A new species of Bulbostylis (Cyperaceae), only known from the Brazilian Amazonian coast, is here described and illustrated. Bulbostylis litoreamazonicola was found growing over dunes, in seasonally flooded restinga vegetation, and in humid fields near the mangroves from the State of Pará, Northern Brazil. This is the fifth Brazilian-native species of Bulbostylis lacking a persistent stylopodium on the mature fruit. The new species is mainly characterized by its annual habit, simple anthelate inflorescences, densely hispid to hispidulous longitudinally ribbed scapes, pubescent glumes, and cordiform nutlets.

<em>Studying the fruit age and proper media formulation is one of the important stages in embryo culture of coffee. The data is highly benefical, especially in saving embryos generated from intra- and inter-species crosses that fall prematurely or experience problems in germination. The aim of this study was to determine the suitable age and media formulation for embryo culture of Arabica, Robusta, and Liberica coffee. The study was conducted at the Tissue Culture Laboratory, Indonesian Industrial and Beverage Crops Research Institute from January 2019 to November 2020. Murashige and Skoog (MS) media with growth regulators adapted to embryonic development were used in this study. The three types of coffee divided into 5 groups, namely pinhead, immature, early mature, almost mature, mature, and used as planting material. The research was designed in a completely randomized design with 10 replications, and media formulation as a treatment. The results showed that embryo culture of the three coffee species was conducted successfully, except for pinhead fruit. The older the cultured fruit, the higher the percentage of germination. There is a difference in germination time between the three coffee species. The  medium for embryo culture should be adjusted with the age of the fruit being cultured. Aside from growing embryos, the cultured mature fruit embryos on MS medium given 0.5 mg/l BA can also be used for propagation by utilizing the secondary somatic embryos formation.</em>

AbstractAngiosperms may be distinguished from their gymnosperm peers by their flowers, and thus a flower is a good proxy of fossil angiosperms. However, flowers and their parts are usually too frail to be preserved in the fossil record. This makes the origin of angiosperms and their flowers the foci of controversy in botany. Eliminating such botanical controversies can only be achieved by studying related plant fossils. Applying routine SEM, LM, and MicroCT technologies, we document a fossil flower bud, Florigerminis jurassica gen. et sp. nov., from the Jurassic of Inner Mongolia, China. This fossil includes not only a leafy shoot but also physically connected fruit and flower bud. The developmentally interpolated existence of a blooming flower between the flower bud and mature fruit in Florigerminis suggests that angiosperm flowers were present in the Jurassic, in agreement with recent botanical progress. Florigerminis jurassica underscores the presence of angiosperms in the Jurassic and demands a re-thinking on angiosperm evolution.

A new subspecies of Sideroxylon mascatense (A.DC.) T.D.Penn. in the Western Hajar Mountains, differing from the type subspecies in the colour of the mature fruit, is described. The two colour forms are genetically isolated, showing that these two are distinct taxa.

Light dependent anthocyanin accumulation contributes to the red pigmentation of the fruit skin of mango (Mangifera indica L.). Light-independent pigmentation has also been reported, but remains poorly characterized. In this study, the pigmentation patterns in the skin of two red mango cultivars, ‘Ruby’ and ‘Sensation’, were evaluated. Metabolomic profiling revealed that quercetin-3-O-glucoside, cyanidin-3-O-galactoside, procyanidin B1, and procyanidin B3 are the predominant flavonoid compounds in the skin of ‘Ruby’ and ‘Sensation’ fruit. Young fruit skin mainly accumulates flavonol and proanthocyanidin, while anthocyanin is mainly accumulated in the skin of mature fruit. Bagging treatment inhibited the biosynthesis of flovonol and anthocyanin, but promoted the accumulation of proanthocyanidin. Compared with ‘Sensation’, matured ‘Ruby’ fruit skin showed light red pigmentation at 120 days after full bloom (DAFB), showing a light-independent anthocyanin accumulation pattern. However, the increase of anthocyanin concentration, and the expression of key anthocyanin structural and regulatory genes MiUFGT1, MiUFGT3, and MiMYB1 in the skin of bagged ‘Ruby’ fruit versus ‘Sensation’ at 120 DAFB was very limited. There was no mutation in the crucial elements of MiMYB1 promoter between ‘Ruby’ and ‘Sensation’. We hypothesize that the light-independent anthocyanin accumulation in the skin of mature ‘Ruby’ fruit is regulated by plant hormones, and that ‘Ruby’ can be used for breeding of new more easily pigmented red mango cultivars.

We examined the effects of applications of N, P, K, Mg, and Ca through an irrigation solution and spraying K, Ca, and Mg salts on cucumber powdery mildew (CPM, Podosphaera xanthii) in potted plants and under commercial-like conditions. Spraying CaCl2 and MgCl2, or KCl and K2SO4, decreased CPM. There were significant negative correlations between the anion-related molar concentrations of the salts and disease severity. Among the sprayed treatments, NaCl provided significantly less CPM control when applied at a low (0.05 M) concentration, as compared with CaCl2 and MgCl2. When sprayed applications of Mg and K salts were analyzed separately from the untreated control, the Cl− salts were found to be more effective than the SO4−2 salts. High N and Mg concentrations in the irrigation water delivered to young, fruit-less cucumber plants reduced CPM, whereas more CPM was observed when the irrigation solution contained a medium amount of P and a high amount of K. In contrast, mature, fruit-bearing plants had less severe CPM at higher N, lower P, and higher K levels. Spraying mature plants with monopotassium phosphate, polyhalite (K2Ca2Mg(SO4)4·2H2O), and the salts mentioned above over an entire growing season suppressed CPM. CPM severity was also reduced by spray applications of Ca, Mg, and KSO4−2 and Cl− salts. Spray applications provided better CPM control than fertigation treatments. Induced resistance is probably involved in the effects of nutrients on CPM.

Mulberry (Morus alba L.) leaves and fruit are traditional Chinese medicinal materials with anti-inflammatory, immune regulatory, antiviral and anti-diabetic properties. Melatonin performs important roles in the regulation of circadian rhythms and immune activities. We detected, identified and quantitatively analyzed the melatonin contents in leaves and mature fruit from different mulberry varieties. Melatonin and three novel isoforms were found in the Morus plants. Therefore, we conducted an expression analysis of melatonin and its isomer biosynthetic genes and in vitro enzymatic synthesis of melatonin and its isomer to clarify their biosynthetic pathway in mulberry leaves. MaASMT4 and MaASMT20, belonging to class II of the ASMT gene family, were expressed selectively in mulberry leaves, and two recombinant proteins that they expressed catalyzed the conversion of N-acetylserotonin to melatonin and one of three isomers in vitro. Unlike the ASMTs of Arabidopsis and rice, members of the three ASMT gene families in mulberry can catalyze the conversion of N-acetylserotonin to melatonin. This study provides new insights into the molecular mechanisms underlying melatonin and its isomers biosynthesis and expands our knowledge of melatonin isomer biosynthesis.

Making raisins (dried grapes) is a laborious process and grape growers around the world are increasingly interested in adopting mechanized methods, especially dry-on-vine (DOV). The DOV method entails severing canes bearing mature fruit, causing them to DOV, after which they may be harvested by machine. The overhead arbor is an important DOV raisin trellis system, but basic agronomic performance of raisin varieties on that system are scant. Therefore, a three-year experiment was conducted to compare the performance of several raisin grape varieties (Vitis vinifera L.), on an overhead arbor trellis, for production of DOV raisins. Grapevine variety, ‘DOVine’, ‘Fiesta’, ‘Selma Pete’, or ‘Thompson Seedless’, was the main-plot factor, and number of canes, six or eight, was the sub-plot. ‘Fiesta’ produced the highest yields, averaging about 12.24 Mg/ha raisins. However, ‘Fiesta’ generally had the lowest soluble solids and among the lowest raisin grades. The later ripening of ‘Fiesta’ fruit delayed harvest pruning to initiate fruit drying compared to the other varieties. Therefore, its raisins did not dry sufficiently to meet industry standards. ‘Selma Pete’, ‘DOVine’, and ‘Thompson Seedless’ had similar yields, generally about 20% less than ‘Fiesta’, but ‘Selma Pete’ produced fruit that were the largest in size, with the highest soluble solids, and that produced raisins of the highest grades, compared to the other varieties. Vines pruned to eight canes produced higher yields without sacrificing raisin quality. In conclusion, ‘Fiesta’ was notable for its high productivity, and ‘Selma Pete’ for its earliness and high quality, and both varieties had sufficient capacity to carry eight 15-node canes.

Watermelon is an important cucurbit vegetable crop grown in most states in the United States of America (USA). Phytophthora fruit rot of watermelon caused by Phytophthora capsici has been a major factor, limiting production for the past 15 years in the southeastern USA. USDA-ARS released five Phytophthora fruit rot-resistant germplasm lines for use in breeding programs. These lines were developed by phenotyping using a local isolate of P. capsici from South Carolina. The present study was undertaken to determine if these resistant lines had broad resistance to diverse P. capsici isolates collected from different states and crops. Five resistant germplasm lines (USVL020-PFR, USVL203-PFR, USVL782-PFR, USVL489-PFR and USVL531-MDR) and two susceptible cultivars Sugar Baby and Mickey Lee used as checks were grown in a field in 2014 and 2015 to produce fruit for evaluation. Mature fruit were harvested and placed in a walk-in growth chamber and inoculated with 20 different P. capsici isolates. The chamber was maintained at 26±2°C and high relative humidity (>95%) using a humidifier. All five resistant germplasm lines were significantly more resistant than the two susceptible checks to all 20 P. capsici isolates. Among the five resistant germplasm lines, USVL020-PFR, USVL782-PFR and USVL531-MDR had broad resistance. Some P. capsici isolates induced minor lesions and rot on USVL489-PFR compared to the other resistant lines. Variation in virulence and genetic diversity among the 20 P. capsici isolates was also observed. The five watermelon germplasm lines will be useful for developing commercial watermelon cultivars with broad resistance to P. capsici.
