Measurement of pathogens on seed tubers is essential for informing likelihood of subsequent potato disease. Here we utilized quantitative PCR assessment of pathogen DNA and visual assessment of disease to measure seed tuber inoculum and used this to model development of disease in potato grown in pathogen-free soil. Analysis by recursive partitioning and modeling using receiver operating curves indicated both abundance of Rhizoctonia solani AG3 and Streptomyces scabies DNA, and disease symptoms associated with these pathogens on seed tubers could predict subsequent disease in progeny tubers and for R. solani, stolons. In contrast, abundance of Spongospora subterranea DNA and disease symptoms on seed tubers were not consistently associated with powdery scab in progeny tubers. The relationship between S. subterranea DNA and seed tuber symptoms on root galling was stronger. Symptomless seed tubers that carried high levels of S. subterranea DNA were also associated with greater root galling than those with low pathogen DNA levels. There was a modest association between root galling and powdery scab in progeny tubers. These results highlight the importance of using certified seed tubers, and demonstrate a statistical tool for measuring the impact of seed tuber-borne inoculum.

Shoots of the early-maturing potato (Solatium tuberosum L.) cultivar Jemseg often do not emerge uniformly when cut seed pieces are planted in cold soil. Under controlled temperatures of 10, 15 and 20 °C, shoots from whole tubers and basal portions of Jemseg tubers emerged later than those from apical portions of seed tubers. The delayed emergence of the basal portion was most pronounced at low temperatures and ranged from 5 to 17 d. Emergence from basal vs. apical portions of Conestoga was delayed by 10 d at 10 °C and by 4 d at 15 °C, only when seed tubers with a single dominant apical sprout were used. There was no delay at 20 °C. The variable time to emergence of shoots in the field can be explained by delayed emergence from basal buds on cut seed pieces.Key words: Potato, cultivars, emergence, bud position, temperature, seed tuber

SummaryThirty-two experiments examining the effects of the weight and within-row spacing of potato seed tubers on graded tuber yields of five varieties were conducted on eight sites from 1980 to 1985. A complex analysis technique was used to combine these data and estimate the optimum tuber planting densities for different ratios of seed cost to small (40–60 mm) and large (60–80 mm) ware value. The same technique could be applied to any other combination of seed cost, ware size and ware value.The optimum tuber planting density decreased with increasing seed-tuber weight. Differences in optimum planting density between varieties were much greater with small (35 g) than with large (105 g) seed tubers and decreased as the cost of seed increased relative to the value of ware. As large ware became worth more than small ware the influence of increasing seed cost on the optimum density was reduced. As the value of large ware increased, net returns increased and the effect of seed cost on net returns was reduced. Mean tuber size decreased with increasing stem density at harvest and at the same stem density was lower in varieties producing more daughter tubers/stem. Changes of mean tuber size (μ) and the spread of yield across size grades (σ) with time were well described by parallel curves in different varieties. It is suggested that in future it may not be necessary to determine optimum tuber planting densities by complex experiments involving several seed-tuber weights and spacings. Instead μ and σ could be estimated from simple experiments and tuber spacings determined by comparison with control varieties.

The period of susceptibility of potato tubers to powdery scab (Spongospora subterranea) was studied by inoculating potato plants with spores, or by watering plants in infested soil, at different stages of plant development in greenhouse conditions. Maximum susceptibility began about 1 week before the stage when 50% of stolons had swollen to at least 5-mm diameter (tuber set), and ended 3-4 weeks later. With holding irrigation water during this period reduced the severity of powdery scab by 65-75% in field experiments in 1981-82 and 1982-83, but had no apparent effect on disease severity in 3 out of 6 large-scale field trials during 1984-85.

Low yield is crucial bottleneck for potato production in Bangladesh. The application of vermicompost may enhance the yield of potato. The experiment was consisted of two factors, i.e., Factor A: - Vermicompost level (Vm1-4): Vm1: 0 t ha-1, Vm2: 3 t ha-1, Vm3: 6 t ha-1 and Vm4: 9 t ha-1; Factor B:- Tuber size (T1-5): T1: 5-10 g, T2: 10-20 g, T3: 20-30 g, T4: 30-40 g and T5: > 40 g. The experiment was conducted in a split-plot design with three replications. Vermicompost had significant effect on most of the yield contributing parameters investigated under present study. Results revealed that yield parameters increased with increasing vermicompost level irrespective of tuber size. Among the twenty (20) treatment combinations, vermicompost at the rate of 9 t ha-1 with tuber size > 40 g produced the maximum yield (31.33 t ha-1) that was 53.53 % higher than that of control with 3273.01 $ of monetary advantage. Therefore, present study suggests that potato growers may use vermicompost for increasing yield of potato in Bangladesh. SAARC J. Agric., 19(1): 71-79 (2021)

Understanding the contribution of seed tuber- and soilborne inocula of Rhizoctonia solani AG 3-PT in causing potato disease epidemics is an important step in implementing effective management strategies for the pathogen. A 2-year study was conducted to evaluate the contribution of each source of inoculum using an integrative experimental approach combining field trials and molecular techniques. Two distinct sets of genetically marked isolates were used as seed tuberborne and soilborne inocula in a mark-release-recapture experiment. Disease assessments were done during tuber initiation and at tuber harvest. Both inoculum sources were found to be equally important in causing black scurf disease, whereas soilborne inocula appeared to be more important for root and stolon infection, and seedborne inocula contributed more to stem canker. However, seed tuber-transmitted genotypes accounted for 60% of the total recovered isolates when genotyped using three polymerase chain reaction restriction fragment length polymorphism markers. The changes in population structure of the experimental R. solani population over the course of the growing season and across two growing seasons were investigated using eight microsatellite markers. The populations at different sampling times were somewhat genetically differentiated, as indicated by Nei’s gene diversity (0.24 to 0.27) and the fixation index (FST). The proportion of isolates with genotypes that differed from the inoculants ranged from 13 to 16% in 2013 and 2014, respectively, suggesting the possibility of emergence of new genotypes in the field. Because both soilborne and tuberborne inocula are critical, it is important to ensure the use of pathogen-free seed tubers to eliminate seed tuberborne inoculum and the introduction of new genotypes of R. solani for sustainable potato production in South Africa.
