Two tomato (Lycopersicon esculentum Mill.) cultivars, Heinz-2653 and Campbell-28, were grown on Fox loamy sand in the subhumid region of southern Ontario from 1979 to 1982. Irrigation increased the marketable yields of H-2653 in a dry year, 1982, but not in the other years. Irrigation substantially increased marketable yields of C-28 in 1979 and 1982. Irrigation, when the available soil moisture (ASM) level reached 50%, was no more effective than when the ASM level in the soil was allowed to drop to 25%. Without irrigation yield increased as plant population increased in normal and wet years, but not in a dry year. Blossom-end rot (BER) of C-28 cultivar was markedly reduced by irrigation. Effects of irrigation or plant population treatments on the incidence of fruit speck did not appear to be significant.Key words: Available soil moisture, Lycopersicon esculentum, Pseudomonas syringae pv. tomato, fruit speck

A severe fruit disorder of tomato (Lycopersicon esculentum Mill.) was induced with a tobacco form of tobacco mosaic virus isolated from blemished tomato fruit (cv. Ohio WR25) from a glasshouse crop in southwestern Ontario. Symptoms on green tomatoes consisted of slightly sunken, pale or yellowish-brown lesions with a thin, dark green margin. Lesions frequently expanded and coalesced along the flutes. The shoulders of the fruit were the most extensively affected areas. On mature fruit, the lesions appeared either silvery or silvery-brown and generally had a scabby appearance due to cracking and lifting of the surface tissues. Lesions were superficial and internal symptoms were not detected. First-truss inoculations prevented or delayed the fruit disorder. Inoculations at truss stages two, three, and four, however, resulted in development of affected fruit on inoculated as well as lower and subsequent trusses with the exception of the first truss. Similar susceptibilities to the fruit disorder occurred among the glasshouse cultivars Michigan Ohio (Burghart strain), Ohio WR25, Vantage, Veegan, and Vendor. No cross protection against the fruit disorder was provided by a mild tomato form of TMV introduced at the second-leaf stage.

Extracts of ripening tomato (Lycopersicon esculentum L. var. Sunny) were tested for their capacity to hydrolyse carboxymethylcellulose (CMC) and seed xyloglucan (XG) at different pH values in the presence or absence of seed XG oligosaccharide subunits (7 to 9 sugar residues). CMC-ase activities were most active at pH 5 and 7 in locule extracts, whereas XG-ase activity was most active in the presence of XG oligosaccharides at pH 6 in pericarp extracts. The cation exchange FPLC column, Mono S, separated three zones of CMC-ase activity in pericarp extracts. One zone was most active at pH 5, and two at pH 7. The three CMC-ases had some XG-ase activity in the presence of oligosaccharide but, in addition, there were two regions of the eluate with XG-ase but no CMC-ase activity. The presence of XG-ase activity, separate from CMC-ases, was also observed after hydrophobic interaction chromatography, and in protein precipitated by lowering the salt content of extracts. During ripening, the CMC-ases and the oligosaccharide-activated XG-ases in pericarp extracts increased in total and specific activities, while endogenous XG-ase activity, that is in the absence of XG oligosaccharides, remained unchanged or declined. It is concluded that multiple, 1,4-β-glucanase activities develop in ripening tomato fruit, including, in pericarp tissue, a specific XG-ase which is latent and activated by XG oligosaccharides.

Advanced lines and backcrosses from interspecific hybridization of processing tomato (Lycopersicon esculentum Mill.) with six L. chmielewskii (Rick, Kesicki, Fobes and Holle) and L. cheesmanii (Riley) accessions were evaluated in the field over 2 yr for soluble solids content (SSC), total solids content (TSC), fruit yield and fruit size to determine the effectiveness of combining elevated solids levels with high fruit yield and size following early generation selection. More than a quarter of the lines had significantly higher SSC levels than the recurrent tomato parent, Purdue 812 (P812). Several lines combined high SSC and TSC with high fruit yield and large fruit size to produce more dry matter yield per hectare or per fruit than P812. Two backcrosses were optimal for combining high SSC and high yield. SSC and TSC were negatively correlated with fruit size and, to a lesser degree, with fruit yield. The cell-wall structures of tomato (Lycopersicon esculentum Mill) and other fruit are intimately linked with the nature of their polyuronides. Cell-wall polyuronides from unripe and ripe tomato fruit were isolated and purified and their molecular size and molecular-size distributions were compared. It was demonstrated that there is a considerable decrease in the weight-average Mr upon ripening (from 160,000 +/- 10,000 to 96,000 +/- 4000) and a corresponding increase in polydispersity, particularly at the low-Mr end of the distribution. The estimates of polyuronide molecular size and molecular-size distribution were obtained without the need for polyuronide standards of known Mr by using gel-filtration chromatography combined with the absolute method of low-speed sedimentation equilibrium.

This experiment was conducted in faculty of Science labs, Kufa University, carried out during 2015 to applied methods for extraction, purification and Quantitative of Lycopene red pigments, from callus tissue and tomato fruits mother plant (Lycopersicon esculentum Mill).This study include of three parts, Firstly; Tomato seeds(Supper queen) hybrid were germinated in free MS medium and callus induction from shoot tip (3cmpieces) by using MS medium supplemented with Dichlorophenoxiactic acid (2,4-D) at different concentration (0.5,1, 1.5mg/l)with benzyl adenine (BA) at concentration of (0.3 mg/l). Secondly; identically callus fresh weight re-cultured in the same MS medium supplemented with high molecular weight polyethylene glycol (PEG) was used as selective agent at level of (5,10,15 and 25%). Thirdly; comparisons study were made between in vitro and in vivo grown plant. Powder of control lycopene used as standard solution. The content of lycopene was done by using high performance liquid chromatography (HPLC), and compare of the quantitatively of lycopene with these content in fruits of mother plant, and callus tissue. Also, include alcohol extraction of Lycopene from tomato fruit by using acetone and hexane mixture. The result showed significant increased (P< 0.05) of lycopene production and the superiority of lycopene content in callus than the content in fruits of mother plant. Antioxidant enzymes activity like Catalase (CAT),Guaiacol peroxidise (POX) and Superoxide dismutase(SOD) were high in callus under drought stress than in fruit of mother plant. However, Proline and total sugar content were at higher levels in callus under drought stress than in fruit of mother plant.

Tomato (Lycopersicon esculentum Mill., cv. BHN 91) fruit were hand harvested at the pink sage of maturity and stored at 12.5, 20, and 30C in air, 3% O2 + 5% CO2, or 0.5% O2 + 20% CO2 for up to six days. Half of the fruit were inoculated with Fusarium. Control fruit retained the best appearance in 3% O2 + 5% CO2 at both 20 and 30C. Inoculated fruit at 12.5, 20 and 30C in air or 3% O2 + 5% CO2 were acceptable for 12, 3 and 2 days, respectively, but they deteriorated more rapidly compared to fruit held in 0.5% O2 + 20% CO2 as temperature and time increased. Off-odors were present in all 0.5% O2 + 5% CO2 treatments by days 12, 9 and 5 at 12.5, 20 and 30C, respectively. A significant time- and temperature-dependent increase in pH of locular and pericarp tissue, and of supernatant pH occurred in inoculated regions of fruit held in air by days 12, 6 and 3 at 12.5, 20 and 30C, respectively. In contrast, reduced acidity occurred at 9 and 5 days in 3% O2 + 5% CO2 at 20 and 30C, respectively. Generally, increased pH followed a trend with air > 3% O2 + 5% CO2 > 0.5% O2 + 20% CO2.

The feasibility of controlling relative humidity in modified atmosphere packages using compounds possessing Type III sorption isotherm behavior was studied. Ten grams each of dry sorbitol, xylitol, NaCl, KCl, or CaCl2 sealed with one maturegreen tomato (Lycopersicon esculentum L.) fruit at 20C in simulated packages for 48 days resulted in stable relative humidities of ≈75%, 80%, 75%, 85%, and 35%, respectively. Relative humidity was a function of the ratio of chemical to fruit mass. Relative humidities within control packages were in the range of 96% to 100% throughout the experiments. A simple system that uses spunbonded polyethylene pouches for the application of this humidity control method to packages is described. The storage life of packaged red-ripe tomato fruit at 20C was extended from 5 days using no pouch to 15 to 17 days with a pouch containing NaCl, mainly by retardation of surface mold development.
