"Green" heavily salted split fish may be pre-cooled within a cold storage area with ice and salt, without imposing an excessive load upon storage refrigeration facilities and with little additional labour. Twelve pounds of ice plus 4 lb of salt per 100 lb of fish, uniformly applied between 3- to 4-inch thick layers of fish, will reduce the bulk fish temperature by 25°F (14 °C) in less than 8 hours.No mechanical equipment is required if flake ice be deemed a commodity. The direct cost may be estimated at less than 15 cents per 100 lb of finished (dried) fish.Pre-cooling is not essential under normal conditions but is recommended for heavily salted fish in which the degree of red bacterial contamination is unknown or suspected to be critical, and the storage refrigeration is not adequate to cool the product in a sufficiently short time.

A systematic study, performed from 2017–2020 looked at the rate of positive post-pasteurization B. cereus findings, the quantity of B. cereus in pasteurized banked human milk (PBM), and the rate of B. cereus toxicogenic isolates from PBM. During the study period, 6815.71 L (30,943 tested bottles) of PBM were tested, with an average amount per year of 1703.93 L (7736 tested bottles). The PBM discard rate per year due to bacterial contamination varied between 8.7–10.0% and contamination with B. cereus was the most frequent reason. The total number of B. cereus positive tests was 2739 and the proportion of its positivity from all positive tests was between 56.7–66.6%. The prevalence of B. cereus positive tests rose significantly in the summer months. The production of enterotoxin was found in 3 of the 20 tested samples (15.0%). The B. cereus CFU-quantities in the PBM were below 10 CFU/mL in 80% of cases (16 of 20 samples tested). The quantitative data can be used in the risk assessment of cold storage of PBM at temperatures above zero and manipulation of PBM prior to its administration.

For research in cell differentiation and in vitro toxicology it is essential to provide a natural state of cell structure as a benchmark for interpreting results. Hypothermosol (Cryomedical Sciences, Rockville, MD) has proven useful in insuring the viability of synthetic human epidermis during cold-storage and in maintaining the epidermis’ ability to continue to differentiate following warming.Human epidermal equivalent, EpiDerm (MatTek Corporation, Ashland, MA) consisting of fully differentiated stratified human epidermal cells were grown on a microporous membrane. EpiDerm samples were fixed before and after cold-storage (4°C) for 5 days in Hypothermosol or skin culture media (MatTek Corporation) and allowed to recover for 7 days at 37°C. EpiDerm samples were fixed 1 hour in 2.5% glutaraldehyde in sodium cacodylate buffer (pH 7.2). A secondary fixation with 0.2% ruthenium tetroxide (Polysciences, Inc., Warrington, PA) in sodium cacodylate was carried out for 3 hours at 4°C. Other samples were similarly fixed, but with 1% Osmium tetroxide in place of ruthenium tetroxide. Samples were dehydrated through a graded acetone series, infiltrated with Spurrs resin (Polysciences Inc.) and polymerized at 70°C.

Cooling is the best preservation method than others because the food that has been cooled will remain fresh and will not experience a change in taste, color and aroma, besides all the activities that cause decay will stop so that the cooled food will last longer. (Hartanto, 1984). With the proper cooling engine planning, it can help with spatial adjustments, adjustments to loading, estimation of the power to be used, and budget plans. That is what is commonly called the cooling load calculation. Calculation of cooling load needs to be carried out before planning. This is necessary because the magnitude of the pending load is very influential on the selection of the cooling engine so that the freezing point for preserving food can be accurate. Pendiginan burden is influenced by external and internal factors. With the experimental method, it is obtained the results of the external cooling load as the external cooling load is 11.6 kW, the inner cooling load is 138.8 kW and the performance work coefficient (COP) is 2.
