Potassium-40 was determined in soil in an area with 40,000 m² situated in the western State of Pernambuco, Brazil. For radiometric measurements, the gamma spectrometry method with a high purity germanium (HPGe) detector was used. Sampling of 78 soil samples has been performed at intervals of 25 m. The specific activities of 40K were calculated based on the photopeak of 1.46 MeV. Values from 541 to 3,572 Bq kg-1 were obtained (mean of 1,827 Bq kg-1). These values allowed the determination of the elemental concentrations as well as the absorbed dose rates in air, 1 m above the ground. The values varied from 1.7 to 11.5% (mean of 6%) and from 23.4 to 154.3 nGy h-1 (mean of 79 nGy h-1), respectively.

The gamma dose rates and specific activity of 137Cs, 60Co and 40K insamples of soil taken from places near the landfill radiation at Al-Tuwaitha site were measured using a portable NaI(Tl) detector. Theresults of gamma dose rates in samples were ranged from 52.6nGy.h-1 to 131nGy.h-1. Then the specific activity of 137Cs, 60Co and40K in soil were determined using high pure germanium (HPGe)detector. The specific activities were varied from 1.9 to 115500 Bq.kg-1 for 137Cs, from 6.37 to 616.5 Bq. kg-1 for 60Co, and from 3 to839.5 Bq. kg-1 for 40K. The corresponding health risk for the annualeffective dose equivalent varied from 1.85×10-14 to 15.7mSv/y. Theresults were compared with various international recommendations.

Among radionuclides in the soil deposited after Chernobyl accident, 137Cs poses considerable environmental and radiological problems because of its relatively long half-life (30.17 y), its abundance in the fallout, high mobility and similarity to potassium as the major plant nutrient. In this study the samples of undisturbed surface soil (n=250) were taken from 70 regions in Belgrade, during 2006-2010. The specific activities of 137Cs were measured by gamma-ray spectrometry. Based on obtained results external effective dose rates were calculated according to the internationally accepted activity to dose rate conversion equations. The specific activities of 137Cs were geographically mapped. The presence of 137Cs has been detected in all soil samples, with high variability of its specific activity, ranging from 3 Bq kg-1 to 87 Bq kg-1. The mean specific activity of 137Cs was 23 Bq kg-1 and the corresponding absorbed dose was 1.5 nSv h-1. The observed range reflects the inhomogeneity of the deposition process following the Chernobyl accident. It could also be attributed to topographic differences and spatial differences in physicochemical and biological soil properties, soil type and vegetation cover. The results of the present study could be valuable database for future estimations of the impact of radioactive pollution.

The developing mammalian fetus is thought to be highly sensitive to ionizing radiation. However, dose, dose-rate relationships are not well established, especially the long term effects of protracted, low-dose exposure. A previous report (1) has indicated that bred beagle bitches exposed to daily doses of 5 to 35 R 60Co gamma rays throughout gestation can produce viable, seemingly normal offspring. Puppies irradiated in utero are distinguishable from controls only by their smaller size, dental abnormalities, and, in adulthood, by their inability to bear young.We report here our preliminary microscopic evaluation of ovarian pathology in young pups continuously irradiated throughout gestation at daily (22 h/day) dose rates of either 0.4, 1.0, 2.5, or 5.0 R/day of gamma rays from an attenuated 60Co source. Pups from non-irradiated bitches served as controls. Experimental animals were evaluated clinically and hematologically (control + 5.0 R/day pups) at regular intervals.

Over the past five years it has become evident that radiation damage provides the fundamental limit to the study of blomolecular structure by electron microscopy. In some special cases structural determinations at very low doses can be achieved through superposition techniques to study periodic (Unwin & Henderson, 1975) and nonperiodic (Saxton & Frank, 1977) specimens. In addition, protection methods such as glucose embedding (Unwin & Henderson, 1975) and maintenance of specimen hydration at low temperatures (Taylor & Glaeser, 1976) have also shown promise. Despite these successes, the basic nature of radiation damage in the electron microscope is far from clear. In general we cannot predict exactly how different structures will behave during electron Irradiation at high dose rates. Moreover, with the rapid rise of analytical electron microscopy over the last few years, nvicroscopists are becoming concerned with questions of compositional as well as structural integrity. It is important to measure changes in elemental composition arising from atom migration in or loss from the specimen as a result of electron bombardment.

Diffraction studies in polymeric and other beam sensitive materials may bring to mind the many experiments where diffracted intensity has been used as a measure of the electron dose required to destroy fine structure in the TEM. But this paper is concerned with a range of cases where the diffraction pattern itself contains the important information.In the first case, electron diffraction from paraffins, degraded polyethylene and polyethylene single crystals, all the samples are highly ordered, and their crystallographic structure is well known. The diffraction patterns fade on irradiation and may also change considerably in a-spacing, increasing the unit cell volume on irradiation. The effect is large and continuous far C94H190 paraffin and for PE, while for shorter chains to C 28H58 the change is less, levelling off at high dose, Fig.l. It is also found that the change in a-spacing increases at higher dose rates and at higher irradiation temperatures.

SummaryAn international collaborative study has been carried out to investigate the reproducibility of hirudin assays in 13 laboratories using four recombinant hirudins and one natural, sulphated product. A simple assay procedure was proposed involving the titration of α-thrombin with inhibitor and measurement of residual activity using a chromogenic substrate. A standard α-thrombin preparation was supplied to ensure that this reagent was of uniform quality throughout the study. The method appeared to present no difficulties and laboratories reported similar potencies for the 5 hirudin samples, in line with expected values. This gave 200–222 Thrombin Inhibitory Units/ampoule (TIU/ampoule) of lyophilised hirudin, with geometric coefficient of variation (gcv) values ranging from 10.15–15.97%. This corresponds to specific activities of approximately 14,300–15,900 TIU/mg protein. This is close to the upper limit of previously reported values of specific activity. We conclude that the precision of this determination compared with the wider range of values in the literature (8,000–16,000 thrombin inhibitory units [TIU]/mg) results from the use of good quality standard α-thrombin by all laboratories. This study has important implications for hirudin standardisation.

SummaryIn an attempt to define sequence elements in human and mouse tissue factor (TF) that are responsible for the species specificity observed in their interaction with human factor VIIa (HVIIa), we constructed human-mouse chimeric TF cDNAs, inserted them into plasmid vectors, and induced their expression in E.coli. Assays for procoagulant activity were carried out with the resulting E. coli lysates using (HVIIa) human and mouse (MVIIa). The ratio of the procoagulant activities, HVIIa/MVIIa, revealed that human TF exon 3 was essential for activity when the TF:VIIa complex was formed with HVIIa. By ligating the maltose binding protein (MBP) gene to TF cDNAs it was possible to construct, express and purify MBP-TF chimeras as well as to estimate their specific activities. With selected MBP-TF chimeras and HVIIa we determined kinetic parameters for the activation of human factor X. Replacement of exon 3 in human TF cDNA with the corresponding exon from mouse TF cDNA resulted in both lower affinity for HVIIa and failure to convert bound HVIIa into a potent protease

ABSTRACT The concentration of plasma corticosteroids was followed during major surgery and during the infusion of synthetic human ACTH at dose rates varying from 2400 ng to 15 000 ng per hour. The results showed that the time course of plasma corticosteroids during major surgery was intermediate between that obtained during the infusion of 7500 and 15 000 ng synthetic human ACTH per hour. This gives an estimated ACTH secretion rate during major surgery of between 7500 ng and 15 000 ng per hour.
