To address concerns regarding the potential impact of antibiotic use in animal husbandry on antibiotic resistance in humans, we conducted a greenhouse-based study examining uptake of the veterinary antibiotics oxytetracycline (OTC) and monensin (MON) by Tifton 85 Bermudagrass (T85), the most commonly grown forage grass in the southeastern U.S.A. Since oxytetracycline is used in both veterinary and human medicine, its accumulation in animal products could impact human resistance to this antibiotic. Monensin is not used in human medicine but has a high potential for accumulating in the environment. Our research examined antibiotic uptake by forage grass T85, the effect of dairy manure application on its uptake, and antibiotic retention in soil. We compared unspiked, wet dairy manure to wet dairy manure spiked with MON or OTC that was soil surface applied to pots or incorporated into soil. After 6 wk, plant stem/leaf and root tissue, as well as soil samples, were assessed for antibiotic residues using enzyme-linked immunosorbent assay (ELISA). Results confirmed Tifton 85 MON and OTC uptake. Six weeks after adding the antibiotics, the greatest plant matter OTC and MON contents were 157.9 ± 70.6 and 234.4 ± 19.6 µg kg−1, respectively, and 17.6 and 369.5 µg kg−1, respectively, for soil. When spiked with OTC, manure incorporation led to decreased OTC uptake by T85 tissue. Bioaccumulation of these antimicrobials in livestock and in the environment is a potential concern for animal, environmental, and human health.

Aeromonas hydrophila, a Gram-negative bacterium commonly found in aquatic environments, is pathogenic to amphibians, reptiles, and mammals. In human medicine, the clinical symptoms of aeromonad infection include not only gastroenteritis but also extraintestinal infections, such as wounds, cellulitis, and septicemia, in immunocompromised and immunocompetent individuals. In this study, ten red-eyed crocodile skinks (Tribolonotus gracilis) that shared the same space were found dead 7 days after being shipped from Indonesia. The necropsy revealed A. hydrophila to be the causative agent, and the isolates were susceptible to most antibiotics, based on an antimicrobial susceptibility test. Seven virulence factors (act, ast, alt, aerA, fla, gcaT, and ahyB) considered to be associated with virulence were detected by PCR. Microscopic examination revealed several necrotic lesions and melano-macrophage centers in the tissue slides. Reptiles caught in the wild for trade experience captivity stress. Furthermore, in the winter, reptiles are easily exposed to the cold atmosphere. These stresses can negatively impact the immunity of these ectotherms, making them vulnerable to A. hydrophila infections. Therefore, to avoid such opportunistic infections and mortality following exposure to severe stress, medical care is recommended. The studies of alternatives, such as bacteriophage and bacteriocin, are needed for a preventive application.

Bacterial isolates from poultry products were tested for their susceptibility to 10 antibiotics commonly used in the therapeutic treatment of poultry. Bacteria were isolated from fresh whole broiler carcasses or from cut-up meat samples (breast with or without skin, wings, and thighs) that were either fresh or stored at 4 or 13°C (temperatures relevant to poultry-processing facilities). The Biolog system was used to identify isolates, and a broth dilution method was used to determine the antibiotic resistance properties of both these isolates and complementary cultures from the American Type Culture Collection. The antibiotics to which the most resistance was noted were penicillin G, sulfadimethoxine, and erythromycin; the antibiotic to which the least resistance was noted was enrofloxacin. Individual isolates exhibited resistances to as many as six antibiotics, with the most common resistance pattern involving the resistance of gram-negative bacteria to penicillin G, sulfadimethoxine, and erythromycin. Differences in resistance patterns were noted among 18 gram-positive and 7 gram-negative bacteria, and comparisons were made between species within the same genus. The data obtained in this study provide a useful reference for the species and resistance properties of bacteria found on various raw poultry products, either fresh or stored at temperatures and for times relevant to commercial processing, storage, and distribution. The results of this study show that resistance to antibiotics used for the therapeutic treatment of poultry occurs in bacteria in the processing environment.

Acinetobacter sp. HO-1-N, a gram-negative hydrocarbon oxidizing bacterium previously designated Micrococcus cerificans, has been shown to sequester the hydrocarbon into intracytoplasmic pools as a result of growth on this substrate. In hydrocarbon grown cells, an intracytoplasmic membrane system was also observed along with a doubling of cellular phospholipids (Z). However, using conventional dehydration and embedding procedures in preparing thin sectioned material, the hydrocarbon is extracted from the cells. This may lead to structural distortion, consequently, the freeze-etch technique was applied to preserve the integrity of the cell.

Bacteria exhibit the ability to adhere to the apical surfaces of intestinal mucosal cells. These attachments either precede invasion of the intestinal wall by the bacteria with accompanying inflammation and degeneration of the mucosa or represent permanent anchoring sites where the bacteria never totally penetrate the mucosal cells.Endemic gram negative bacteria were found attached to the surface of mucosal cells lining the walls of crypts in the rat colon. The bacteria did not intrude deeper than 0.5 urn into the mucosal cells and no degenerative alterations were detectable in the mucosal lining.

Periplasm of bacteria is the space outside the permeability barrier of plasma membrane but enclosed by the cell wall. The contents of this special milieu exterior could be regulated by the plasma membrane from the internal, and by the cell wall from the external environment of the cell. Unlike the gram-negative organism, the presence of this space in gram-positive bacteria is still controversial because it cannot be clearly demonstrated. We have shown the importance of some periplasmic bodies in the secretion of penicillinase from Bacillus licheniformis.In negatively stained specimens prepared by a modified technique (Figs. 1 and 2), periplasmic space (PS) contained two kinds of structures: (i) fibrils (F, 100 Å) running perpendicular to the cell wall from the protoplast and (ii) an array of vesicles of various sizes (V), which seem to have evaginated from the protoplast.

Blood cultures are positive in approximately only 50 per cent of the patients with nongonococcal bacterial infectious arthritis and about 20 per cent of those with gonococcal arthritis. But the concept that gram-negative bacteria could be involved even in chronic arthritis is well-supported. Gram stains are more definitive in staphylococcal arthritis caused by gram-positive bacteria than in bacterial arthritis due to gram-negative bacteria. In the latter situation where gram-negative bacilli are the problem, Gram stains are helpful for 50% of the patients; they are only helpful for 25% of the patients, however, where gram-negative gonococci are the problem. In arthritis due to gram-positive Staphylococci. Gramstained smears are positive for 75% of the patients.

Perhaps the data about periplasmic enzymes are obtained through biochemical methods but lack of morphological description. We have proved the existence of periplasmic bodies by electron microscope and described their ultrastructures. We hope this report may draw the attention of biochemists and mrophologists to collaborate on researches in periplasmic enzymes or periplasmic bodies with each other.One or more independent bodies may be seen in the periplasmic space between outer and inner membranes of Gram-negative bacteria, which we called periplasmic bodies. The periplasmic bodies have been found in seven species of bacteria at least, including the Pseudomonas aeroginosa. Shigella flexneri, Echerichia coli. Yersinia pestis, Campylobacter jejuni, Proteus mirabilis, Clostridium tetani. Vibrio cholerae and Brucella canis.

Keratitis and conjunctivitis (infections of the cornea or conjunctiva) are ocular infections caused by various bacteria, fungi, viruses or parasites; bacteria, however, are usually prominent. Systemic conditions such as alcoholism, diabetes, debilitating disease, AIDS and immunosuppressive therapy can lead to increased susceptibility but trauma and contact lens use are very important factors. Gram-negative bacteria are most frequently cultured in these situations and Pseudomonas aeruginosa is most usually isolated from culture-positive ulcers of patients using contact lenses. Smears for staining can be obtained with a special swab or spatula and Gram staining frequently guides choice of a therapeutic rinse prior to the report of the culture results upon which specific antibiotic therapy is based. In some cases staining of the direct smear may be diagnostic in situations where the culture will not grow. In these cases different types of stains occasionally assist in guiding therapy.
