Induction of a proteinase K inhibitor in a potato cultivar with a high degree of field resistance against Phytophthora infestans

Wheat is the second most cultivated cereal in the world and is equally important in India. Leaf (brown) rust, caused by Puccinia triticina, was most prevalent among the three rusts in all the wheat-growing areas of India, Bhutan, and Nepal during 2016 to 2019. Leaf rust samples from wheat crops in these countries were pathotyped using the wheat differential genotypes and binomial Indian system of nomenclature. To facilitate international communication, each pathotype identified was also tested on the North American differentials. A total of 33 pathotypes were identified from 1,086 samples, including 3 new pathotypes, 61R47 (162-5 = KHTDM) and 93R49 (49 = NHKTN) from India and 93R57 (20-1 = NHKTL) from Nepal. Two pathotypes, 121R60-1 (77-9/52 = MHTKL) and 121R63-1 (77-5 = THTTM), accounted for 79.46% of the population. Virulence on Lr19 was identified in 0.27% of the samples and from Nepal only. The proportion of pathotype 121R60-1 (77-9 = MHTKL) increased during these years to 57.55%. Virulence was not observed to Lr9, Lr24, Lr25, Lr28, Lr32, Lr39, Lr45, and Lr47 in the population of the Indian subcontinent. Eighteen polymorphic simple sequence repeat (SSR) primer pairs tested on the isolates amplified 48 alleles with an average of 2.66 alleles per primer pair. Based on SSR genotyping, these pathotypes could be grouped into two clades with further two subclades each. Many of the Lr genes present in Indian wheat germplasm (Lr1, Lr3a, Lr10, Lr11, Lr14a, Lr15, Lr16, Lr17, Lr20, Lr23, and Lr26) were ineffective to a majority of the pathotypes. Most of these varieties possessed a high degree of leaf rust resistance. The field resistance of wheat varieties could be attributed to the interaction of genes, unknown resistance, or adult plant resistance.

Defender (A90586-11) is a new late blight-resistant potato cultivar which was released from the Tri-State Potato Variety Development Program in 2004. Conventional and reduced fungicide spray programs were compared on Defender and Russet Burbank (3 years) and Ranger Russet (1 year) in Wisconsin experimental field trials. Useful levels of field resistance to both late blight and early blight were observed in Defender in the absence of fungicide sprays and reduced fungicide input programs. Disease progressed slowest on Defender regardless of fungicide program, relative to Russet Burbank and Ranger Russet. Organic, conventional, and reduced fungicide spray programs also were compared on Defender and Russet Burbank in experimental greenhouse and field tests in Washington. Fungicide spray programs performed similarly on both Defender and Russet Burbank; however, area under the disease progress curve values for no-fungicide treatments were either three times (greenhouse) or six times (field) lower on Defender compared with Russet Burbank. Regardless of the fungicide program, total yield was higher for Defender than Russet Burbank. Mean economic returns associated with Defender also were higher than for Russet Burbank ($6,196 versus $4,388/ha). Fungicide and nonfungicide treatment programs generated similar returns on Defender whereas conventional and reduced fungicide programs generated comparable but higher returns than the nonfungicide program on Russet Burbank.

A potato cysteine protease (cyp) cDNA expressed at an early stage of an incompatible interaction with Phytophthora infestans was isolated. Both the nucleotide and deduced amino acid sequences are highly homologous to those of a tomato cysteine protease, CYP1. Striking protein similarity to all known cathepsins in animals, particularly cathepsin K, was also observed. However, unlike cathepsins, a granulin binding domain is located near the carboxyl terminus of the putative CYP protein. In animals, granulins bind to receptors in the plasma membrane and signal cell growth and division. A ribonuclease protection assay demonstrated that the cyp gene is tightly regulated and is induced 15 h post inoculation with P. infestans in potato leaves either with high field resistance or in which a resistance (R) gene is activated. We conclude that a common signaling pathway is activated in each form of resistance.

ABSTRACTObjective. The purpose of this study was to characterize a population of domestic pig (Sus scrofa domestica) in Cereté, Córdoba, using 20 microsatellite; calculate heterozygosity per locus and average heterozygosity. Materials and methods. Hair samples were collected from 62 specimens. DNA was extracted by proteinase K digestion and phenol-chloroform purification. Information from 20 microsatellites was selected out of those recommended for swine biodiversity studies. PCR products were separated by a vertical polyacrylamide gel electrophoresis. The bands were visualized by staining with silver nitrate. Results. All microsatellites used were polymorphic. Between 3 (SW1067) and 15 (IFNG) alleles were detected with an average number of 6.7 and a total de 134 alleles. The average expected and observed heterozygosities were 0.5278 and 0.5479, respectively. PIC values ranged between 0.1999 and 0.8300 for loci SW1067 and SW911, respectively. Conclusions. Levels of observed and expected heterozygosity found in the present study indicate that the domestic pig (Sus scrofa domestica) in Córdoba Cereté show high degree of genetic variability

Solanum balbocastanum Dun. was collected extensively throughout its range in Mexico and Guatemala. Experimental self- and cross-pollinations showed that the species is highly self-sterile and consists of cross-sterile and cross-fertile individuals. Self-sterility may be due to the presence of incompatibility factors or to triploidy. Authentic hybrids were produced between S. bulbocastanum and S. trifidum Correll, and between S. bulbocastanum and S. pinnatisectum Dun.After inoculation with race 1.2.3.4 of Phytophthora infestans three types of reaction were observed among 1148 seedlings of S. bulbocastanum: immunity with no perceptible lesions, resistance expressed by non-sporulating lesions of the hypersensitive type, and susceptibility indicated by large sporulating necrotic lesions. Seedlings resistant to an isolate of race 1.2.3.4 of Canadian origin did not always show the same level of resistance to a Mexican isolate of the same race. Varying degrees of field resistance were observed among seedling plants, while tuber-propagated plants were generally field immune. Solanum bulbocastanum is considered a mixture of resistant and susceptible genotypes.
