Antioxidant status of liver, erythrocytes, and blood serum of rats in acute methanol intoxication

Introduction. The overwhelming number of publications contains only data on the content of individual antioxidants, but not on the overall antioxidant activity of the blood in patients with vibration disease.The aim of the study was to determine the total antioxidant activity of blood serum in patients with vibration disease.Materials and methods. Th e main group consisted of 30 people diagnosed with “Vibration disease” of 1 degree (n=21) and 2 degrees (n=9). Th e control group consisted of 30 clinically healthy men, comparable in age with the main group (p=0.66). Th e total activity of antioxidant systems of blood plasma was evaluated photometrically using the test system “Total antioxidant status-Novo” (“Vector-best”, Russia).Results. The indicator of the total antioxidant status (TAS) was 1,038±0.232 mmol/l in the examined main group, against 1,456±0.225 mmol/l in the examined control group (p<0.000001). Th e coefficient of variation (CV) in patients with vibration disease was 22.35%, 1.45 times higher than in the control group (15.45%). In the main group there was a positive correlation between age and TAS (R=0.525), in the control group there was no such relationship (R=0.095). Th e degree of decrease depended on the severity of vibration disease.Conclusions. 1. The development of vibration disease is accompanied by a decrease in the antioxidant status of blood serum. 2. Th e degree of decrease in the antioxidant status of blood serum correlates with the severity of vibration disease. 3. Reduction of TAS can serve as a pathogenetic justification of the need to include drugs and/or biologically active additives with antioxidant activity in therapy

The objective of this study was to evaluate the effects of dietary supplementation with tannins (TANs) on productive performance, carcass characteristics, meat quality, oxidative stability, and blood serum antioxidant capacity of sheep through a meta-analysis. Using Scopus, Web of Science, ScienceDirect, and PubMed databases, a systematic search was performed for studies published in scientific journals that investigated the effects of TANs supplementation on the variables of interest. Only studies with weaned or older sheep were included. The data analyzed were extracted from 53 peer-reviewed publications. The sheep included in the present study were between 2 and 6 months old, and between 12 and 31 kg of body weight. The effects of TANs were analyzed using random-effects statistical models to examine the standardized mean difference (SMD) between treatments with TANs and control (no TANs). Heterogeneity was explored by meta-regression and a subgroup analysis was performed for covariates that were significant. Supplementation with TANs did not affect dry matter intake, pH, color (L* and b*), Warner–Bratzler shear force, cooking loss and meat chemical composition (p > 0.05). Supplementation with TANs increased daily weight gain (SMD = 0.274, p < 0.05), total antioxidant capacity (SMD = 1.120, p < 0.001), glutathione peroxidase enzyme activity (SMD = 0.801, p < 0.001) and catalase (SMD = 0.848, p < 0.001), and decreased malondialdehyde (MDA) concentration in blood serum (SMD = −0.535, p < 0.05). Supplementation with TANs decreased feed conversion rate (SMD = −0.246, p < 0.05), and the concentration of MDA (SMD = −2.020, p < 0.001) and metmyoglobin (SMD = −0.482, p < 0.05) in meat. However, meat redness (SMD = 0.365), hot carcass yield (SMD = 0.234), cold carcass yield (SMD = 0.510), backfat thickness (SMD = 0.565) and the Longissimus dorsi muscle area (SMD = 0.413) increased in response to TANs supplementation (p < 0.05). In conclusion, the addition of tannins in sheep diets improves productive performance, antioxidant status in blood serum, oxidative stability of meat and some other characteristics related to meat and carcass quality.

Activities of superoxide dismutase (SOD), catalase, glutathione peroxidase (GSH-Px) and glutathione reductase (GSSG-R) and concentration of ascorbate, alpha-tocopherol, non-protein and protein-bound sulfhydryl compounds and thiobarbituric acid-reactive substances (TBA-rs) were measured in liver and serum of rats 6, 12 and 24 h and 2, 5 and 7 days after intoxication with 1.5 g or 3.0 g methanol/kg b.w. Liver GSH-Px and GSSG-R activities and SH-groups and ascorbate content were significantly diminished at 6 and 24 h, while TBA-rs were increased. Serum SOD, GSH-Px and GSSG-R activities and SH-groups concentration were reduced while TBA-rs were elevated. The changes were more intensive after application of the higher dose of methanol. It is concluded that methanol impairs the liver and blood serum antioxidant mechanisms in rats.

By incubation unicellular alga Chlorella vulgaris Biej. in aquaculture with sodium selenite and chrome (III) sulphate there was received and allocated stable lipid and selenium, chrome-lipid substance and studied their effects on oxidative status in healthy rats experiment. Putting substances at a dose of 1.85 mсg of selenium, chrome 1.1 mcg and 0.5 mg lipid per 1 ml of 1 % aqueous starch slurry body in healthy rats every day for 14 days suppressed prooxidative processes activated antioxidant status by glutation system by reduction of catalase and superoxidedismutase activity partly in experimental animals in the liver and in blood serum.

<p>There were obtained and allocated stable selen-lipid and selen-zinc-lipid substances in a result of incubationof unicellular algae Chlorella vulgaris Biej. In aquaculture with sodium selenite and zinc sulfate their influence on oxidative and energetic status of healthy rat was investigated. There were suppressed prooxidative processes,activated antioxidant status, succinate dehydrogenase and cytochrome oxidase activity and glutamate dehydrogenase way of formation of glutamate were increased when substance at a dose of 0.4 mcg of selenium, 2.5 mcg of zinc<br />and 0.5 mg of lipid per 1 ml of 1 % starch-water suspensions was introduced in organisme of healthy rats. Also it contributed of formation and functioning of the active components of the antioxidant system – catalase, ceruloplasmin,<br />reduction glutathione in a liver and in a blood serum of experimental animals.</p>

Repeated administration of methyl alcohol to Rhesus monkeys (Maccaca mulata) by intragastric tube resulted in ultrastructural abnormalities of hepatocytes, which persisted in one animal twelve weeks after discontinuation of the methyl alcohol regime. With dosages ranging between 3.0 to 6.0 gms. of methanol per kg. of body weight, the serum levels attained within a few hours averaged approximately 475 mg. per cent.
