Abstract A comprehensive atlas of sex steroid distribution in multiple tissues is currently lacking and how circulating and tissue sex steroid levels correlate remains unknown. Here, we adapted and validated a gas chromatography-tandem mass spectrometry method for simultaneous measurement of testosterone (T), dihydrotestosterone (DHT), androstenedione, progesterone (Prog), estradiol, and estrone in mouse tissues. We then mapped the sex steroid pattern in 10 different endocrine, reproductive, and major body compartment tissues and serum of gonadal intact and orchiectomized (ORX) male mice. In gonadal intact males, high levels of DHT were observed in reproductive tissues, but also in white adipose tissue (WAT). A major part of the total body reservoir of androgens (T and DHT) and Prog was found in WAT. Serum levels of androgens and Prog were strongly correlated with corresponding levels in the brain while only modestly correlated with corresponding levels in WAT. After ORX, the levels of active androgens T and DHT decreased markedly while Prog levels in male reproductive tissues increased slightly. In ORX mice, Prog was by far the most abundant sex steroid, and again, WAT constituted the major reservoir of Prog in the body. In conclusion, we present a comprehensive atlas of tissue and serum concentrations of sex hormones in male mice, revealing novel insights in sex steroid distribution. Brain sex steroid levels are well reflected by serum levels and WAT constitutes a large reservoir of sex steroids in male mice. In addition, Prog is the most abundant sex hormone in ORX mice.

SUMMARYLiver samples from 10Schistosomamansoni-infected baboons all yielded eggs but neither their egg counts nor duration of infection (< 226 weeks) correlated with the slightly elevated liver collagen levels or minimal histological fibrosis observed. Approximately 10% of inert 9 and 15 μm diameter microspheres injected into the mesenterio veins of 2 baboons with acute, challengeS. mansoniinfection reached their lungs (mainly 9 μm microspheres). Lung egg recoveries over 1000 were significantly less common among 175 baboons exposed once toS. monsoniinfections than among 56 baboons exposed more than once. Among 221S. mansoni-andS. haematobium-infected baboons, female worm, total tissue egg and lung egg recoveries were significantly correlated with each other but not with liver or (where available) spleen weights. Liver and spleen weights were strongly correlated with total body weight. Baboons did not develop significant liver fibrosis, even after prolonged schistosome infections. However, some liver ‘leakiness’ developed during acute primary and challenge infections, allowing small inert particles and eggs to pass to the lungs, but this ‘leakiness’ was not associated with resistance to challenge. In contrast to mice, such resistance in baboons cannot, therefore, be explained simply in terms of pathological changes due to previous infections.

The importance of radiation damage to the skin and its vasculature was recognized by the early radiologists. In more recent studies, vascular effects were shown to involve the endothelium as well as the surrounding connective tissue. Microvascular changes in the mouse pinna were studied in vivo and recorded photographically over a period of 12-18 months. Radiation treatment at 110 days of age was total body exposure to either 240 rad fission neutrons or 855 rad 60Co gamma rays. After in vivo observations in control and irradiated mice, animals were sacrificed for examination of changes in vascular fine structure. Vessels were selected from regions of specific interest that had been identified on photomicrographs. Prominent ultrastructural changes can be attributed to aging as well as to radiation treatment. Of principal concern were determinations of ultrastructural changes associated with venous dilatations, segmental arterial stenosis and tortuosities of both veins and arteries, effects that had been identified on the basis of light microscopic observations. Tortuosities and irregularly dilated vein segments were related to both aging and radiation changes but arterial stenosis was observed only in irradiated animals.

The heart is generally considered a radioresistant organ, and has received relatively little study after total-body irradiation with doses below the acutely lethal range. Some late damage in the irradiated heart has been described at the light microscopic level. However, since the dimensions of many important structures of the blood vessel wall are submicroscopic, investigators have turned to the electron microscope for adequate visualization of histopathological changes. Our studies are designed to evaluate ultrastructural changes in the mouse heart, particularly in the capillaries and muscle fibers, for 18 months after total-body exposure, and to compare the effects of 240 rad fission neutrons and 788 rad 60Co γ-rays.Three animals from each irradiated group and three control mice were sacrificed by ether inhalation at 4 days, and at 1, 3, 6, 12, and 18 months after irradiation. The thorax was opened and the heart was fixed briefly in situwith Karnofsky's fixative.

The extent to which the Minnesota Child Development Inventory (MCDI), could be used to estimate levels of language development in 2-year-old children was examined. Fifty-seven children between 23 and 28 months were given the Sequenced Inventory of Communication Development (SICD), and at the same time a parent completed the MCDI. In addition the mean length of utterance (MLU) was obtained for each child from a spontaneous speech sample. The MCDI Expressive Language scale was found to be a strong predictor of both the SICD Expressive scale and MLU. The MCDI Comprehension-Conceptual scale, presumably a receptive language measure, was moderately correlated with the SICD Receptive scale; however, it was also strongly correlated with the expressive measures. These results demonstrated that the Expressive Language scale of the MCDI was a valid predictor of expressive language for 2-year-old children. The MCDI Comprehension-Conceptual scale appeared to assess both receptive and expressive language, thus complicating its interpretation.
