The synthesis of a small library of 8-substituted 2-methyl-5,6,7,8-tetrahydroquinoline derivatives is presented. All the compounds were tested for their antiproliferative activity in non-cancer human dermal microvascular endothelial cells (HMEC-1) and cancer cells: human T-lymphocyte cells (CEM), human cervix carcinoma cells (HeLa), human dermal microvascular endothelial cells (HMEC-1), colorectal adenocarcinoma (HT-29), ovarian carcinoma (A2780), and biphasic mesothelioma (MSTO-211H). Compounds 3a, 5a, and 2b, showing significant IC50 values against the whole panel of the selected cells, were further synthesized and tested as pure enantiomers in order to shed light on how their stereochemistry might impact on the related biological effect. The most active compound (R)-5a was able to affect cell cycle phases and to induce mitochondrial membrane depolarization and cellular ROS production in A2780 cells.

The capacity to induce fibrin retraction has been considered a specific property of platelets until recently, when Niewiarowski et al. (Proc. Soc. Exp, Biol. Med. 140, 1199, 1972) observed fibrin retraction induced by human fibroblasts. As a part of a larger study on the interactions of cultured cells with fibrin, we have investigated the ability of the following cell lines to retract fibrin: KB (human oral epidermoid carcinoma); HeLa (human cervix carcinoma); Chang Liver (human, normal epithelium; Chang Conjunctiva (human normal epithelium); NCTC clone 929 (L) (fibroblasts from C3H/AN mice) and BA 1112 (rhabdomyosarcoma developed on WAG/Rji inbred rats). Cells were cultured in Eagle’s MEM, in Hank’s blanced salt solution plus 10% calf serum, removed from the flasks by trypsin treatment and resuspended at a concentration of 2 × 106/ml in Tyrode-albumin solution, containing Ca++ and Mg++. Human citrated platelet-poor plasma was clotted in a test tube at 37° C by thrombin in the presence of either the cell suspensions or buffer. Only BA 1112 cells were able to retract fibrin; the presence of Ca++, cellular integrity and random distribution in the sample were required for this activity. BA 1112 cells were able to modify the structuration of thrombin-induced fibrin as indicated by the marked increase of the maximal amplitude of thrombelastogram. BA 1112-induced fibrin retraction was inhibited by PGE1 and by some pyrimido-pyrimidine derivatives, not by apsirin. No retraction occurred when reptilase instead of thrombin was used as the clotting agent, even if the cells were preincubated with ADP. These results suggest that BA 1112 cells have a susceptibility to thrombin similar to that of platelets; this hypothesis is interesting in view of the muscular origin of these cells.(Supported by Grant NIH-PHRB-10RO1 CA 12764–01.)

Researchers have reported significant effects from Danshen (Salvia miltiorrhiza) in terms of inhibiting tumor cell proliferation and promoting apoptosis in breast cancer, hepatocellular carcinomas, promyelocytic leukemia, and clear cell ovary carcinomas. Here we report our data indicating that Danshen extracts, especially alcohol extract, significantly inhibited the proliferation of the human oral squamous carcinoma (OSCC) cell lines HSC-3 and OC-2. We also observed that Danshen alcohol extract activated the caspase-3 apoptosis executor by impeding members of the inhibitor of apoptosis (IAP) family, but not by regulating the Bcl-2-triggered mitochondrial pathway in OSCC cells. Our data also indicate that the extract exerted promising effects in vivo, with HSC-3 tumor xenograft growth being suppressed by 40% and 69% following treatment with Danshen alcohol extract at 50 and 100 mg/kg, respectively, for 34 days. Combined, our results indicate appreciable anticancer activity and significant potential for Danshen alcohol extract as a natural antioxidant and herbal human oral cancer chemopreventive drug.

The present study probes into the biocompatibility testing of Phycobiliproteins (PBPs) from Gracilaria changii, in-vitro. The cell survivability and cytotoxicity of PBPs to cultured Human Cervix Carcinoma (Hep-2C) cells were tested and observed using MTT (3-(4, 5- Dimethylthiazol-2-yl)-2, 5-diphenyltetrazolium bromide) assay. For ten-fold dilutions, Phycobiliproteins (PBPs) promotes highest cell survivability at 10-4 dilution rate with 239.62% and highest cytotoxic effect, with 40.21%, at 10-1 dilution. The compositions of phycobiliproteins were determined using Ultraviolet (UV-Vis) spectrophotometer giving visible absorbance peaks at 498.5, 614 and 651 nm for the three PBPs composition: Phycoerythrin (PE), Phycocyanin (PC) and Allophycocyanin (APC), respectively. The effects of time duration in centrifugation process of Phycobiliproteins (PBPs) from Gracilaria changii in 3, 5 and 7 minutes, were proven significant on the quantity of the extracted PBPs. The highest average concentration of 20.37 µg/ml for APC, 13.31 µg/ml for PC and 18.57 µg/ml for PE were obtained at the 5 minute-extraction process.
