Reliability of the sulfur hexafluoride tracer technique for methane emission measurement from individual animals: an overview

Measurements of enteric methane (CH4) emissions from individual animals have traditionally been made with indirect calorimetry techniques, which are both accurate and reliable. However, the expense and need for animal training and the extent to which calorimetric results can be extrapolated to free-ranging animals have been questioned and stimulated the development of the sulfur hexafluoride (SF6) tracer technique. The tracer technique is now widely used in New Zealand and many other countries for CH4 emission measurements on grazing and pen-fed cattle, sheep, deer and alpacas. Few studies with cattle and sheep have examined the validity of the SF6 tracer technique. Most of these studies have concluded that estimations of CH4 emission by this technique do not differ from those of calorimetric techniques, though some exceptions have been reported. There is general agreement that the tracer technique is associated with large between-animal variability in the CH4 emission estimates from animals on the same diet, but it remains unknown whether this is due to the environment, housing conditions or the technique itself. High within-animal variability has also been reported from tracer CH4 measurements. There is growing evidence that CH4 emission estimates by the tracer technique are positively influenced by the permeation rate (PR) of the SF6 gas from permeation tubes and it has been suggested that fate of the tracer in the rumen rather than unrepresentative breath sample collection is the likely reason for the latter. It is concluded that although some issues related to the tracer technique need to be clarified, using a narrow range in PR and balancing of PR between treatments should be practised in order to overcome the relationship between PR and CH4 emission estimates.

Daily methane (CH4) emissions and dry matter intake (DMI) were measured on 14 mature ewes (3–4 years old) and 13 lambs when the lambs were 13, 17, 25 and 35 weeks of age. During the four CH4 measurement periods, all animals were kept in individual metabolism cages and fed pasture cut daily and fed at 1.5 times maintenance. Feed was offered in equal amounts at 0800 and 1500 hours daily. Methane emissions were measured using the sulfur hexafluoride tracer technique and values reported were the mean of measurement on 4–5 days for each animal. In the intervals between CH4 measurements, ewes and lambs grazed separate paddocks containing predominantly ryegrass. Daily CH4 emissions for the ewes ranged from 21.5 to 22.5 ± 1.50 g/day and were significantly higher than those of the lambs which ranged from 10.7 to 17.5 ± 1.50 g/day. Averaged across all four periods, the overall mean CH4 emission for lambs was 8% lower (P < 0.05) than for ewes (21.9 v. 23.8 ± 0.95 g CH4/kg DMI). However, within each measurement period, the emissions of CH4/kg DMI from lambs was significantly lower (P < 0.05) than those of ewes only in the fourth period when the lambs were 35 weeks of age (17.9 v. 21.9 g/kg DMI for lambs and ewes, respectively). The pasture offered to both ewes and lambs in this period was of higher quality than in the other periods (organic matter digestibility of 80% DM v. 68–71% DM at other times) and CH4 emission per kg DMI was lower in both groups of animals than in the other periods. This study supports the hypothesis that young sheep have lower CH4 emissions per unit of intake than mature animals. However, the age at which the lambs produced similar CH4/kg DMI to adult sheep could not be determined precisely because of the changes in pasture quality between different measurement periods.

To identify relationships between animal, dietary and management factors and the resulting methane (CH4) emissions, and to identify potential mitigation strategies for CH4 production, it is vital to develop reliable and accurate CH4 measurement techniques. This review outlines various methods for measuring enteric CH4 emissions from ruminants such as respiration chambers (RC), sulphur hexafluoride (SF6) tracer, GreenFeed, sniffer method, ventilated hood, facemask, laser CH4 detector and portable accumulation chamber. The advantages and disadvantages of these techniques are discussed. In general, RC, SF6 and ventilated hood are capable of 24 h continuous measurements for each individual animal, providing accurate reference methods used for research and inventory purposes. However, they require high labor input, animal training and are time consuming. In contrast, short-term measurement techniques (i.e., GreenFeed, sniffer method, facemask, laser CH4 detector and portable accumulation chamber) contain additional variations in timing and frequency of measurements obtained relative to the 24 h feeding cycle. However, they are suitable for large-scale measurements under commercial conditions due to their simplicity and high throughput. Successful use of these techniques relies on optimal matching between the objectives of the studies and the mechanism of each method with consideration of animal behavior and welfare. This review can provide useful information in selecting suitable techniques for CH4 emission measurement in ruminants.

AbstractThe objective of this study was to determine whether feeding tannin-containing hays to heifers and mature beef cows influences enteric methane (CH4) emissions and nitrogen (N) excretion relative to feeding traditional legume and grass hays. Fifteen mature beef cows (Exp. 1) and 9 yearling heifers (Exp. 2) were each randomly assigned to treatment groups in an incomplete bock design with 2 periods and 6 types of hays with 3 hays fed each period (n = 5 cows and 3 heifers per treatment). Groups were fed tannin-containing [birdsfoot trefoil (BFT), sainfoin (SAN), small burnet (SML)] or non-tannin-containing [alfalfa (ALF), cicer milkvetch (CMV), meadow bromegrass (MB)] hays. Each period consisted of 14 d of adjustment followed by 5 d of sample collection. Nine cows and 9 heifers were selected for the measurement of enteric CH4 emissions (sulfur hexafluoride tracer gas technique), and excretion of feces and urine, while dry matter intake (DMI) was measured for all animals. The concentration of condensed tannins in SAN and BFT was 2.5 ± 0.50% and 0.6 ± 0.09% of dry matter (DM), respectively, while SML contained hydrolyzable tannins (4.5 ± 0.55% of DM). Cows and heifers fed tannin-containing hays excreted less urinary urea N (g/d; P &lt; 0.001) and showed lower concentrations of blood urea N (mg/dL; P &lt; 0.001) than animals fed ALF or CMV, indicating that tannins led to a shift in route of N excretion from urine to feces. Additionally, cows fed either BFT or CMV showed the greatest percentage of retained N (P &lt; 0.001). Enteric CH4 yield (g/kg of DMI) from heifers (P = 0.089) was greatest for MB, while daily CH4 production (g/d) from heifers (P = 0.054) was least for SML. However, digestibility of crude protein was reduced for cows (P &lt; 0.001) and heifers (P &lt; 0.001) consuming SML. The results suggest that tannin-containing hays have the potential to reduce urinary urea N excretion, increase N retention, and reduce enteric CH4 emissions from beef cattle. The non-bloating tannin-free legume CMV may also reduce environmental impacts relative to ALF and MB hays by reducing N excretion in urine and increasing N retention.

AbstractGenerally, nonreproductive sex is thought to act as “social grease,” facilitating peaceful coexistence between subjects that lack close genetic ties. However, specifc nonreproductive sexual behaviors may fulfill different functions. With this study, we aimed to test whether nonreproductive mounts in Barbary macaques are used to 1) assert dominance, 2) reinforce social relationships, and/or 3) solve conflicts. We analyzed nonreproductive mounts (N = 236) and postmount behavior in both aggressive and nonaggressive contexts, in 118 individuals belonging to two semi-free-ranging groups at La Montagne des Singes (France). As predicted by the dominance assertion hypothesis, the probability to be the mounter increased with rank difference, especially in aggressive contexts (increasing from 0.066 to 0.797 in nonaggressive contexts, and from 0.011 to 0.969 in aggressive contexts, when the rank difference was minimal vs. maximal). The strength of the social bond did not significantly predict the proportion of mounts across dyads in nonaggressive contexts, providing no support for the relationship reinforcement hypothesis. Finally, in support of the conflict resolution hypothesis, when individuals engaged in postconflict mounts, 1) the probability of being involved in further aggression decreased from 0.825 to 0.517, while 2) the probability of being involved in grooming interactions with each other increased from 0.119 to 0.606. The strength of the social bond between former opponents had no significant effect on grooming occurrence and agonistic behavior after postconflict mounts. Overall, our findings suggest that nonreproductive mounts in Barbary macaques have different functions that are not affected by the strength of the social bond.

Forage of six pure species was harvested for hay at several maturity stages during four years. The digestible energy content of 102 different lots of hay was determined by feeding to four groups of sheep during the same period, and by in vitro digestions and energy analysis of the undigested residues. The relationship between digestible energy content assayed by the two methods was highly significant (r = 0.85) and did not differ between years and species. Exclusion from regression of the hays containing less than 2 or more than 3 digestible kcal/g revealed that the in vitro assay could reproduce the in vivo digestible energy value with a standard deviation of 0.31 in over 70% of the hays. This represented the maturity and quality range of forage commonly fed to cattle and sheep. The in vitro assay therefore appeared promising for commercial quality determinations.

Throughout the primate order, individuals are highly motivated to handle infants that are not their own. Given the differing and often conflicting interests of the various participants in handling interactions (handler, infant, and mother), most functional hypotheses are specific to particular handling roles. Here we explore one hypothesis that may apply to all participants, but that has received relatively little attention: that handling may facilitate the formation and maintenance of social bonds. Using free-ranging rhesus macaques (Macaca mulatta) on Cayo Santiago, we examine the relationship between infant handling in the early weeks and the strength and diversity of infant social bonds months later, when infant relationships were more independent from those of their mothers. Our results largely confirm the influence of several social characteristics (kinship, rank, sex, and age) in governing handling interactions. They also provide the first evidence that early handling is associated with later social bonds that are stronger than expected based on these social characteristics. However, the enhancement of bonds is largely confined to related handlers; frequent unrelated handlers did not generally go on to form strong bonds with infants. This suggests that kinship may be a sort of prerequisite to the enhancement of social bonds via handling. Given the adaptive benefits of strong social bonds among adult primates, future research should investigate whether early infant handling may have longer term fitness effects.
