Introduction: Oxidative stress caused by an excess of reactive oxygen species (ROS) is the main reason behind many civilization diseases. The fruits and leaves of Prunus domestica L. contain antioxidants such as vitamins, polyphenols (including flavonoids) and organic acids.The aim of the study was to evaluate the antioxidant activity and total polyphenol content of extracts of leaves and fruits of the plum harvested in 2 ripening phases.Materials and methods: Antioxidative activity was measured by 2,2-diphenyl-1-picrylhydrazyl (DPPH) and 2,2’-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) – ABTS methods, and total polyphenol content by Folin–Ciocalteu technique. Extracts in 40%, 70% and 96% (v/v) of ethanol as well as 99.8% (v/v) methanol were used. Ultrasound-assisted extraction was applied for 15, 30 and 60 min.Results: The antioxidant potential of the extracts, measured by DPPH method, was highest for leaf methanolic extracts of plum harvested before ripening (3.83 ±0.01 mg trolox/g of raw material), while for the ABTS method, highest activity was found in the fruit peel extract in methanol (12.89 ±0.04 mg trolox/g raw material). The highest polyphenol levels (5.89 ±0.16, 6.06 ±0.09 and5.76 ±0.13 mg gallic acid/g raw material) were found in the extracts of leaves harvested at fruit ripening stage, prepared in 70% (30 and 60 min) ethanol and 40% ethanol (60 min), respectively.Conclusion: The alcohol based extracts from the leaves and fruit of Prunus domestica L. were characterized by high antioxidant potential, which suggests the possibility of their use as a valuable source of antioxidants. Ethanol seems to be the most appropriate extractant.Keywords: alcohol based extracts; antioxidant activity; Prunus domestica L.; ripening stage; total polyphenol content.

Background: The natural antioxidants have an important role in the prevention of many diseases. The aim of study is to investigate phytochemical components, antioxidant activity and acute oral toxicity of Pomegranate (Punica granatum L.) fruit peel (PFP) extract. Materials and methods: Phytochemicals of PFP were determined by qualitative chemical tests, thin layer chromatography, total polyphenol and flavonoid contents. The PFP extract was evaluated for antioxidant activity by DPPH assay and MDA assay. In vivo acute oral toxicity test was conducted using Karber-Behrens method to determine LD50. Results: Results illustrated that PFP mainly contains flavonoids, alkaloids, tannins, triterpenes, saponins, and coumarins. PFP extract exhibited the total polyphenol and flavonoid contents with 189.97 mg gallic acid equivalent/g dry weight and 9.42 mg quercetin equivalent/g dry weight, respectively. The DPPH free radical scavenging and anti-lipid peroxidation activities of PFP extract were expressed with IC50 value of 4.80 μg/mL and 0.38 μg/ mL, sequentially. Simultaneously, the Dmax (the maximum dose administered to mice that no toxicity was observed) of PFP extract was determined to be 21.28 g/kg, equivalent to 35.64 g dried herb. Conclusion: The PFP extract is relatively safe and revealed high antioxidant activity. Using natural products as antioxidant agents has been beneficial to replace synthetic products. Efforts have been made to profile the antioxidant capacities of natural resources, such as medicinal plants. The polyphenol content of Himalayan rhubarb, Rheum emodi wall, was measured and the antioxidant activity was determined using DPPH and ABTS+ assay, and the oxidative stress was assessed using SOD enzymatic assay. Five different solvent fractions, n-hexane, n-butanol, ethyl acetate, dichloromethane, and water, were used for screening the antioxidant capacity in effort to determine the optimum extraction solvent. The total phenolic contents for R. emodi fractions ranged from 27.76 to 209.21 mg of gallic acid equivalents (GAE)/g of dry weight. DPPH and ABTS+ assay results are presented into IC50 values, ranged from 21.52 to 2448.79 μg/mL and 90.25 to 1718.05 μg/mL, respectively. The ethyl acetate fraction had the highest antioxidant activity among other fractions. Also, n-butanol and water fractions showed significantly lower IC50 values than the positive control in DPPH radical scavenging activity. The IC50 values of SOD assay of fractions ranged from 2.31 to 64.78 μg/mL. A similar result was observed with ethyl acetate fraction showing the highest SOD radical scavenging activity. The study suggests that the ethyl acetate fraction of R. emodi possess the strongest antioxidant activity, thus the most efficient in extracting antioxidant contents. Moreover, a highly significant correlation was shown between total polyphenol content and antioxidant activity screening assays. The compounds related to the antioxidant activity of R. emodi were identified to myricitrin, myricetin 3-galloyl rhamnoside, and myricetin, which have not been reported in studies about R. emodi before.

  Objective: Indonesia is a country which has various natural resources including medicinal plants. Among the plants, red dragon fruit is interesting to be studied. In this study, antioxidant activity of red dragon fruit peel (Hylocereus polyrhizus (F.A.C. Weber) Britton and Rose) isolate was tested using 2,2-diphenyl-1-picrylhydrazyl method.Methods: Isolation of active compounds was performed through the application of vacuum liquid chromatography and preparative thin-layer chromatography (TLC). Antioxidant activity was tested using both TLC and spectrophotometry.Result: Results revealed that the values of IC50 of isolate 1 and 2 were 2.952, 14 μg/mL and 25.635,95 μg/mL, respectively.Conclusion: Isolate of dragon fruit has antioxidant activity. The compound which results in antioxidant activity is terpenoid and steroid.

This study revealed polyphenolic content, nutritive content, antioxidant activity, and phenolic profile of methanol and aqueous extracts ofPunica granatumpeel extract. For this, extracts were screened for possible antioxidant activities by free radical scavenging activity (DPPH), hydrogen peroxide scavenging activity and ferric-reducing antioxidant power (FRAP) assays. The total phenolics and flavonoid recovered by methanolic (MPE) and the water extract (AQPE) were ranged from 185 ± 12.45 to 298.00 ± 24.86 mg GAE (gallic acid equivalents)/gm and 23.05 ± 1.54 to 49.8 ± 2.14 quercetin (QE) mg/g, respectively. The EC50of herbal extracts ranged from 100 µg/ml (0.38 quercetin equivalents), for AQPE, 168 µg/ml (0.80 quercetin equivalents), for MPE. The phenolic profile in the methanolic extracts was investigated by chromatographic (HPLC) method. About 5 different flavonoids, phenolic acids, and their derivatives including quercetin (1), rutin (2), gallic acid (3), ellagic acid (4), and punicalagin as a major ellagitannin (5) have been identified. Among both extracts, methanolic extract was the most effective. This report may be the first to show nutritive content and correlation analysis to suggest that phenols and flavonoids might contribute the high antioxidant activity of this fruit peel and establish it as a valuable natural antioxidant source applicable in the health food industry.
