BackgroundIn adults, excess adiposity has been associated with low-grade, chronic inflammation and compromised bone health, but less is known about these linkages in children. The purpose of this study was to compare the circulating levels of inflammatory cytokines, adipokines, osteokines, and bone markers at rest and in response to plyometric exercise between obese and normal weight adolescent females.MethodsTen normal weight (BMI = 21.3 ± 2) and 10 obese (BMI = 32.9 ± 4), postmenarcheal females, aged 13–17 years, performed one bout of plyometric exercise (5 circuits; 120 jumps). Blood samples were taken at rest, 5 min, 1 h, and 24 h post-exercise. Tumor necrosis factor alpha (TNF-α), interleukin 6 (IL-6), insulin, leptin, osteocalcin, carboxy-terminal telopeptide (CTX), sclerostin, and parathyroid hormone (PTH) were measured in serum.ResultsCytokines were not different between groups at rest or over time with IL-6 increasing (+31%; p = 0.04) 5 min post-exercise and TNF-α decreasing (-9%; p = 0.001) 1 h post-exercise. Insulin and leptin were higher in the obese compared to the normal weight females. In both groups, insulin significantly increased 5 min post-exercise but remained elevated 1 h post-exercise only in the obese group. Leptin did not change in response to exercise. Osteocalcin was lower in the obese group across time and increased (+12%; p = 0.02) 24 h post-exercise in both groups. CTX was similar between groups at rest and decreased (-24%; p < 0.001) 1 h post-exercise. Sclerostin was similar between groups at rest, but there was a significant interaction reflecting a significant increase (+29%; p = 0.04) 5 min post-exercise in the obese group and a non-significant decrease (-13%; p = 0.08) in normal weight controls. PTH increased 5 min post-exercise, dropped 1 h post-exercise to lower than pre-exercise, and returned to baseline 24 h post-exercise in both groups.ConclusionObese adolescent females from our study had no evidence of resting inflammation or differences in bone resorption but show blunted bone formation when compared to normal weight controls. The direction and temporal changes in inflammatory cytokines, adipokines, and bone turnover markers to exercise were similar in both groups, reflecting an overall bone anabolic response for most biomarkers, except sclerostin, which increased only in the obese females immediately post-exercise, suggesting a different systemic regulation of sclerostin depending on adiposity.

The problem of overweight and obesity is one of the most urgent health issues in the world. 13% of girls and 21% of boys aged 11 suffer from overweight in the Russian Federation.The main causes of pubertal obesity are endocrine pathology, lifestyle and genetic disorders including mutation and polymorphisms of different metabolic pathways. Leptin produced in adipose tissue participates in reproduction regula-tion, glucose homeostasis, bone formation, etc. These effects are provided by leptin receptors coding LEPR gene. Q223R (rs1137101) polymorphism is associated with an increased serum level of leptin and overweight. There is no exact information about association between this polymorphism and obesity of adolescent females. The objective was to reveal LEPR Q223R polymorphism association between overweight and obesity in adolescent females.123 Caucasian adolescent females were involved in this study. All samples could be separated into two groups: the girls with normal weight (SDS BMI ±1.0; controlgroup), girls with overweight and obesity (SDS BMI≥ +1.0–2.0; studied group). Anthropometric measurements (weight, height, waist and hip circumference, body fat percentage) were taken, and genotyping was performed using polymerase chain reaction with electrophoresis detection.G-allele frequency was 43.1% in control and 40% in the clinical group. We found no significant differences of the prev-alence of polymorphism Q223Rbetween the studied groups (р=0,862). Furthermore, there was no association between the carriage of AG and GG with weight, BMI, body fat percentage, waist and hip circumference in both groups (р> 0.05).We have not found any association between LEPR Q223R and overweight and obesity in adolescent females.

The purpose of the present study was to determine if there exists an inverse relationship between perceived age and perceived physical attractiveness, i.e., are older faces evaluated as less attractive? Four groups of 15 subjects were studied: adult males and females (aged 31 to 38 yr., M = 34.53 yr.), adolescent males and females (aged 14 to 16 yr., M = 14.63 yr.). Subjects were given written instructions asking them to arrange two sets of photographs, male and female, according to physical attractiveness and then according to age. Perceived age and perceived physical attractiveness were negatively correlated ( rho = –.91, p < .01), i.e., as perceived age increased perceived attractiveness decreased. Differences in rankings by 30 male vs 30 female subjects and by 30 adolescent vs 30 adult subjects were not significant. However, there was greater agreement among the adolescent females than among members of the other groups for rankings of both males' attractiveness and females' age. All subjects showed greater agreement for what constitutes females' physical attractiveness than for what constitutes males' physical attractiveness.

The Spexin gene is considered the most dysregulated in obese human fat. Limited data suggest that the novel peptide spexin may potentially impact food intake, weight regulation and body adiposity. The aim of this case-control study was to compare fasting serum spexin concentrations between normal weight (NW) and overweight/obese (OB/OW) adolescent females and explore the relationship between circulating spexin and anthropometric, bone and fat mass, metabolic and hormonal parameters. Eighty post-menarcheal females (mean age ± SD 16.23 ± 2.26 years); 55 NW (mean BMI ± SD 19.72 ± 2.52 kg/m2) and 25 OB/OW (mean BMI ± SD 29.35 ± 3.89 kg/m2) participated in the study. Circulating spexin levels did not differ significantly (p = 0.378) between NW (median (interquartile range), 0.26 (0.17) ng/mL) and OB/OW (median (interquartile range), 0.28 (0.06) ng/mL) adolescents and did not correlate with BMI (rs = −0.090, p = 0.438), % body fat (rs = −0.173, p = 0.409), glucose or insulin resistance indices derived from fasting and oral glucose tolerance states. In the total study sample, spexin concentrations correlated positively with lipoprotein(a) (rs = 0.402, p = 0.046). In the OB/OW adolescents spexin levels correlated positively with testosterone (rs = 0.727, p = 0.011) and free androgen index (rs = 0.755, p = 0.007). In the NW adolescents, spexin concentrations correlated negatively with dehydroepiandrosterone sulphate (rs = −0.445, p = 0.038). Results may suggest potential involvement of spexin in the regulation of lipoprotein(a) and of the reproductive/adrenal axis in post-menarcheal adolescent females.
