The purpose of this reseach was to determine the composition of the right planting media for the growth of rubber plant seeds. This research was conducted for 3 (three) months The State Agricultural Polytechnic at Samarinda. This reseach consists of three levels of media composition treatment namely 75% top soil media and 25% sand (m1), 50% top soil media and 50% sand (m2) and 25% top soil soil media and 75% sand (m3) ). The composition of the planting media of top soil 50% and 50% sand gives better results to the growth of plant height and number of leaves compared to the treatment of the composition of other planting media, namely the composition of the top soil media 75% and 25% sand and the treatment of top soil planting media 25% and 75% sand.

RingkasanHeterogenitas batang bawah pada sistem okulasi Hevea brasiliensis dapat menyebabkan interaksi batang bawah dengan batang atas dapat menimbulkan berbagai tingkat keragaman respons antar individu batang atas dari klon yang sama. Tujuan penelitian ini adalah untuk mengetahui pengaruh berbagai jenis batang bawah terhadap respons batang atas dari klon karet yang sama pada okulasi, berdasarkan perubahan pola pita protein, dan isoenzim esterase (EST), asam fospatase (AP), malat dehidrogenase (MDH), fosfogluko oksaloasetat (PGD), fosfo glukosa isomerase (PGI), peroksidase (PER), sikimik dehidrogenase (SKD), glutamat oksaloasetat (GOT) dan leusin aminopeptidase (LAP) dari daun atau lateks batang atas. Kombinasi diuji adalah klon (a) batang atas klon PB260 yang dikombinasikan dengan PR255, BPM1, LCB1320, PR300, AVROS2037, RRIM712 dan GT1, sebagai kontrol GT1/GT1. (b) sebagai batang atas adalah BPM1, BPM24, RRIC100 dan RRIC102 yang dikombinasikan dengan batang bawah BPM1, BPM24, RRIC100, RRIC101, RRIC102 dan RRIC110. Hasil yang diperoleh menunjukkan adanya perubahan pola pita protein lateks PB260 yang diokulasikan pada PR255, BPM1, LCB1320, PR300, AVROS2037, RRIM712 sebagai batang bawah. BPM1, BPM24, RRIC100, RRIC102 sebagai batang atas dengan BPM1, BPM24, RRIC100, RRIC101, RRIC102 dan RRIC110 menyebabkan terjadinya perubahan pola pita protein lateks dari klon yang sama. Terjadi induksi pembentukan protein baru dengan BM 24 kDa pada kombinasi okulasi PB260/ PR255, BPM1/RRIC100, BPM24/ RRIC100, BPM24/RRIC102 dan BPM24/ RRIC110. Hasil analisis isoenzim pada lateks dan daun batang atas menunjukkan bahwa isoenzim MDH, PGD, PGI, PER, SKD, GOT dan LAP menghasilkan pita yang monomorfik untuk seluruh kombinasi okulasi yang diuji. Polimorfisme EST ditemukan pada PR255/PB260, LCB1320/ PB260, GT1/PB260, BPM24/ RRIC100 dan BPM24/RRIC101. Sedang polimorfisme AP ditemukan pada PR255/ PB260, LCB1320/PB260, GT1/ PB260, BPM24/ RRIC101, BPM24/ RRIC102, dan RRIC100/RRIC110. Polimorfisme untuk MDH dan SKD juga ditemukan pada PR255/PB260, LCB1320/PB260 dan GT1/ PB260. Berdasarkan sidik gerombol dan UPGMA dari penggabungan data analisis SDS-PAGE protein dan isoenzim menunjukkan bahwa batang atas dari klon yang sama yang diokulasikan pada berbagai batang bawah yang berbeda, umumnya berada dalam satu sub kelompok yang sama. Namun tingkat kesamaan genetiknya beragam, hal ini menunjukkan adanya perbedaan pengaruh batang bawah terhadap batang atas yang sama. SummaryThe heterogenity of root stocks leads to stock-scion interactions resulting in considerable variation at different level among the population of a single clone. The aim of this research is to study the effect of several rootstock on scion of the rubber clone, based on the changes on budding patterns of leaf or latex protein and isozymes esterase (EST, acid phosphatase (AP), malat dehydrogenase (MDH), phosphogluco oxaloacetate (PGD), phospho glucose isomerase (PGI), peroxidase (PER), shikimic dehydrogenase (SKD), glutamate oxaloacetate (GOT) and leucyne aminopeptidase (LAP), bands pattern. The combinations stocks/scions tested were (a) scion PB260 clone combined with PR255, BPM1, LCB1320, PR300, AVROS2037, RRIM712 and GT1. Control combinations used were GT1/GT1 and (b) as a scion BPM1, BPM24, RRIC100 dan RRIC102 combined with BPM1, BPM24, RRIC100, RRIC101, RRIC102 and RRIC110. The results showed that latex protein bands pattern of PB 260 as a scion were changed in combination with PR255, BPM1, LCB1320, PR300, AVROS2037, RRIM712 as a rootstocks. BPM1, BPM24, RRIC100, RRIC101, RRIC102 and RRIC110 as a rootstocks caused the changes of the latex protein patterns of BPM1, BPM24, RRIC100, RRIC102 as a scion. The interaction among stock/scion showed by the changes on the levels proteins or the prsence of new proteins with MW 24-63 kDa especially in combination of BPM1/ RRIC100, BPM24/ RRIC100, BPM24/ RRIC102 and BPM24/ RRIC110. BPM1, BPM24, RRIC100, RRIC102 and PB260 in combined with all the stocks were tested. Analyses of leaf and latex isozymes showed that MDH, PGD, PGI, PER, SKD, GOT and LAP produced monomorphic isozyme bands. The polymorphism were found on EST of PR255/PB260, LCB1320/PB260, GT1/PB260, BPM24/RRIC100 and BPM24/ RRIC101. While the combination of PR255/ PB260, LCB1320/ PB260, GT1/ PB260, BPM24/RRIC101, BPM24/ RIC102, and RRIC100/RRIC110 produced AP polymorphism. Polymorphism for MDH and SKD were also found in PR255/ PB260, LCB1320/PB260 and GT1/PB260. Cluster and UPGMA analyses of protein and isozyme pattern showed combinations of a scion with several different rootstocks belong to the same sub group. However, genetic symilarities among individuals were varieties, it showed that the differences effect of rootstock to the scion from the same clone.

This study aimed to determine the growth eye stump of rubber after laying away in coconut fiber media. The design of this study was Completely Randomized Design with laying duration in coconut fiber medium (P) as a treatment which consisting of 7 treatment levels e.g. P0: 0 day; P1:5 days; P2:10 days, P3:15 days; P4:20 days, P5: 25 days, and P6: 30 days storage. The result of this research showed that the growth of eye stump rubber after laying away on the coconut fiber medium was significantly influence the percentage of several parameters e.g. :starting of stump sprout, sprout height during storage, plant height, stem diameter, leaf number and leaf area total for each treatment except for the parameter percentage of eye stump rubber which not active yet.  The freshness and the seedling viability of rubber plant originating from the eye stump rubber after laying away for 0-30 days in coconut fiber can be maintained. The highest growth was observed in the 20 to 30 days of laying away.Keywords: stump sleeping eye grafting, storage, coconut huskPenelitian ini bertujuan untuk mengetahui pertumbuhan stum okulasi mata tidur Karet (Hevea brasiliensis Muell Arg.) setelah mengalami  penyimpanan pada media sabut kelapa.  Rancangan penelitian yang digunakan adalah Rancangan Acak Lengkap (RAL) dengan perlakuan percobaan lama penyimpanan stum okulasi mata tidur dalam  media sabut kelapa (P) yang terdiri dari 7 taraf perlakuan yaitu penyimpanan:P0: 0 hari; P1:5 hari ; P2:10 hari, P3: 15 hari ; P4: 20 hari, P5: 25 hari, dan P6: penyimpanan 30 hari. Hasil penelitian menunjukkan bahwa pertumbuhan stum okulasi mata tidur karet (Hevea brasiliensis Muell Arg.) setelah mengalami penyimpanan pada media sabut kelapa berpengaruh nyata terhadap peubah persentase stum okulasi mulai bertunas, tinggi tunas selama penyimpanan, tinggi tanaman, diameter batang, jumlah daun dan luas daun total untuk setiap perlakuan kecuali pada peubah persentase stum okulasi mata tidur yang belum aktif tumbuh. Kesegaran dan daya tumbuh bibit tanaman karet yang berasal dari stum mata tidur setelah disimpan selama 0–30 hari dalam media sabut kelapa dapat dipertahankan. Pertumbuhan tertinggi untuk setiap peubah yang diamati terdapat pada lama penyimpanan 20 hingga 30 hari.Kata kunci : stump okulasi mata tidur, penyimpanan, sabut kelapa

Effort to increase rubber plant production should be done, especially rejuvenation for old rubber plant which in turn require high numbers of seedling within short time period. One of the method to provide rubber plant seedlings within short time period is through grafting technique which combine plant having strong rootas base stem and high production plant as upper stem (entres). The objective of this research was to study response of entres age and clone to growth of rubber seedling (Hevea brasiliensis Muell, Arg.) produced from green and brown grafting. This study was conducted from February 2019 to Februari 2020. This study used Factorial Randomized Block Design with entres age as the first factor consisting of 3, 6, 9, 12 and 15 months as well as clone as the second factor consisting of PB 260 and BPM 24. Data was analyzed by using analysis of variance and followed bywith Honestly Significant Different (HSD) test and regression analysis. The study results showed that the best treatment was entres having 9 months of age as indicated by grafting success, the speed of stum out, stumlength, stumdry weight, root length androot dry weight which had the highest values than that of other treatments. Clone of PB 260 was the best entres that was very compatible with base stem of GT1 clone than that of BPM 24 clone because it gave the highest yield in term of all observed parametes. The interaction between 9 month entres age and PB 260 clone was the best treatment

Rubber (Hevea brasiliensis Muell. Arg., Euphorbiaceae) is a tropical tree crop mainly grown for the industrial production of latex. The rubber sector in Cameroon is faced with the effects of global economic crisis and a drop in the yield of latex due to several pests and diseases. This study focused on leaf diseases caused by Fusarium oxysporum and Pestalotiopsis microspora. The fungi were isolated from the leaves of the rubber plant and cultured for characterization on two media (Potato Dextrose Agar (PDA) and Potato Carrot Dextrose Agar (PCDA)) at various temperature and pH levels. Using the poison food technique, an in vitro assay was done to assess the effect of two fungicides (Synthetic Fungicide 1 (SF1) and Synthetic Fungicide 2 (SF2)) at the respective doses of 0.006, 0.013 and 0.025 g.L-1, and 0.005, 0.01 and 0.02 g.L-1 on the growth of the two fungi. Results obtained show that PDA is most favourable for the growth of both fungi compared to PCDA. Both fungi attained optimal growth at pH 7 and at an incubation temperature of 25 oC. Neither F. oxysporum nor P. microspora grew on SF1-amended PDA and PCDA at all doses of the fungicide. SF2 significantly (P>0.05) inhibited the growth of P. microspora by 53.3% at the dose of 0.025 g.L-1 while F. oxysporum was less sensitive to the fungicide at all doses. In vitro pathogenicity test revealed similar disease symptoms as those observed in the field. This study shows that SF1 is the best fungicide against F. oxysporum and P. microspora as it gave 100% inhibition on the growth of both fungi.
