Abstract Tissue culture techniques can increase the number of garlic seedlings. The purpose of this research is to determine the effect of NAA and coconut water in increasing the number of garlic seeds. This research used a Completely Randomized Design of two factors. The treatment used is NAA with concentrations of 0 ppm, 0.5 ppm, 1 ppm, 1.5 ppm, and coconut water concentrations of 0%, 10%, 20%. The variables observed were shoot emergence time, root emergence time, number of shoots, number of roots, number of leaves, shoot height, root length, and number of plantlets. The results showed that the addition of coconut water 20% without the addition of NAA in 1 bulb can produce 3.33 planlets and the results of explant propagation in 1 bulb can produce the number of shoots as many as 15.33 shoots. Giving coconut water with concentrations of 10% and 20% can increase the number of leaves, shoot height, and some planlets. The concentration of NAA 0.5 ppm can accelerate the root emergence time on garlic explant.

Tribulus (Tribulus terrestrisL.) is a medicinal plant with considerable implementation such as aphrodisiac and anti‑inflammation drugs. This research was conducted to study the effectivity of coconut water and IAA (Indole‑3‑acetic acid) application on various concentration as the growth regulator for tribulus growth on thein vitroand its effect toward observation variables. The material used were cotyledon from tribulus embryo and MS (Murashige and Skoog) medium. The research method used a Completely Randomized Design with first factor of coconut water concentration of 0 mL L-1, 50 mL L-1, 100 mL L-1, and 150 mL L-1and IAA concentration of 0 ppm, 0.15 ppm, 0.20 ppm, and 0.25 ppm as the second factor. Observed variables were shoots emergence time, number of shoots, shoot height, number of leaves, roots emergence time, number of roots and regression between observation variables. The result showed that the combination of 150 mL L-1coconut water and 0.25 ppm IAA gave the highest shoot height, roots emergence time, and number of roots. Coconut water treatment on 150 mL L-1concentration alone gave the best result on shoots emergence time, number of shoots, and number of leaves, while IAA concentration of 0.25 ppm independently gave the highest number of leaves. Regression analysis result indicate that the number of roots has a positive correlation with shoots height, number of shoots and number of leaves.

Black orchids (Coelogyne pandurata Lindl.) are epiphytic and endemic in Borneo. Habitat damage and over exploitation by the society can cause black orchids to be threatened with extinction. For this reason, alternative ways to increase black orchids are needed through tissue culture techniques with the addition of ambon banana extract and Benzyl Amino Purine BAP. The aim of this study was to determine the effect of ambon banana extract and BAP on the multiplication of black orchids. This research was conducted for 3 months from January to March 2018 at Pontianak Aloe Vera Center (AVC) Tissue Culture Laboratory. This study uses a Completely Randomized Design (CRD) with two factors. The first factor was ambon banana extract (0%; 2,5%; 5%; 7,5% and 10%) and the second factors BAP (0 M; 10-7M; 10-6M; 5x10-6M) with 3 replications, each of which obtained 60 trial unit. The results showed that the ambon banana extract had a significant effect on the time of budding, number of buds and number of leaves. The concentration of ambon banana extract 2,5% resulted in the fastest budding time appearing on the 11,33th day, the number of buds was 8,33 tillers, and the number of leaves was 17,67 strands.

Aims: This study was conducted to enhance germplasm conservation of some indigenous plantain landraces through tissue culture techniques in Cameroon. Study Design: The experiment was laid out in a completely randomized design (CRD) with three treatments in four replications. Place and Duration of Study: The study was conducted in the tissue culture laboratory of the Institute of Agricultural Research for Development (IRAD), Bambui, Cameroon, in the first half of 2018. Methodology: Explants were gotten from three plantain land races (Kwah, Ngumbe and Sangmoh) gotten from Bambui. Shoot tips were excised and cultured on Murashige and Skoog (MS) medium supplemented with 30 g of sucrose, 5 ml of ascorbic acid, 4 ml of 6- benzylaminopurine (BAP 1 mg/l), 1 ml indole-3- acetic acid (IAA 1 mg/l) and 6 g of agar at pH of 5.8±0.1 for shoot initiation and proliferation. Data was collected over a period of 12 weeks every 4th week on the average number of buds, shoot length, number of leaves, number contaminated and number dead. Results: All the landraces responded positively to shoot tip culture, since they could adapt and regenerate by producing buds with Ngumbe giving the highest mean number (7.0) of buds and Sangmoh giving the lowest (3.0). Analysis of variance revealed significant differences (p = 0.05) in most of the parameters measured except for number contaminated. High dead rate was recorded in Kwah with dead mean of 3.0. Ngumbe was found to regenerate better than Kwah and Sangmoh. Conclusion: It is recommended that the three plantain landraces, germplasm be conserved. The ramification of this finding vis-à-vis germplasm conservation and increase productivity is explained.

Acclimatization is the final stage of plant propagation in tissue culture techniques that can determine the success of the nursery process. This study aimed to observe the growth response during the acclimatization stage of the acid-adapted of Leucaena leucocephala Tarramba variety, which developed from tissue culture techniques. The design used in this study was a completely randomized design (CRD) with the cultivation of 11 mutant lines from tissue culture, namely M1-M11 (an acid-adapted mutant from tissue culture addition of 1 ppm IBA) and 2 controls (lamtoro broodstock without gamma irradiation) namely K0 (lamtoro broodstock resulting from tissue culture addition of 0 ppm IBA), K1 (breeding lamtoro from tissue culture addition of 1 ppm IBA). The variables observed were the level of plant viability, plant height, and number of leaves. The results showed that the acclimatization of the plant Leucaena leucocephala to tissue culture production on the M3 and M9 mutant lines gave the best response to plant morphological growth up to 5 WAP (weeks after planting). Background: Pineapple fruit used in this study was obtained from Sipahutar sub-district, North Tapanuli regency, North Sumatra which was a superior commodity fruit. Propagation of the Sipahutar pineapple plant was conducted vegetatively using bud stems and crown shoots, but the amount was limited to be planted in large areas. Therefore, propagation through tissue culture techniques could be used as an alternative solution.Objective: Explants source were taken from plantlets in vitro aged 3 months were grown on Murashige and Skoog (MS) basic medium with the addition of Indole Acetic Acid (IAA) 1 ppm.Methods: Sipahutar pineapple tissue culture work has been done in YAHDI Plant Tissue Culture Laboratory, Medan. The study design used a factorial completely randomized design. The first factor is coconut water with concentration of 0%, 10%, 20%, and 30%, second factor is Benzyl Amino Purine (BAP) with concentration 0 ppm, 2 ppm and 4 ppm, so there were 12 combination with 3 repetition.Result: The results showed that the addition of coconut water on MS base medium with IAA of 1 ppm gave a significant effect on the number of leaf yield and shoot height. The provision of BAP on basic MS medium with IAA of 1 ppm gave a significant effect on the number of shoots produced, the number of leaves produced, the length of the leaves, the width ofleaves and the length of shoots. The interaction of coconut water and BAP addition on MS base medium with IAA of 1 ppm gave significant effect on leaf length, leaf width and shoot length as well.Conclusions: MS + IAA basic media combined with coconut water + BAP could stimulate the growth of Sipahutar pineapple in vitro.

Explants play an important role in thepropagation system of stevia (Stevia rebaudiana Bertoni). A Completely Randomized Design (CRD) was used in this experiment with factorial pattern consisting of two factors, namely three types of explants (shoot tip, first node, and second node) and eleven accessions of stevia namely a1 (Bogor), a2 (Garut), a3 (Canada), a4 (Tawangmangu), a5 (STG1), a6 (SBG 4), a7 (SBG 10), a8 (SGB 2), a9 (BR5), a10 (SGR 7.5), a11 (TR 3.5). The results showed that shoot tip explant was the best explant than first node and second node for the number of shoots (18.11 shoots), number of leaves (93.49 leaves) and wet weight (3.56 grams). The best accession of shoot height was a10(SGR 7.5) (19.95 cm), the highest number of shoots wasa7(SBG 10) (21.87 shoots), the highest number of leaves wasa7(SBG 10) (138.00 leaves), the heaviest wet weight wasa7(SBG 10) (3.56 grams), the highest leaf chlorophyll content was a10 (SGR 7.5) (0.63 µg mL-1). Accessions with the fastest root initiation time at the rooting stage was a11 (TR 3.5) (4.00 DAC), the highest number of roots wasa10 (TR 3.5) (27.11 roots), the best root length wasa2(Garut) (4.51 cm). Information on the best explant types and stevia accessions in the in-vitro multiplication stage can be used as the basis for stevia breeding programs in Indonesia.AbstractEksplan berperan penting dalam sistem perbanyakan Stevia (Stevia rebaudiana Bertoni). Penelitian ini menggunakan Rancangan Acak Lengkap (RAL) dengan pola faktorial yang terdiri dari dua faktor yaitu tiga jenis eksplan (ujung pucuk, ruas pertama, dan ruas kedua) dan sebelas aksesi stevia yaitu a1 (Bogor), a2 (Garut), a3 (Canada), a4 (Tawangmangu), a5 (STG1), a6 (SBG 4), a7 (SBG 10), a8 (SGB 2), a9 (BR5), a10 (SGR 7.5), a11 (TR 3.5). Hasil penelitian menunjukkan bahwa eksplan ujung pucuk merupakan eksplan terbaik dibandingkan buku pertama dan kedua untuk jumlah tunas (18,11 tunas), jumlah daun (93,49 daun) dan berat basah (3,56 gram). Aksesi terbaik pada tinggi pucuk adalah a10(SGR 7.5) (19,95 cm), jumlah pucuk tertinggi a7 (SBG 10) (21,87 pucuk), jumlah daun terbanyak a7 (SBG 10) (138.00 helai daun), bobot basah terberat a7 (SBG 10) (3,56 gram), kandungan klorofil daun tertinggi adalah GR 7,5 (0,63 µg mL-1). Aksesi dengan waktu inisiasi akar tercepat pada tahap perakaran adalah TR 3,5 (4,00 HST), jumlah akar terbanyak adalah TR 3,5 (27,11 akar), panjang akar terbaik adalah a2(Garut) (4,51 cm). Informasi jenis eksplan dan aksesi stevia terbaik pada tahap multiplikasi in-vitro dapat dijadikan dasar program pemuliaan stevia di Indonesia.

This study aims to know the effect of Benzyl Amino Purine (BAP) and Coconut Water on shoot bud growth and Broccoli Sulforaphane content (Brassica oleracea L. var italic Plenck). The study was conducted in the laboratory of Biotechnology Department of Aquaculture, Faculty of Agriculture of Sam Ratulangi University, Manado, that conducted from August-December 2017. This study used a Complete Randomized Design (RAL), consisting of 8 treatments and each repeated as many 4 times, so we get 32 unit experiment. The variables observed were number of buds, number of leaves, plant height, wet weight, root number and Sulforaphane content analysis. The result of research shows that analysis of variance showed that in the use of Benzyl Amino Purine (BAP) concentration 3 ppm tends to increase the number of leaves aged 4 Weeks After Culture (MSK) and increase the number of shoots age 2 and 6 Weeks After Culture (MSK). Benzyl Amino Purine (BAP) 3 ppm can increase the wet weight of age 6W eeks After Culture ((MSK). Coconut water 20% tends to increase the number of leaves at age 6 Weeks After Culture (MSK) and increase the number of shoots aged 6 Weeks After Culture (MSK), while for combination of 3 ppm Benzyl Amino Purine (BAP) and coconut water 20% tends to increase the number of leaves aged 2 Weeks After Culture (MSK) and the number of shoots aged 2 Weeks After Culture (MSK). Combination of coconut water and Benzyl Amino Purine (BAP) is not detected by the content of Sulforaphane.

Salah satu alternatif untuk perbanyakan anggrek hitam (Coelogyne pandurata Lindl.) adalah melalui multiplikasi tunas anggrek secara in vitro. Tujuan penelitian adalah untuk membuktikan pengaruh pemberian NAA (Naphthalene Acetic Acid) dan ekstrak biji jagung (Zea mays) terhadap multiplikasi tunas anggrek hitam. Penelitian dilakukan di Laboratorium Kultur Jaringan Jurusan Biologi Fakultas Matematika dan Ilmu Pengetahuan Alam Universitas Tanjungpura Pontianak. Penelitian ini menggunakan Rancangan Acak Lengkap (RAL) pola faktorial dengan dua faktor perlakuan. Faktor pertama adalah NAA terdiri dari 5 taraf konsentrasi yaitu A0 (0 M/ kontrol) A1 (10-7 M), A2 (10-6 M), A3 (5x10-7 M) dan A4 (5x10-6 M ) dan faktor ekstrak biji jagung (B) dengan 5 taraf konsentrasi yaitu B0 (0%), B1 (2,5%), B2 (5%); B3 (7,5%) dan B4 (10%). Pemberian kombinasi NAA dan ekstrak biji jagung berpengaruh nyata terhadap semua parameter yaitu jumlah tunas, jumlah daun, dan tinggi tunas. Hasil terbaik rerata jumlah tunas pada perlakuan A4+B4 yaitu 5x10-6M NAA+10% ekstrak biji jagung. Hasil terbaik pada rerata jumlah daun pada perlakuan A2+B2 yaitu 5x10-7M NAA+5% ekstrak biji jagung dan hasil terbaik pada rerata tinggi tunas pada perlakuan A1+B1 yaitu 10-7M NAA+2,5% ekstrak biji jagung.Kata Kunci: multiplikasi; tunas anggrek hitam; ekstrak biji jagung; NAA. ABSTRACTGenerative reproduction of orchid plants it takes a requires the help of mycorriza mushrooms for seed germination, whereas conventional propagation business takes a long time to obtain large quantities of plants. One alternative to the propagation black orchids (Coelogyne pandurata Lindl.) is required through tissue culture techniques. The purpose of this study is to find the influence and concentration corn seed extract (Zea mays) and NAA (Naphthalene Acetic Acid) on the multiplication black orchids. This research was conducted in the tissue culture laboratory Biology Department Faculty of Mathematics and Natural Sciences Tanjungpura University Pontianak. The study used a Complete Randomized Design (RAL) of factorial patterns with two treatment factors. The first factor is that the NAA consists of 5 concentration levels  A0 (0 M) A1 (10-7 M), A2 (10-6 M), A3 (5x10-7 M) and A4 (5x10-6 M ) and the second factor is that corn seed extract of 5 levels concentratio B0(0%), B1 (2,5%), B2 (5%); B3 (7,5%) and B4 (10%). The administration NAA and corn seed extract in combination has a real effect on all parameters namely the number shoots, the number leaves, and the height shoots. The best results where the average number of shoots in the treatment of A2+B2 namely 5x10-6M NAA + 10% corn seed extract. The best results average number of leaves in the treatment  A2+B2 namely 5x10-7M NAA + 5% corn seed extract and in the best results for shoot height in the treatment of A1+B1 namely 10-7M NAA + 2.5% corn seed extract.Keywords: Multiplication; black orchid’s shoot; corn  seed extract; NAA

Vanilla has a potential to be developed through tissue culture techniques to anticipate the limitations of the parent plant as a source of planting material. The in vitro propagation ability of vanilla shoots needs to be controlled with the regulation of Kinetin and Benzyl Amino Purines. The interests of this study are 1) analysis of the response of vanilla explants at several Kinetin concentrations; 2) analysis of the response of vanilla explants at several concentrations of BAP and 3) analysis of the interaction of Kinetin and BAP on the response of vanilla explants to form shoot multiplication. The research was conducted at the Tissue Culture Laboratory Politeknik Negeri Jember from June to December 2020 using a factorial Completely Randomized Design (CRD). Factor 1 was the Kinetin concentration of 0.0, 1.0, 2.0 mg.L-1 and the second factor was the concentration of BAP 0.5, 1.5, 2.5 mg.L-1. The results proved that the fastest shoot multiplication occurred on MS medium + Kinetin 2 mg.L-1 with a mean of 8.7 days after inoculation. The mean number of shoots was 7.6 shoots/explant with the highest average wet weight of 0.9 grams/explant at the addition of BAP 1.5 mg. L-1 at measurement 70 days after inoculation.

Abstract Cashew (Anacardium occidentale L.) is a plant with high economic value. Conventional propagation of this plant still has obstacles, so an alternative techniques using tissue culture could be tried. One of the factors that determine the success of tissue culture techniques is the type and concentration of growth regulators was used. Growth regulator which have effect on shoot elongation is a cytokinin and gibberellin, this research tried to investigate the influence of combination cytokinin and gibberelin on in vitro shoot elongation of cashew. This research was conducted at BB-Biogen, Bogor on June-November 2010. The method in this research was to design experimental method with completely randomized design. The result was cytokinin could increase the length of shoots and quantity of shoots very well until 4 cm and quantity of shoot for 5. With the most effective cytokinin is zeatin of 5 mg/l.   Key words: Anacardium occidentale L., cytokinin, elongation of shoots, gibberelin
