Background and Aim: Marker-assisted selection has many advantages over conventional selection in animal breeding. The candidate gene approach has been applied to identify genetic markers associated with economically important traits in livestock. This study was established to investigate variation in the hormone-sensitive lipase (HSL) and leptin receptor (LEPR) genes, and their association with growth traits in Barki lambs. Materials and Methods: Records for birth weight (BW), pre-weaning average daily gain (ADG1), weaning weight (WW), post-weaning average daily gain (ADG2), and marketing weight (MW) were obtained from 247 Barki lambs. Polymerase chain reaction–single-stranded conformational polymorphism analyses were used to detect variation in exon 9 of HSL and exon 19 of LEPR. General linear models were used to test for associations between the variation in ovine HSL and LEPR, and growth traits. Results: The SSCP banding patterns for HSL showed three variants (H1, H2, and H3), which contained two nucleotide-sequence differences (c.1865C>T and c.2038T>C). Two SSCP banding patterns (L1 and L2) were observed for LEPR and these contained two nucleotide-sequence differences (c.2800G>A and c.2978C>G). The HSL genotype showed no effect on the studied traits. The LEPR genotype was proven to have significant effects (p<0.05) on ADG2 and MW. The presence of the L1 variant was associated (p<0.01) with decreased ADG2 and MW. Conclusion: The finding of an association between LEPR gene variation and growth rate after weaning in Barki lambs warrants efforts to improve this trait.

AbstractThe aim of this research was to identify variation in the yak lipin-1 gene (LPIN1) and determine whether this variation affects milk traits. PCR-single stranded conformational polymorphism (PCR-SSCP) analysis was used to detect variation in the 5′ untranslated region of LPIN1 in 500 yaks from four populations: Tianzhu white yaks, Qinghai yaks, wild × domestic-cross yaks and Gannan yaks. Four unique PCR-SSCP patterns, representing four different DNA sequence variants (named A, B, C and D), were observed. These contained six single nucleotide polymorphisms. Female Gannan yaks with BC genotype produced milk with a higher fat content (P < 0.001) and total milk solids (P < 0.001), than those with the AA, AB and BB genotypes. These results would suggest that LPIN1 is having an effect on yak milk fat synthesis.

Fatty acid synthase (FASN) is an enzyme involved in the synthesis of fatty acids (FA) and plays a central role in de novo lipogenesis in mammals. This study was conducted to ascertain the relative level of expression of the FASN gene (FASN) in tissues from the yak (Bos grunniens), and to search for variation in two regions of yak FASN using polymerase chain reaction single-stranded conformational polymorphism (PCR-SSCP) analyses; it also ascertains whether that variation is associated with yak milk traits. The gene was found to be expressed in twelve tissues, with the highest expression detected in the mammary gland, followed by subcutaneous fat tissue. Two regions of the gene were analyzed in 290 Gannan yaks: A region spanning exon 24-intron 24 and a region spanning exon 34. These regions both produced two PCR-SSCP patterns, which, upon sequencing, represented different DNA sequences. This sequence variation resulted from the presence of three nucleotide substitutions: c.4296+38C/T (intron 24), c.5884A/G, and c.5903G/A, both located in exon 34. The exon 34 substitutions would result in the amino acid substitutions p.Thr1962Ala and p.Gly1968Glu if expressed. Four haplotypes spanning from the exon 24-intron 24 region to exon 34 were identified. Of these, two were common (A1-A2 and B1-A2), and two were rare (A1-B2 and B1-B2) in the yaks investigated. The presence of A1-A2 was associated with an increase in milk fat content (p = 0.050) and total milk solid content (p = 0.037), while diplotype A1-A2/B1-A2 had a higher milk fat content (p = 0.038) than the other diplotypes. This study suggests that further characterization of the FASN gene might provide for an improved understanding of milk traits in yaks.

Abstract. Variation in KRTAP6-1 has been reported to affect wool fibre traits in Merino cross-breed sheep and Chinese Tan sheep, but little is known about whether these effects persist in other breeds. In this study, variation in KRTAP6-1 was investigated in 290 New Zealand (NZ) Romney ewes sired by 16 different rams. Polymerase chain reaction single-stranded conformational polymorphism (PCR-SSCP) analysis revealed four variants (A, B, E and F) of KRTAP6-1, and nine genotypes (AA, AB, AE, AF, BB, BE, BF, EE and FF) in these ewes. Among the 243 ewes that had genotypes with a frequency of over 5 % (i.e. AA, AB and BB), the presence of A was found to be associated with reduced mean fibre diameter (MFD) and increased coefficient of variation in fibre diameter (CVFD), whereas the presence of B had a trend of association with decreased coarse edge measurement (CEM). A genotype effect was also detected for MFD and CVFD. No associations were detected for fibre diameter standard deviation (FDSD), mean fibre curvature (MFC) and medulation. These results suggest that variation in KRTAP6-1 affects wool fibre diameter in NZ Romney ewes, confirming the finding in Merino cross-breed sheep.

AbstractKRTAP8-1 was the initial high-glycine-tyrosine keratin-associated protein gene recognized in sheep, but little is known about the functional influence of this gene. The current study used polymerase chain reaction-single stranded conformational polymorphism analysis to genotype KRTAP8-1 in 391 Southdown × Merino-cross sheep from six sire-lines. Five previously described variants (named A to E) of KRTAP8-1 were identified with frequencies of 67.0, 14.2, 7.0, 10.7 and 1.0%, respectively. Of the four variants (A, B, C and D) that occurred at a frequency greater than 5%, the presence of C was found to be associated with a reduction in mean fibre curvature (MFC) and the presence of D was associated with an increase in mean staple strength (MSS), whereas the presence of A had a trend of association with reduced MSS. Associations were not identified with other wool traits. These results suggest that variation in KRTAP8-1 affects MSS and MFC, and that KRTAP8-1 has the potential to be used as a genetic marker for improving these traits.

Abstract. The aim of this study was to estimate the effect of variation in the fatty acid binding protein 4 gene (FABP4) on milk production traits in Greek Sfakia sheep. Polymerase chain reaction – single-stranded conformational polymorphism (PCR-SSCP) analysis was used to genotype a total of 374 Sfakia ewes for two regions of FABP4 located around exon 2–intron 2 (Region 1) and exon 3–intron 3 (Region 2). Each month, for a period of 6 months, milk samples were collected from the ewes to measure total milk yield, fat content, protein content, lactose content, non-fat solid content, pH, and somatic cell count (SCC). A general linear model was used to test the association between the variation observed in FABP4 and milk production traits. Four gene variants (A1–A4) were found in Region 1 and two variants (C1–C2) were found in Region 2. In the first region, the FABP4 genotype significantly affected (P<0.05) non-fat solid levels, fat content, and SCC. The presence of the A2 variant was significantly associated (P<0.05) with decreased SCC, while the presence of A4 was significantly associated with decreased milk yield (P<0.01), increased non-fat solid content (P<0.05), decreased fat content (P<0.01), increased lactose content (P<0.05), and increased pH (P<0.05). In the second region, FABP4 genotype had an effect (P<0.05) on protein content and the presence of the C2 variant was associated (P<0.05) with increased protein content, decreased SCC, and lower pH. The results suggest an association between variation in ovine FABP4 and milk production traits in Greek Sfakia sheep. Nevertheless, further analyses in independent sheep populations of increased size will strengthen these findings.

AbstractKeratins are the main structural proteins of wool fibres, and it is thought that variation in the keratins may affect wool fibre characteristics. Polymerase chain reaction-single stranded conformational polymorphism (PCR-SSCP) analyses were used to investigate four regions of the ovine keratin gene KRT31 including a portion of the promoter, the exon 1, exon 3 and exon 7 regions. Initially, in a screening panel of 300 New Zealand Romney, Merino and White Dorper sheep obtained from 26 farms, three, two, two and two PCR-SSCP banding patterns were observed for these four regions, respectively. The promoter region, the exon 1 and exon 3 regions contained two single nucleotide polymorphisms (SNPs) and the exon 7 region contained one SNP. The effect of the variation found in the promoter region on wool traits was subsequently investigated in 485 Southdown × Merino-cross lambs from seven sire-lines. The three variants identified in the original 300 sheep (named A, B and C) were observed with frequencies of 56, 29 and 15%, respectively. The presence of A and B had no significant effect on wool traits, but the presence of C was found to be associated with an increase in greasy fleece weight (GFW), clean fleece weight (CFW) and mean staple length (MSL). There was an effect of genotype on CFW and MSL, with BC sheep producing wool of higher CFW and MSL than AA, AB, AC and BB sheep. These results suggest that ovine KRT31 might be a useful candidate gene for improving wool traits.

For the association between of Follicle stimulating hormone receptor (FSHR) gene (partial part of exon 10) polymorphisms and litter size trait in Egyptian Ossimi sheep, polymerase chain reaction-single stranded conformational polymorphism (PCR-SSCP) and DNA sequencing techniques were developed. Fifty female Ossimi sheep reared under Egyptian conditions were selected according to their litter size. DNA from blood samples of these animals was isolated to amplify 250-bp of the FSHR gene influencing litter size production trait in sheep. Based on litter size, 50 animals were selected from the highest to the lowest litter size productivity during three seasons. PCR-SSCP analysis of the FSHR gene (250-bp) showed two various genotypes AA and with frequencies 0.64 and 0.36, respectively. The frequencies of the A and B alleles were 0.82 and 0.18, respectively. PCR fragment of FSHR gene (191-bp) was sequenced only in the high and low litter size productivity animals (GenBank accession numbers from MG973191 to MG973207, sequentially). The result indicated that 6SNPs (G/71, G/72, G/77, A/110, A/111, A/191) in high fertile animals, while, 10 SNPs (T/1, C/2, T/14, A/69, A/70, A/71, A/74, G/74, A/75, A/136) have found in low fertile animals. Statistically, AA and genotypes have no significant differences (p> 0.05) on litter size trait in Ossimi sheep. FSHR (exon 10) locus was moderate polymorphic (PIC= 0.25) and it can be used for high litter size productivity in Ossimi sheep as a marker-assisted selection (MAS).
