Mastitis is one of the diseases with the highest incidence in dairy cows, causing huge economic losses to the dairy industry all over the world. The aim of the study was to characterize mastitic milk metabolome through untargeted nuclear magnetic resonance spectroscopy (1H-NMR). Taking advantage of the high reproducibility of 1H-NMR, we had the opportunity to provide quantitative information for all the metabolites identified. Fifty-four molecules were characterized, sorted mainly into the chemical groups, namely amino acids, peptides and analogues, carbohydrates and derivates, organic acids and derivates, nucleosides, nucleotides and analogues. Combined with serum metabolomic investigations, several pathways were addressed to explain the mechanisms of milk metabolome variation affected by clinical mastitis, such as tricarboxylic acid cycle (TCA cycle) and phenylalanine, tyrosine and tryptophan biosynthesis. These results provide a further understanding of milk metabolome altered by clinical mastitis, which can be used as a reference for the further milk metabolome investigations.

Abstract Introduction: The aim of the study was to evaluate the occurrence of enterococci in inflammatory secretions from mastitic bovine udders and to assess their antimicrobial resistance. Material and Methods: A total of 2,000 mastitic milk samples from cows were tested in 2014–2017. The isolation of enterococci was performed by precultivation in buffered peptone water, selective multiplication in a broth with sodium azide and cristal violet, and cultivation on Slanetz and Bartley agar. The identification of enterococci was carried out using Api rapid ID 32 strep kits. The antimicrobial susceptibility was evaluated using the MIC technique. Results: Enterococci were isolated from 426 samples (21.3%). Enterococcus faecalis was the predominant species (360 strains), followed by E. faecium (35 isolates), and small numbers of others. The highest level of resistance was observed to lincomycin, tetracycline, quinupristin/dalfopristin (Synercid), erythromycin, kanamycin, streptomycin, chloramphenicol, and tylosin. Single strains were resistant to vancomycin and ciprofloxacin. All isolates were sensitive to daptomycin. E. faecalis presented a higher level of resistance in comparison to E. faecium, except to nitrofurantoin. Conclusion: The results showed frequent occurrence of enterococci in mastitic cow’s milk and confirmed the high rate of their antimicrobial resistance.

The aim of this study was to isolate fungi from mastitic milk of dairy cows and to identify fungal microorganisms involved in bovine mastitis. A total of 608 milk samples from clinical mastitis quarters from 580 animals in Northern Greece were collected, during the years 1997-2001. Antibacterial treatment was administrated to 50% of the sampled animals. Forty two (42) fungi were isolated and identified using mycological media and methods. From the 42 fungi isolates, 38 were yeasts and 4 moulds. The yeasts isolated were classified into the genera Candida, Geotrichum, Rhodotorula. From the thirty four (34) Candida species, 14 were identified as G tropicalis and 6 as G krusei. Furthermore, 4 isolates were classified as G pseudotropicalis, 4 as G albicans, 3 as G parapsilosis and 3 as G rugosa. Geotrichum candidum and Rhodotorula spp. were represented with 2 isolates. The isolated moulds were classified into the genus Aspergillus. Fungi were isolated in pure culture from 38 milk samples and in mixed culturewith bacteria from 4. In 510 samples only bacteria were cultured, while in 56 samples no growth of microorganism was observed. The results of the present study indicate that a level of 6.9% of mycotic mastitis is significant and yeasts are apparently implicated in mammary gland pathology causing economic loss.
