ABSTRACTParent-of-origin effects (POEs) in mammals typically arise from maternal effects or from imprinting. Mutations in imprinted genes have been associated with psychiatric disorders, as well as with changes in a handful of animal behaviors. Nonetheless, POEs on complex traits such as behavior remain largely uncharacterized. Furthermore, although perinatal environmental exposures, such as nutrient deficiency, are known to modify both behavior and epigenetic effects generally, the architecture of environment-by-POE is almost completely unexplored. To study POE and environment-by-POE, we employ a relatively neglected but maximally powerful POE-detection system: a reciprocal F1 hybrid population. We exposed female NOD/ShiLtJxC57Bl/6J and C57Bl/6JxNOD/ShiLtJ mice, in utero, to one of four different diets, then after weaning recorded their whole-brain gene expression, as well as a set of behaviors that model psychiatric disease. Microarray expression data revealed an imprinting-enriched set of over a dozen genes subject to POE; the POE on the most significantly affected gene, Carmil1 (a.k.a. Lrrc16a), was validated using qPCR in the same and in a new set of mice. Several behaviors, especially locomotor behaviors, also showed POE. Interestingly, Bayesian mediation analysis suggests Carmil1 expression suppresses behavioral POE, and Airn suppresses POE on Carmil1 expression. A significant diet-by-POE was observed on one behavior, one imprinted gene, and over a dozen non-imprinted genes. Beyond our particular results, our study demonstrates a reciprocal F1 hybrid framework for studying POE and environment-by-POE on behavior.

Abstract Inadequate knowledge of the population structure and diversity present often hamper the efficient use of germplasm collections. Using a high through-put system, twelve microsatellite loci were used to analyze genetic diversity and population structure in a national field genebank repository of 243 cacao accessions grouped into 11 populations based on their known sources. Based on multi-locus profiles, the Bayesian method was used for individual assignment to verify membership in each population, determine mislabeling and ancestry of some important accessions used in breeding program. A total of 218 alleles was revealed with a mean number of 18.2 alleles per locus. Gene diversity (He = 0.70) and allelic richness (4.34 alleles per locus) were highest in the F1 hybrid population. Differential mating system was suggested as responsible for the observed deficit and excess of heterozygotes observed among the populations. Analysis of molecular variance showed that within-population variance accounted for 63.0% of the total variance while the rest 37% was accounted for by the among-population variance. Cluster dendrogram based on UPGMA revealed two main subsets. The first group was made up of the Amelonado/Trinitario ancestry and the other of Nanay/Parinari ancestry. We found that Nanay and Parinari populations were the major source of Upper Amazon genes utilized while a large proportion of genetic diversity in the field genebank remained under-utilized in development of improved cultivars released to farmers in Nigeria. This study showed that the presence of alleles of the Upper Amazon Forasteros (Nanay, Parinari and Iquitos Mixed Calabacillo) genetic materials in the locally available accessions predated the formal large scale introduction of Upper Amazon materials in 1944. This is the first report of population structure of field genebank collections of cacao in Nigeria since more than seven decades of formal cacao breeding research.

Beany flavor of soybean (Glycine max (L.) Merr.) is caused by oxidation of polyunsaturated fatty acids by the action of three lipoxygenases (LOX1, LOX2 and LOX3) present in mature seeds. The unpleasant flavor restricts human consumption of soybean products. This problem could be solved through genetic elimination of alleles that code these enzymes. Parental cultivars and two hybrid population were selected and analyzed using genetic markers for alleles locus, encoding Lox1, Lox2 and Lox3 free. The SSR marker Satt212 confirmed the presence of the homozygous null-allele Lx3 in the cultivar BRS 213, which were used for hybridization with BR 36. Heterozygote F1 hybrid plants and homozygous Lx3 lines in F2 segregating populations were successfully identified.  The SSR markers Sat090 and Sat417 was the most effective diagnostic marker among the all SSR markers tested. Satt090 and Satt417 confirmed the presence of the homozygous Lx2 null-allele in the parental cultivar BRS 213 by flanking Lx2 loci at 3,00 and 2,77 cM, respectively. The presence of Lx2 null allele in The F2 segregating populations between BRS 213 and BRS 155 were successfully identified with a selection efficiency of 98% and have great potential for further application in the Brazilian breeding program aimed at improving soybean seed quality.

In the present investigation, an effort was made to characterize and estimate the variation of 20 tea germplasm on the basis of cytological and anatomical characteristics. All the germplasm showed highly significant difference amongst them in all the parameters except the ploidy level. Here, all the germplasm were observed to be diploid having 2n=30 no. of chromosome. From the anatomical parameters it was observed that THT8 followed by THT5, THT7, THT13 showed characteristic more likely towards Cambod type also recorded with higher phloem index. The planting material noted with higher phloem index in the present study can be considered as hybrid population which may be a good parent material for breeding programme.

Purpose and objectives. To establish the gene interaction types in the performance inheritance, to evaluate the combining abilities and effects of genes for this trait in spring barley accessions. Materials and methods. We investigated 75 F1 spring barley hybrid combinations derived from crossing 25 female forms with three male ones (chaffy and naked). Crossing was conducted in accordance with topcross design. F1 seeds and parents were sown with a cassette breeding planter SKS-6A. Grain pea was the forecrop. The plot area was 0.20 m2. The interrow distance was 0.20 m; the inter-plot tracks were of 0.50 m. Crossings were performed in two replications. Plants were harvested manually, with roots. For structural analysis, 20 typical plants were chosen from each F1 hybrid population, and the performance inheritance was determined by dominance degree (hp). The obtained data were grouped and gene interaction types were determined as per G.M. Beil and R.E. Atkins’s classification. Using two-factor analysis of variance in STATISTICA 10, we found significant differences between the GCA and SCA variances for the performance and evaluated the combining ability effects. Results and discussion. In 2019–2020, the features of 22 spring barley cultivars and three lines were described in terms of the gene interaction types in the performance inheritance and combining ability. On this basis, the prospects of biotype selections were evaluated. In the arid conditions of 2019, F1 only showed positive over dominance, while in favorable 2020 the inheritance types varied from positive to negative overdominance. In the two years, the general combining ability was high in female cultivars Khors and Troian. The male components did not show consistently high GCA for the both years. Hybrids between accessions with a high GCA and accessions with a lower or intermediate GCA may be promising in breeding due appearance of positive transgressions in the offspring. In 2019, the SCA effects were significantly strong in female forms Troian, Datcha, Gladys, Grace, Gatunok, Modern, and Herkules and in male forms Ahrarii and Scrabble. Conclusions. The study found that the gene interaction types in the spring barley performance inheritance depended on cross combinations and growing conditions. In unfavorable 2019, F1 only showed positive overdominance (heterosis), while in favorable 2020, positive overdominance, positive dominance and intermediate inheritance were observed. In 2019–2020, the high GCA was seen in female cultivars Khors and Troian. The strong SCA effects were noticed in female components Troian, Datcha, Gladys, Grace, Gatunok, Modern, and Herkules and male forms Agrarii and Scrabble. The hybrid combinations with the maximum probability of producing transgressive segregants have been selected
