The objective of this study is to prepare nanogels were prepared via charged gellan gum. It was prepared by in situ cross linking reaction between two oppositely charged materials by green method without use of chemical cross linking agents. The prepared nanogels were characterized by Dynamic light scattering, scanning electron microscopy, differential scanning calorimetry and X- Ray diffractometry. The prepared formulation had average particle size of 226 nm with polydispersity index of 0.3. The doxorubicin loaded nanogel demonstrated sustained release for 20 h. The prepared nanogels were hemocompatible and cyctocompatible as revealed by hemocompatibility and MTT assay respectively. All results confirmed that these nanogels can be used for cancer treatment. At this work hydrogel and composite hydrogel systems based on ԑ-polylysine (EPL), hyaluronic acid (HA) and nanocrystalline hydroxyapatite (nHAp) were synthesized via chemical cross-linking method followed by in situ precipitation of nHAp into hydrogel copolymer matrix. Molecular structure, phase composition and morphology of EPL-HA and EPL-HA/nHAp systems were investigated using Fourier transform infrared spectroscopy (FTIR), X-ray powder diffractometry (XRD) and scanning electron microscopy (SEM). The fabricated hydrogels and composite hydrogels were evaluated by hydrogels characteristics such as gel fraction and swelling behavior. This study provides a new insight to develop cutting-edge bioactive hydrogels and composite hydrogels for bone tissue engineering as injectable biomaterials due to beneficial properties of system components.

Abstract Wound management is a major global challenge and a big financial burden to the healthcare system due to the rapid growth of chronic diseases including the diabetes, obesity, and aging population. Modern solutions to wound management include hydrogels that dissolve on demand, and the development of such hydrogels is of keen research interest. The formation and subsequent on-demand dissolution of hydrogels is of keen interest to scientists and clinicians. These hydrogels have excellent properties such as tissue adhesion, swelling, and water absorption. In addition, these hydrogels have a distinctive capacity to form in situ and dissolve on-demand via physical or chemical reactions. Some of these hydrogels have been successfully used as a dressing to reduce bleeding in hepatic and aortal models, and the hydrogels remove easily afterwards. However, there is an extremely wide array of different ways to synthesize these hydrogels. Therefore, we summarize here the recent advances of hydrogels that dissolve on demand, covering both chemical cross-linking cases and physical cross-linking cases. We believe that continuous exploration of dissolution strategies will uncover new mechanisms of dissolution and extend the range of applications for hydrogel dressings.

Mitochondrial protein interactions and complexes facilitate mitochondrial function. These complexes range from simple dimers to the respirasome supercomplex consisting of oxidative phosphorylation complexes I, III, and IV. To improve understanding of mitochondrial function, we used chemical cross-linking mass spectrometry to identify 2,427 cross-linked peptide pairs from 327 mitochondrial proteins in whole, respiring murine mitochondria. In situ interactions were observed in proteins throughout the electron transport chain membrane complexes, ATP synthase, and the mitochondrial contact site and cristae organizing system (MICOS) complex. Cross-linked sites showed excellent agreement with empirical protein structures and delivered complementary constraints for in silico protein docking. These data established direct physical evidence of the assembly of the complex I–III respirasome and enabled prediction of in situ interfacial regions of the complexes. Finally, we established a database and tools to harness the cross-linked interactions we observed as molecular probes, allowing quantification of conformation-dependent protein interfaces and dynamic protein complex assembly.
