AbstractRheum australe is an endangered medicinal herb of high altitude alpine region of Himalayas and is known to possess anti-cancerous properties. Unlike many herbs of the region, R. australe has broad leaves. The species thrives well under the environmental extremes in its niche habitat, therefore an understanding of transcriptome of R. australe to environmental cues was of significance. Since, temperature is one of the major environmental variables in the niche of R. australe, transcriptome was studied in the species growing in natural habitat and those grown in growth chambers maintained at 4 °C and 25 °C to understand genes associated with different temperatures. A total of 39,136 primarily assembled transcripts were obtained from 10,17,74,336 clean read, and 21,303 unigenes could match to public databases. An analysis of transcriptome by fragments per kilobase of transcript per million, followed by validation through qRT-PCR showed 22.4% up- and 22.5% down-regulated common differentially expressed genes in the species growing under natural habitat and at 4 °C as compared to those at 25 °C. These genes largely belonged to signaling pathway, transporters, secondary metabolites, phytohormones, and those associated with cellular protection, suggesting their importance in imparting adaptive advantage to R. australe in its niche.

The response of root vole (Microtus oeconomus) to least weasel (Mustela nivalis) odours during breeding (August) and nonbreeding seasons (November) was studied in the root voles' natural habitat. The aim of this study was to determine if antipredator behaviour of the root vole changed between breeding and nonbreeding seasons and if this change was closely related to individual environmental cues, e.g., weasel odours. It was found that when bait and weasel odour was used, trappability was greater in breeding season than in nonbreeding season. Moreover, in the breeding season, heavier (i.e., older) voles were more frequently captured than lighter (i.e., younger) ones. Results show that voles' antipredator behaviour of weasel avoidance changes seasonally and that during the breeding season voles display a trade-off conflict between predator risk and foraging, if predator risk is high. It is possible that older voles are driven to reproduce in the current breeding season and thus display less weasel avoidance than do younger voles, which have higher probability of surviving to the next breeding season. Results indicate that indirect (nonlethal) effects of mammalian predators on voles' behaviour strongly depend on age, sexual activity, and season.

Picrorhiza kurroa plants were collected from its natural habitat. In vitro plants were raised from the leaves of high yielding collection screened in an earlier study. Leaves, roots and internodal segments were cultured for 15 days. The effect of thidiazuron (1-phenyl-3-(1, 2, 3- thiadiazol-5-yl) urea; TDZ) pretreatment for 15 days on regeneration potential of different explants viz. leaves, roots and internodes of Picrorhiza kurroa was studied. Regeneration potential varied significantly with the type of explant. Regeneration response of 100% with 46.25 shoots per explant was obtained in leaf segments of 2.0 cm length pretreated with 0.5 µM TDZ for 15 days and then transferred to 2.32 µM kinetin (Kn) containing MS basal medium. In case of root explants maximum shoot number (17.12) was obtained on 0.5 µM TDZ pretreated for 15 days and then to 3.64 µM Kn. Maximum shoots per explants (12.33) were obtained in case of internodes pretreated with 0.5 µM TDZ for 15 days and transferred to 1.16 µM Kn. Regenerated shoots from different explants developed in vitro rooting on MS basal medium within 7-8 days. Conclusively, an efficient and repeatable protocol for rapid regeneration from different explants and in vitro rooting has been developed in P. kurroa which can be effectively used for its conservation.
