Polymorphisms of kappa-casein gene and their effects on milk production traits in Holstein, Jersey and Brown Swiss cattle

The objectives of this study were to detect kappa-casein (κ-CN, CSN3, CASK) gene polymorphisms and to investigate the association between the κ-CN gene polymorphisms and milk yield and composition in cattle. For this purpose, a total of 365 blood and milk samples were collected from Holstein, Jersey and Brown Swiss cows. Polymorphisms of κ-CN gene were detected by the PCR and restriction fragment length polymorphism. A 776-bp fragment of exon IV was amplified by PCR and digested with HindIII and HaeIII restriction endonuclease to distinguish allele A, B and E. The analysis of associations between κ-CN genotypes and milk yield and composition were conducted with the use of the general linear model procedure. As a result, allele frequencies were detected as 0.736, 0.357, 0.308 for A allele, 0.186, 0.643, 0.692 for B allele in Holstein, Brown Swiss and Jersey breeds, respectively, and 0.079 for E allele in Holstein. This study also demonstrated the presence of E allele in Turkish Holstein. Although κ-CN genotypes were associated with protein and solids-not-fat content of milk in Hostein and Brown Swiss, they were associated with milk fat content in Jerseys (P < 0.05).

This Research Communication describes the association between genetic variation within the prolactin (PRL) gene and the milk production traits of Italian Mediterranean river buffalo (Bufala mediterranea Italiana). High resolution melting (HRM) techniques were developed for genotyping 465 buffaloes. The association of genetic polymorphism with milk production traits was performed and subsequently the effects of parity and calving season were evaluated. Single nucleotide polymorphisms (SNPs) were identified at exons 2 and 5 and at introns 1 and 2. All the SNPs were in Hardy–Weinberg equilibrium, and statistical analysis showed that the polymorphism of intron1 was significantly (P < 0·05) associated with milk yield, milk protein content and peak milk yield. The average contribution of the intron1 genotype (r2intron1) to total phenotypic variance in milk production traits was 0·09, and the TT genotype showed lower values than CC and CT genotypes. A nonsynonymous SNP was identified in exon 2, which resulted in an amino acid change from arginine to cysteine. Moreover, the polymorphism of exon 2 was associated significantly with milk fat content (P < 0·05), and the buffaloes with TT genotype showed higher total fat content than the buffaloes with CT genotype. These findings provide evidence that polymorphisms of the buffalo PRL gene are associated with milk production traits and PRL can be used as a candidate gene for marker-assisted selection in Italian Mediterranean river buffalo breeding.

Context Genetic selection based on genetic markers for economically important traits in Sfakia sheep. Aims The aim of the present study was to investigate variation in the ovine myostatin gene (MSTN) and calpain 3 gene (CAPN3), and their association with milk-production traits. Methods Records for milk yield, milk fat content, protein content, lactose content, and non-fat solid content, pH and somatic-cell score (log), were obtained from 376 Sfakia ewes. Polymerase chain reaction–single-strand conformational polymorphism (PCR–SSCP) analyses were used to detect variation in intron 1 of MSTN and exon 10 of CAPN3. General linear models were then used to test for associations between the variation in MSTN and CAPN3, and milk-production traits. Key results The SSCP banding patterns for MSTN showed four variants (A1, A2, A3 and A4), which contained nine nucleotide sequence differences. Four SSCP banding patterns (C1, C2, C3 and C4) were observed for CAPN3 and these contained eight nucleotide-sequence differences. The MSTN variation was associated (P &lt; 0.05) with variation in milk yield and non-fat milk solid content. Variation in CAPN3 was associated with milk yield (P &lt; 0.001), fat content (P &lt; 0.05) and lactose content (P &lt; 0.05). Association analyses between the presence/absence of MSTN and CAPN3 variants and milk-production traits showed that a variant of MSTN that had previously between associated with muscle hypertrophy was associated with decreased milk yield (P &lt; 0.05) and a lower non-fat milk solid content (P &lt; 0.01). A CAPN3 variant that had previously been associated with increased sheep-carcass loin lean-meat yield was associated with a decreased milk yield (P &lt; 0.01) and a decreased milk fat content (P &lt; 0.05). Conclusions Our results have provided an insight into the effects of variation in ovine MSTN and CAPN3 on milk-production traits in sheep. Implications To preserve the dual-purpose characteristics of Sfakia sheep, breeding goals should take into account the possible antagonism between meat and milk traits.

Abstract. The objective of this study was to estimate the effect of acylCoA-diacylglycerol-acyltransferase 1 (DGAT1), leptin and thyroglobulin (TG) loci on the milk fat, milk protein and milk yield in Holstein Friesian, Jersey and Hungarian Simmental cows. Leptin and DGAT1 genotypes were determined by qPCR assay, while TG genotypes were identified using the polymerase chain reaction-restriction fragment length polymorphism technique (PCR-RFLP). DGAT1 GC/GC cows had the highest 305-day milk yield values. Difference between AA/AA and GC/GC genotypes was significant (P<0.05). Leptin CC animals produced significantly higher 305-day milk protein percent values (P<0.05) than other genotypes in Hungarian Simmental breed. At TG locus TT cows showed the highest 305-day milk fat percent values, although differences between genotypes proved to be significant (P<0.05) only in Jersey breed.

The aim of this work was to study bioenergetic and ethological indicators in crossbred first-calf heifers compared to purebred first-calf heifers. The research was conducted on the first-calf heifers of Ukrainian Black-Pied dairy breed and first-generation crossbreeds obtained as a result of crossing with Brown Swiss breed, as well as, on the first-calf heifers of Ukrainian Red-Pied dairy breed and first-generation crossbreeds obtained as a result of crossing Ukrainian Red-Pied dairy with Montbéliarde breed. The purebred Black-and Red-Pied first-calf heifers prevailed over crossbreed first-calf heifers on the indicator of milk yield for 305 days of lactation by 106.45 and 218.36 kg, respectively. Accordingly, the indicators of average daily milk yield were also higher by 0.35 and 0.72 kg and the indicators of average daily milk yield adjusted by 4 % fat content per 0.15 and 0.16 kg. At the same time, the indicator of average fat content in milk prevailed in crossbreeds of Black-Pied and Brown Swiss breed by 0.08 % compared to purebred Black-Pied breeds, and in Red-Pied crossbreeds with Montbéliarde breed by 0.16 % compared to purebred Red-Pied breeds. The purebred Black-Pied first-calf heifers had an advantage by 2.04 MJ compared to crossbreeds, while the Red-Pied first-calf heifers had a slight advantage by 0.06 MJ compared to crossbreed first-calf heifers on the indicator of daily cost of exchange energy (EE). The purebred Black-and Red-Pied first-calf heifers had higher cost of exchange energy allocated with milk compared to crossbreed first-calf heifers by 0.42 and 0.82 MJ. They also had higher cost of exchange energy rates for heat products by 0.79 and 0.25 MJ. According to the energy index indicator, purebred Black - and Red-Pied first-calf heifers prevailed over crossbreeds first-calf heifers by 0.16 and 0.07 %, respectively. The same trend was observed in relation to the productive index indicator. The purebred Black-and Red-Pied first-calf heifers had a slight advantage by 0.001 and 0.002 kg adjusted by 4 % of milk fat content per 1 MJ. According to the indicators of daily behavioral reactions, no special differences between a purebred and crossbreed were found.

AbstractThis research was carried out to quantify the effects of a range of variables on milk fat globule (MFG) size for a herd of Holstein-Friesian cows managed through an automatic milking system with year-round calving. We hypothesised that the overall variation in average MFG size observed between individual animals of the same herd cannot sufficiently be explained by the magnitude of the effects of variables that could be manipulated on-farm. Hence, we aimed to conduct an extensive analysis of possible determinants of MFG size, including physiological characteristics (parity, days in milk, days pregnant, weight, age, rumination minutes, somatic cell count) and milk production traits (number of milkings, milk yield, fat yield, protein and fat content, fat-protein ratio) on the individual animal level; and environmental conditions (diet, weather, season) for the whole herd. Our results show that when analysed in isolation, many of the studied variables have a detectable effect on MFG size. However, analysis of their additive effects identified days in milk, parity and milk yield as the most important variables. In accordance with our hypothesis, the estimated effects of these variables, calculated using a multiple variable linear mixed model, do not sufficiently explain the overall variation between cows, ranging from 2.70 to 5.69 µm in average MFG size. We further show that environmental variables, such as sampling day (across seasons) or the proportion of pasture and silage in the diet, have limited effects on MFG size and that physiological differences outweigh the effects of milk production traits and environmental conditions. This presents further evidence that the selection of individual animals is more important than the adjustment of on-farm variables to control MFG size.

Abstract. A study where Casein loci were used to be markers for improving milk yield, fat content and protein yield in a marker assisted selection experiment focused on possible side effects of this process on longevity traits. Using the data of two samples of cows a Simulation on the data was carried out evaluating the selection differences in period of use, period of life and production of the whole life indirectly. The selection criteria were flexible using one of the milk production traits each as well as marker information. During the investigated period no negative effect of simulated selection using EBV for milk yield, fat content and protein yield alone or in combination with the Casein loci used as markers on longevity traits occurred in the population. There are no significant differences regarding the longevity traits caused by selection for Single milk trait based on pure EBV and MAS.

Abstract The Signal Transducer and Activator of Transcription 5A (STAT5A) gene involved in activating the transcription of milk protein genes was predicted to be influencing milk production traits. The present study was undertaken to investigate the suitability of the polymorphism of STAT5A as a marker for milk traits in Ongole, crossbred cattle and Murrah buffaloes from Southern India. Blood samples (n = 502) for DNA isolation and milk samples (n = 222) from different genetic groups were collected from various farms. The gene variants upon polymerase chain reaction–restriction fragment length polymorphism (PCR-RFLP) on the exon 7 region of STAT5A were subjected to GLM analysis to evaluate their association with milk production traits. The frequencies of C and T alleles at the STAT5A/AvaI locus were 0.98 and 0.02 (Jersey crossbred), 0.94 and 0.06 [Holstein-Friesian (HF) crossbred], 0.97 and 0.03 (Ongole). T allele was not observed in Murrah buffaloes. The least squares mean lactation milk yield of CC and CT genotypes of STAT5A were 2,096.90 ± 48.63 and 2,294.41 ± 215.85 kg in Jersey crossbred, 2,312.92 ± 91.01 and 2,392.82 ± 207.66 kg in HF crossbred and 528.40 ± 22.10 and 396.37 ± 76.17 kg in Ongole cattle, respectively. The milk fat content of the CC genotype was higher (P > 0.05) in Jersey crossbred cattle. The CT genotypes of Ongole and HF crossbred cattle recorded a higher fat per cent than the CC genotypes. Significant associations were not observed in support of STAT5A as a marker for milk production traits in either Ongole or crossbred cattle of indicine admixture and no reason could be found to consider this locus as universal markers for milk production traits in indicine cattle and buffaloes. Considering the monomorphic nature of the gene in buffaloes and their higher milk fat content as compared to bovine milk, much remains to be explored regarding the underlying differences across the bovine and the bubaline species.

CD4+T cells play a key role in the immune response of pathogen-induced mastitis in dairy cattle. Mammary gland factor STAT5b is involved in the regulation of CD4+T cell differentiation during inflammatory response and milk production. Little is known about the genetic variation effects of bovineCD4andSTAT5bgenes on somatic cell score (SCS) and milk production traits in dairy cattle. The aim of the study was to investigate the single nucleotide polymorphisms (SNPs) of bovineCD4andSTAT5bin Chinese Holsteins and to analyse their association with estimated breeding values (EBVs) for SCS and milk production traits. In the present study, SNPs ofCD4(NC_007303 g.13598C>T) andSTAT5b(NC_007317 g.31562 T>C) were identified and genotyped in Chinese Holstein population. The results showed that both SNPs were significantly associated with the EBVs for milk yield and protein yield in Chinese Holstein cows, and the SNP inCD4was associated with the EBV for SCS (P<0·01). The additive effect ofCD4SNP on protein yield was significant (P<0·05), and the dominant effect ofSTAT5bSNP was significant on milk yield and protein yield (P<0·01). Cows with combination genotype C7 (CCTT:CD4g.13598C>T andSTAT5bg.31562 T>C) had the highest SCS EBV but lower milk yield, while cows with C2 (TTTC) produced more milk, fat and protein than the other eight combination genotypes. These results suggested that the SNPs inCD4andSTAT5bmay be potential genetic markers for SCS and milk/protein yields selecting and warrant further functional research.

Abstract. The aim of this study was to estimate the effect of variation in the fatty acid binding protein 4 gene (FABP4) on milk production traits in Greek Sfakia sheep. Polymerase chain reaction – single-stranded conformational polymorphism (PCR-SSCP) analysis was used to genotype a total of 374 Sfakia ewes for two regions of FABP4 located around exon 2–intron 2 (Region 1) and exon 3–intron 3 (Region 2). Each month, for a period of 6 months, milk samples were collected from the ewes to measure total milk yield, fat content, protein content, lactose content, non-fat solid content, pH, and somatic cell count (SCC). A general linear model was used to test the association between the variation observed in FABP4 and milk production traits. Four gene variants (A1–A4) were found in Region 1 and two variants (C1–C2) were found in Region 2. In the first region, the FABP4 genotype significantly affected (P<0.05) non-fat solid levels, fat content, and SCC. The presence of the A2 variant was significantly associated (P<0.05) with decreased SCC, while the presence of A4 was significantly associated with decreased milk yield (P<0.01), increased non-fat solid content (P<0.05), decreased fat content (P<0.01), increased lactose content (P<0.05), and increased pH (P<0.05). In the second region, FABP4 genotype had an effect (P<0.05) on protein content and the presence of the C2 variant was associated (P<0.05) with increased protein content, decreased SCC, and lower pH. The results suggest an association between variation in ovine FABP4 and milk production traits in Greek Sfakia sheep. Nevertheless, further analyses in independent sheep populations of increased size will strengthen these findings.
