasin B (MIC 2.3 μg/ml)]. None of the other compounds showed free radical scavenging and cell aggregation inhibitory activities.

Objectives: The present study was carried out to determine the phytochemical constituents and in vitro antioxidant activities of leaf extracts of Alternanthera brasiliana (L). Kuntze (A. brasiliana) and Alternanthera bettzickiana regel (A. bettzickiana).Methods: Preliminary phytochemical screening was performed in petroleum ether, chloroform, acetone, ethanol, hydroethanolic and water extracts of leaves of A. brasiliana and A. bettzickiana. The level of alkaloids, flavonoids, total phenolic content, tannins, Vitamin C, Vitamin E, GSH, and total proteins were determined in hydroethanolic, ethanol, and water extracts. Antioxidant activity of the hydroethanolic leaf extracts of A. brasiliana and A. bettzickiana were determined by 2,2-diphenyl-1-picryl-hydrazyl-hydrate free radical scavenging assay, nitric oxide scavenging assay, superoxide anion scavenging assay, ferric reducing antioxidant power assay, total antioxidant capacity, and reducing power assay.Results: The phytochemical screening of six different extracts of A. brasiliana and A. bettzickiana revealed the presence of various phytonutrients. Quantitative analysis of secondary metabolites in ethanol, hydroethanolic and water extracts of leaves of A. brasiliana and A. bettzickiana showed the presence of high amount of secondary metabolites in the hydroethanolic extract. In vitro, antioxidant assay of two plant extracts revealed that A. bettzickiana was more potent than A. brasiliana in scavenging free radicals.Conclusion: The different extracts from A. brasiliana and A. bettzickiana and specifically the hydroethanolic extract of A. bettzickiana revealed several properties such as rich source of phytonutrients, higher free radical scavenging properties, and significant antioxidant capabilities. Therefore, the bioactive compound should be isolated in future studies and could be used as a safe and serve as a potential source of natural antioxidants.

Cayratia trifolia Linn. Domin Syn. Vitis trifolia (Family: Vitaceae) is commonly known as Fox grape in English; Amlabel, Ramchana in Hindi and Amlavetash in Sanskrit. It is native to Asia and Australia. This plant is used for chronic fever, rheumatic, anti-inflammatory etc. In the present study, quantitative analysis and free radical scavenging activities of stem ethanolic extract of Cayratia trifolia was investigated. The extract was found to possess more secondary metabolites and it exhibit radical scavenging activities, Based on the results it can be concluded that, the stem ethanolic extract of Cayratia trifolia which contains high amount of secondary metabolites and exhibits free radical scavenging activities, phytochemistry from leaves and stems using spectral techniques. This research paper provides information mainly on various biological activities like antimitotic, antidiabetic and anti-implantation and several medicinal uses. Biological activities of few of them have been studied maximum concentration of Proteins (58.4mg/gdw) and minimum concentration in total soluble sugar (0.7mg/gdw) and maximum concentration of Lipid peroxidase (5.01mg/gdw) and minimum concentration in FRAP (0.197mg/gdw). Mangrove, a specialized group of plant communities, provide immense ecological and protective benefits to the coastal areas of the tropical and subtropical world where they thrive. Demographic obligation and climatic hostilities have massively altered their vegetation pattern and, even ruined some key species to large extent. The present study aims to consider Reactive Oxygen Species (ROS) scavenging skills in some degrading mangrove taxa of Indian Sundarbans (Xylocarpus granatum and Heritiera fomes) compared with some opulently growing ones (Excoecaria agallocha, Bruguiera gymnorrhiza and Phoenix paludosa) in increasing salinity gradient, in relation to their sustainability. Non-enzymatic antioxidants (secondary metabolites) Proanthocyanidin and Tannin were estimated and Free radical scavenging ability was evaluated by Singlet Oxygen Scavenging Activity Assay, Reducing power assay, effects on Peroxynitrite, Nitric Oxide Radical Scavenging, Hydrogen Peroxide Scavenging Activity Assay, Reaction with Hypochlorous Acid, Superoxide Radical Scavenging Activity Assay and Hydroxyl Radical Scavenging Activity. Relatively higher occurrence of secondary metabolites and improved antioxidant ability were recorded in E. agallocha, B. gymnorrhiza and P. paludosa; than the other two plants X. granatum and H. fomes; where the trend showed a decline in the ROS scavenging after a certain increase in salinity. Strong positive correlation of both secondary metabolites and radical scavenging ability with salinity pose the three stable taxa more viable in the higher salty environment of the Indian Sundarbans. But relatively less ROS scavenging ability in more saline zones, may be the potential reason for the unfavorable existence of the two red listed plants, which would ultimately lead to gradual natural extinction of them from the Indian Sundarbans.
