Response of Longleaf Pine Seeds to Storage Conditions and Pregermination Treatments

Abstract Although longleaf pine (Pinus palustris Mill.) seeds are considered the most susceptible of the southern pines to damage during collection, processing, and storage, results of these studies show that high seed quality can be assured for periods up to 20 yr through proper handling and storing techniques. Recommendations for long-term storage include drying seeds to moisture contents of 10% or less and storing at subfreezing temperatures, preferably near 0°F. Reevaluation of stratification treatments applied under operational conditions indicates that the soaking in water that is necessary for seed imbibition reduces total germination in an amount proportional to the length of the soak. The organization of the sowing material stocks is a necessary condition for stable seed production of any crop. Insurance, reserve, breeding and transi-tional funds are such stocks in corn seed production. Their creation and maintenance largely depend on the economic longev-ity of seeds, which can be formed at the stages of cultivation, harvesting, post-harvest processing and storage of seed. However, the data on the peculiarities of the formation of seed longevity of corn hybrids and their ability to long-term storage are lim-ited. That is why research in this area is considered relevant and important. The results of study of regularities of formation and factors of longevity of corn hybrids seeds of domestic selection are summarized in the executed work. Ecological, technical-technological and varietal factors are formed dur-ing corn cultivation, har-vesting and processing. The greatest dependence is noted on the hydrothermal conditions, start-ing from the stage of cultivation, and then – in the stages after harvesting - the modes of drying and separation of seed. High economic durability is provided under the following conditions: hydro-term coefficient at the level of 0,5–0,7 – for vegetation and 0,3–0,4 – for maturing, dry-ing temperature within 36–400С at humidity of seeds of 20–30%, fractional separation, removal from the seeds mass of the unstable frac-tion containing 18–20% of the total. Some of the factors included a sign of seed size, which is clearly manifested at the stage of long-term storage of seed. Sowing fractions differ significantly in terms of size, including the weight of 1000 seeds and their linear size. The greatest economic durability is inherent in the first and second of all factions. It is recommended to obtain them by calibrating the seeds on separating sieves with round holes with a diameter of 8–10 mm, depending on the hybrids. Prolonged storage is proposed on the basis of a combination of two factors – low humidity and airtight packaging of seeds. This significantly reduces resource costs and extends the economic longevity of corn hybrid seeds to 4–5 years.

The objectives of this study were to understand how genotype, storage time, and storage conditions affect cooking time of beans and to indicate storage techniques that do not affect the cooking time. The grains were subjected to five different storage periods and six different storage conditions. The cooking time was estimated using the Mattson Cooker. The data were subjected to analysis of variance and a subsequent adjustment of simple linear regression for deployment of the interactions between the factors. Contrasts were used to determine the best levels of the factor storage condition. Genotype did not impact cooking time when the storage time and storage conditions were considered. Time and storage conditions affect the cooking time of beans in a dependent manner, but time of storage had the biggest influence. The best conditions for long-term storage of beans ensuring a smaller increase in cooking time is plastic storage at low temperatures. Thus, plastic freezer storage is a practical alternative for consumers.

Sample handling and storing are critical steps for the reliable measurement of circulating biomolecules in biological fluids. This study evaluates how cytokine measurements in pig seminal plasma (SP) vary depending on semen handling and SP storage. Thirteen cytokines (GM-CSF, IFNγ, IL-1α, IL-1β, IL-1ra, IL-2, IL-4, IL-6, IL-8, IL-10, IL-12, IL-18 and TNFα) were measured using Luminex xMAP® technology in individual seminal plasma (SP) samples (n = 62) from healthy breeding boars. Three separate experiments explored the delay (2 h and 24 h) in SP collection after ejaculation (Experiment 1) and SP storage, either short-term (5 °C, −20 °C and −80 °C for 72 h, Experiment 2) or long-term (at −20 °C and −80 °C for two months, Experiment 3), before analysis. Levels in fresh SP-samples were used as baseline control values. Delays in SP harvesting of up to 24 h did not substantially impact SP cytokine measurements. Some cytokines showed instability in stored SP samples, mainly in long-term storage. Ideally, cytokines in pig SP should be measured in fresh samples harvested within 24 h after ejaculation. If storage of SP is imperative, storage conditions should be adjusted for each cytokine.

The introduction and active use of information transmission and storage systems in the Ministry of Defense (MoD) of Ukraine form the need to develop ways of guaranteed removal of data from media after their use or long-term storage. Such a task is an essential component of the functioning of any information security system. The article analyzes the problems of guaranteed destruction of information on magnetic media. An overview of approaches to the guaranteed destruction of information on magnetic media of different types is presented, and partial estimates of the effectiveness of their application are given by some generally accepted indicators of performance evaluation. The article also describes the classification of methods of destruction of information depending on the influence on its medium. The results of the analysis revealed the main problems of application of software methods and methods of demagnetization of the information carrier. The issue of guaranteed destruction of information from modern SSD devices, which are actively used in the formation of new systems of information accumulation and processing, became particularly relevant in the article. In today's conditions of development of the Armed Forces of Ukraine, methods of mechanical and thermal destruction are more commonly used today. In the medium term, the vector of the use of information elimination methods will change towards the methods of physical impact by the pulsed magnetic field and the software methods that allow to store the information storage device, but this today requires specialists to develop new ways of protecting information in order to avoid its leakage.

Storage of tissue slides has been claimed to induce dramatically reduced antigen detection particularly for immunohistochemistry (IHC). With tissue microarrays, the necessity to serially cut blocks in order to obtain as much material as possible is obvious. The presumed adverse effect of storage might hamper such an approach. The authors designed an experimental setting consisting of four different storage conditions with storage time of tissue slides of up to 1 year. Detection of proteins, DNA, and mRNA was performed using IHC and in situ hybridization techniques. Slight but significant changes in IHC occurred over time. The most important factor is the primary antibody used: four showed no significant changes, whereas limited decreases in 8 antibodies could be detected by image analysis. Whether the antigen was nuclear or cytoplasmic/membranous did not matter. No major differences between different storage conditions could be shown, but storage at 4C was overall the best procedure. Furthermore, gene copy number aberrations, chromosomal translocations, and the presence of mRNA could be detected on slides stored up to 1 year. In conclusion, in tissues optimally formalin fixed and using modern histological techniques, only minute changes in tissue antigenicity are induced by long-term storage.

The fatty acid's contents of non-conform pooled human milk can be affected by different processing and storage operational conditions. Besides, the knowledge of changes in the human milk fatty acid profile can help indicate its use in a given storage period, according to each newborn's specific need. Thus, in the present work, changes in the fatty acid profiles of three types of human milk (raw; pasteurized at 62.5 °C for 30 min; homogenized at 40 oC for 30 s followed by pasteurization) were studied during storage for six months in a freezer at -18 oC. Large variations were observed in the concentrations of polyunsaturated fatty acids, particularly of docosahexaenoic acid, with a reduction of almost 50% of its total. Palmitic and stearic acid contents also changed according to the conditions of processing and storage. Correlations between the decrease of long fatty acid chains and the increase of medium and short chains were verified.  Thus, we observe that operational conditions of processing and storing change human milk lipid profile, with some nutritional losses.
