Low Genic Variation between Black Ducks and Mallards

Abstract We used allozyme electrophoresis to estimate the degree of genetic differentiation among allopatric and sympatric populations of American Black Ducks (Anas rubripes) and Mallards (A. platyrhynchos). Mallards were collected in California, Saskatchewan, Manitoba, and Ontario, and Black Ducks were collected in Newfoundland, Nova Scotia, and Ontario. The mean genetic distances, D̄, between Black Duck populations (0.0007), between Mallard populations (0.0010), and between Mallard and Black Duck populations (0.0006) were virtually identical; there was as much genetic differentiation within the two species as there was between them. Such small genetic distances are characteristic of local populations of avian species in other orders, and are consistent with what is known about the lack of reproductive isolation between Black Ducks and Mallards. Although the two taxa are still somewhat split on an east-west basis, our genetic data do not support even subspecific status for the Black Duck.

The Thoroughbred (TB) horse is the best racehorse breed used in the racing industry. This breed has had a closed studbook for about 300 years. In Russia TB horses have been bred since the second half of the XVIII century. The modern Russian Stud Book register of TB horses is partially presented by stallions and broodmares imported from different countries. The genealogical structure of the breed is represented by 17 lines, among which the Northern Dancer line dominates (30.9 %). The study of features of different lines of TB was carried out on 17 loci of DNA microsatellites (VHL20, HTG4, AHT4, HMS7, HTG6, AHT5, HMS6, ASB23, ASB2, HTG10, HTG7, HMS3, HMS2, ASB17, LEX3, HMS1 and CA425) to assess genetic differentiation of the genealogical structure. The results of the DNA typing of 8091 Thoroughbred horses across microsatellite loci show that the gene pool of the domestic population is represented by 100 alleles typical of the breed. A comparative analysis of the genotypes of horses representing different lines indicates that they differ in the number of alleles (85–99), allele frequencies, the level of polymorphism Ae (2.93–3.48) and the degree of the observed heterozygosity Ho (0.653–0.739). The genetic distances between the lines varied in a wide range from 0.014 (Nasrullah – Northern Dancer) to 0.125 (Massine – Teddy). The correspondence to HWE was maintained in most lines, which is confirmed by the negative values of Fis. Cluster analysis demonstrated the correspondence of the obtained dendrogram of Nei’s genetic distances to its genealogical scheme of lines. Genetic differentiation of lines by index Fst varied in a range 0.005–0.073 at the mean value Fst = 0.024. The data indicate genetic differentiation of lines of TB at the STR markers and confirm the effectiveness of the system of linear breeding for the maintenance of interbreed biodiversity.

Abstract A new statistic for detecting genetic differentiation of subpopulations is described. The statistic can be calculated when genetic data are collected on individuals sampled from two or more localities. It is assumed that haplotypic data are obtained, either in the form of DNA sequences or data on many tightly linked markers. Using a symmetric island model, and assuming an infinite-sites model of mutation, it is found that the new statistic is as powerful or more powerful than previously proposed statistics for a wide range of parameter values.

Patterns of variability of genetic, morphometric, and physiological traits in Turkish chestnut (Castanea sativa Mill.) are investigated. Previous studies suggested a marked genetic differentiation of Turkish demes, arranged in an east-west cline. Genetic distances based on 21 isozyme loci, discriminant analysis of 13 fruit traits, and analysis of variance of carbon isotope discrimination were carried out. The results agree with genetic divergence reported earlier, and reveal remarkably consistent variation patterns for the three types of biological traits. Historical and anthropogenic effects are discussed as causes of chestnut evolution after the Wurm glaciation and of chestnut dispersal in historical times. Particular attention is focused on probable selective forces that molded the phenotypic variation of C. sativa and lead to the observed similarity of patterns of genetic and morphophysiological levels of variation.Key words: Castanea sativa, isozymes, morphometries, carbon isotype discrimination.

Surian Toona sinensis Roem is one of the most widely planted species in Indonesia. This study aimed to estimate the genetic diversity between a number of surian populations in a progeny test using RAPD markers, with the goal of proposing management strategies for a surian breeding program. Ninety-six individual trees from 8 populations of surian were chosen as samples for analysis. Eleven polymorphic primers (OP-B3, OP-B4, OP-B10, OP-H3, OP-Y6, OP-Y7, OP-Y8, OP-Y10, OP-Y11, OP-Y14, and OP-06) producing reproducible bands were analyzed for the 96 trees, with six trees per family sampled. Data were analyzed using GenAlEx 6.3, NTSYS 2.02. The observed percentage of polymorphic loci ranged from 18.2% to 50%. The mean level of genetic diversity among the surian populations was considered to be moderate (He 0.304). Cluster analysis grouped the genotypes into two main clusters, at similarity levels of 0.68 and 0.46. The first two axes of the PCoA explained 46.16% and 25.54% of the total variation, respectively. The grouping of samples into clusters and subclusters did not correspond with family and their distances, but the grouping was in line with the genetic distances of the samples.

Pseudechiniscus is a morphologically homogeneous heterotardigrade genus with a relatively low number of morphological features useful for the species discrimination. The species of the Pseudechiniscus suillus-facettalis complex are some of the most challenging tardigrades to identify. Here, we examine several populations from Antarctica, Italy, Madagascar and Norway that would have most likely been attributed to Pse. suillus prior to the recent redescription of the species. Populations were analysed using integrative taxonomy—a combination of classical morphology and morphometry, as well as genetic data. Besides minute differences in dorsal sculpture and morphometry, we found clear, species-specific differences in ventral sculpture which are very useful in discrimination of Pseudechiniscus species. Based on morphology (mainly ventral sculpture) and significant genetic distances in COI and ITS-2 sequences, we describe five new species: Pse. angelusalas sp. nov. from Madagascar, Pse. dastychi sp. nov. from Antarctica, Pse. ehrenbergi sp. nov. from Italy as well as Pse. indistinctus sp. nov. and Pse. lacyformis sp. nov. from Norway. Finally, we provide an updated phylogenetic tree of the genus Pseudechiniscus based on COI sequences.

Four samples of Neoplecostomus yapo were analyzed through the allozyme electrophoresis technique in corn starch gel. The allozyme pattern was similar to those found in N. paranensis with 24 loci scored. Two samples (ribeirão Atlântico and ribeirão Uraí) showed monomorphic bands for all 24 loci, whereas the other two (rio Verde and rio Fortaleza) showed 8.3% of polymorphic loci. The He genetic variability estimates for the rios Verde and Fortaleza populations were 0.0195 and 0.0179, respectively, too much inferior to the mean heterozygosity summed to species from the whole world (0.051). The Wright statistical values F IS = 0.5181, F IT = 0.5681 and F ST = 0.1039 and the genetic distance of Nei values showed that the four samples are genetically very similar to each other and that there is homozygote excess in the polymorphic loci.

This study presents the ampelographic and molecular characterization of 44 grapevine cultivars. Ampelographic data were obtained during two vegetation periods using the latest version of the descriptors. Based on the mean values transformed by the method indicated in IBPGR publications, a dendrogram was constructed. ISSR analysis was also employed to characterize the genotypes at the DNA level. Twenty primers, selected on the basis of their discriminating potential, generated a total of 157 bands, of which 140 were polymorphic. The dendrograms constructed by the two approaches were largely similar in both the clustering position and divergence of varietal groups. The least distance was observed between Yuvarlak Cekirdeksiz and Superior Seedless. The clustering position of cultivars throughout the dendrograms was basically related to the genetic distances and main uses, as well as to geographic origins.
