Herein, the extraction of bioactive compounds from umbu fruit peel was optimized using thermal-assisted solid–liquid extraction. In parallel, antioxidant, antimicrobial, and inhibitory effects against α-amylase of optimized extract were also evaluated. The combination of operational conditions including the temperature (32–74 °C), ethanol concentration (13–97%), and solid/liquid ratio (1:10–1:60; w/v) was employed using a rotational central composite design for optimization. The extracts were evaluated for total phenolic compounds (TPC), total flavonoid compounds (TFC) and antioxidant capacity by ABTS•+, DPPH• and FRAP assays. The bioactive profile of the optimized extract was obtained by ultra-performance liquid chromatography coupled to quadrupole/time-of-flight mass spectrometry in electrospray ionization in both negative and positive modes. The statistically evaluated results showed that the optimal operational conditions for the recovery of bioactive compounds from umbu fruit peel included 74 °C, 37% ethanol, and a solid–liquid ratio of 1:38. Under these conditions, the obtained values were 1985 mg GAE/100 g, 1364 mg RE/100 g, 122 µmol TE/g, 174 µmol/TE g and 468 µmol Fe2+/g for TPC, TFC, ABTS•+, DPPH•, and FRAP assays, respectively. In addition, the optimized extract was effective against Gram-positive and Gram-negative bacteria (MBC ranged from 0.060 to 0.24 mg GAE/mL), as well as it was effective to inhibit α-amylase (IC50 value of 0.076 mg GAE/mL). The optimized extract showed to be mainly constituted by phenolic acids and flavonoids.

AbstractLocomotive design is a highly complex task that requires the use of systems engineering that depends upon knowledge from a range of disciplines and is strongly oriented on how to design and manage complex systems that operate under a wide range of different train operational conditions on various types of tracks. Considering that field investigation programs for locomotive operational scenarios involve high costs and cause disruption of train operations on real railway networks and given recent developments in the rollingstock compliance standards in Australia and overseas that allow the assessment of some aspects of rail vehicle behaviour through computer simulations, a great number of multidisciplinary research studies have been performed and these can contribute to further improvement of a locomotive design technique by increasing the amount of computer-based studies. This paper was focused on the presentation of the all-important key components required for locomotive studies, starting from developing a realistic locomotive design model, its validation and further applications for train studies. The integration of all engineering disciplines is achieved by means of advanced simulation approaches that can incorporate existing AC and DC locomotive designs, hybrid locomotive designs, full locomotive traction system models, rail friction processes, the application of simplified and exact wheel-rail contact theories, wheel-rail wear and rolling contact fatigue, train dynamic behaviour and in-train forces, comprehensive track infrastructure details, and the use of co-simulation and parallel computing. The co-simulation and parallel computing approaches that have been implemented on Central Queensland University’s High-Performance Computing cluster for locomotive studies will be presented. The confidence in these approaches is based on specific validation procedures that include a locomotive model acceptance procedure and field test data. The problems and limitations presented in locomotive traction studies in the way they are conducted at the present time are summarised and discussed.

Welding is accompanied by the presence of weld residual stresses, which in case of dissimilar metal welds even with post weld heat treatment cannot be removed completely therefore they should be considered when assessing possible welding defects. The measurement of residual stress in metal weld is a very complex procedure and also in the investigated case could not be carried out as it is the part of a working plant. However, by modelling these processes, the residual stresses and deformation of the components caused by this manufacturing method can be determined. It is important to calculate these values as accurately as possible to determine the maximum load capacity of the structure. The structure under examination was the dissimilar metal weld of a VVER-440 steam generator. 2D simulations were performed, where temperature and phase-dependent material properties were implemented. Different loading scenarios were considered in the numerical analysis. The results can be useful to determine the real loading conditions of a given component and can be used to predict stress corrosion crack initiation locations, as well as to evaluate the lifetime and failure mode prediction of welded joints.

A typical 1100 MWe pressurized water reactor (PWR) is a second unit installed at the coastal site of Pakistan. In this paper, verification analysis of reactivity control worth by means of rod cluster control assemblies (RCCAs) for startup and operational conditions of this typical nuclear power plant (CNPP) has been performed. Neutronics analysis of fresh core is carried out at beginning of life (BOL) to determine the effect of grey and black control rod clusters on the core reactivity for startup and operating conditions. The combination of WIMS/D4 and CITATION computer codes equipped with JENDL-3.3 data library is used for the first time for core physics calculations of neutronic safety parameters. The differential and integral worth of control banks is derived from the computed results. The effect of control bank clusters on core radial power distribution is studied precisely. Radial power distribution in the core is evaluated for numerous configurations of control banks fully inserted and withdrawn. The accuracy of computed results is validated against the reference values of Nuclear Design Report (NDR) of 1100 MWe typical CNPP. It has been observed that WIMS-D4/CITATION shows its capability to effectively calculate the reactor physics parameters.

Abstract Background ICU operational conditions may contribute to cognitive overload and negatively impact on clinical decision making. We aimed to develop a quantitative model to investigate the association between the operational conditions and the quantity of medication orders as a measurable indicator of the multidisciplinary care team’s cognitive capacity. Methods The temporal data of patients at one medical ICU (MICU) of Mayo Clinic in Rochester, MN between February 2016 to March 2018 was used. This dataset includes a total of 4822 unique patients admitted to the MICU and a total of 6240 MICU admissions. Guided by the Systems Engineering Initiative for Patient Safety model, quantifiable measures attainable from electronic medical records were identified and a conceptual framework of distributed cognition in ICU was developed. Univariate piecewise Poisson regression models were built to investigate the relationship between system-level workload indicators, including patient census and patient characteristics (severity of illness, new admission, and mortality risk) and the quantity of medication orders, as the output of the care team’s decision making. Results Comparing the coefficients of different line segments obtained from the regression models using a generalized F-test, we identified that, when the ICU was more than 50% occupied (patient census > 18), the number of medication orders per patient per hour was significantly reduced (average = 0.74; standard deviation (SD) = 0.56 vs. average = 0.65; SD = 0.48; p < 0.001). The reduction was more pronounced (average = 0.81; SD = 0.59 vs. average = 0.63; SD = 0.47; p < 0.001), and the breakpoint shifted to a lower patient census (16 patients) when at a higher presence of severely-ill patients requiring invasive mechanical ventilation during their stay, which might be encountered in an ICU treating patients with COVID-19. Conclusions Our model suggests that ICU operational factors, such as admission rates and patient severity of illness may impact the critical care team’s cognitive function and result in changes in the production of medication orders. The results of this analysis heighten the importance of increasing situational awareness of the care team to detect and react to changing circumstances in the ICU that may contribute to cognitive overload.

Abstract The “muon-to-electron conversion” (Mu2e) experiment at Fermilab will search for the charged lepton flavour violating neutrino-less coherent conversion of a muon into an electron in the field of an aluminum nucleus. The observation of this process would be the unambiguous evidence of the existence of physics beyond the standard model. Mu2e detectors comprise a straw-tracker, an electromagnetic calorimeter and an external veto for cosmic rays. In particular, the calorimeter provides excellent electron identification, a fast calorimetric online trigger, and complementary information to aid pattern recognition and track reconstruction. The detector has been designed as a state-of-the-art crystal calorimeter and employs 1348 pure Cesium Iodide (CsI) crystals readout by UV-extended silicon photosensors and fast front-end and digitization electronics. A design consisting of two identical annular matrices (named “disks”) positioned at the relative distance of 70 cm downstream the aluminum target along the muon beamline satisfies the Mu2e physics requirements. The hostile Mu2e operational conditions, in terms of radiation levels (total expected ionizing dose of 12 krad and a neutron fluence of 5 × 1010 n/cm2 @ 1 MeVeq (Si)/y), magnetic field intensity (1 T) and vacuum level (10−4 Torr) have posed tight constraints on scintillating materials, sensors, electronics and on the design of the detector mechanical structures and material choice. The support structure of each 674 crystal matrix is composed of an aluminum hollow ring and parts made of open-cell vacuum-compatible carbon fiber. The photosensors and front-end electronics for the readout of each crystal are inserted in a machined copper holder and make a unique mechanical unit. The resulting 674 mechanical units are supported by a machined plate of vacuum-compatible plastic material. The plate also integrates the cooling system made of a network of copper lines flowing a low temperature radiation-hard fluid and placed in thermal contact with the copper holders to constitute a low resistance thermal bridge. The data acquisition electronics are hosted in aluminum custom crates positioned on the external lateral surface of the disks. The crates also integrate the electronics cooling system as lines running in parallel to the front-end system. In this paper we report on the calorimeter mechanical structure design, the mechanical and thermal simulations that have determined the design technological choices, and the status of component production, quality assurance tests and plans for assembly at Fermilab.
