Heteromorphic incompatibility system under disruptive selection

Forty populations of Pemphis acidula sampled from Africa to Taiwan have been analysed for heterostylic incompatibility. All populations except one are distylic. The exceptional population comes from southern Madagascar. It consists of 100% long homostyled plants that are self-compatible. A semi-homostyle which is self-incompatible in some and self-compatible in others is found in nine populations at 10% frequency. Measurements of anthers, styles and pollen grains reveal that all the distylic populations are similar. Both the long- and short-styled plants have two levels of anthers with two sizes of pollen grains. In a more extreme form this is a feature usually found only in tristylic species. If distylic Pemphis has been derived from tristyly by the loss of the ‘mid’ type, then the end point of selection would be equal anthers and pollen within each type. The widespread distribution of the forty populations and their similarity indicate that this final equilibrium is unattainable. The explanation of this paradox comes from the homostyle population. The two rows of anthers are at equal heights and the two classes of pollen grains are equal in size. Thus in a self-compatible, single type population, selection has gone to completion. In the distylic populations the selection within the two types, for equality of anthers and pollen grains, is in opposite directions in each type. The 'mid’ anthers have to be raised in the short-styled and lowered in the long-styled plants. Similarly the selection of ‘mid’ pollen is in opposite directions. This disruptive selection keeps the equilibrium at an intermediate point.

Program pemuliaan ikan mas varietas Punten dilakukan dengan seleksi individu terhadap karakter bobot ikan. Pembentukan populasi dasar untuk kegiatan seleksi dilakukan dengan memijahkan secara massal induk ikan mas yang terdiri atas 20 induk betina dan 21 induk jantan yang dikoleksi dari daerah Punten, Kepanjen (delapan betina dan enam jantan), Kediri (tujuh betina dan 12 jantan), Sragen (27 betina dan 10 jantan), dan Blitar (15 betina dan 11 jantan). Larva umur 10 hari dipelihara selama empat bulan. Selanjutnya dilakukan penjarangan sebesar 50% dan benih dipelihara selama 14 bulan untuk dilakukan seleksi dengan panduan hasil sampling 250 ekor individu setiap populasi. Seleksi terhadap calon induk dilakukan saat umur 18 bulan pada populasi jantan dan betina secara terpisah dengan memilih berdasarkan 10% bobot ikan yang terbaik. Calon induk yang terseleksi kemudian dipelihara hingga matang gonad, kemudian dipilih sebanyak 150 pasang dan dipijahkan secara massal. Didapatkan respons positif dari hasil seleksi berdasarkan bobot ikan, yaitu 49,89 g atau 3,66% (populasi ikan jantan) dan 168,47 g atau 11,43% (populasi ikan betina). Nilai heritabilitas untuk bobot ikan adalah 0,238 (jantan) dan 0,505 (betina).Punten carp breeding programs were carried out by individual selection for body weight trait. The base population for selection activities were conducted by mass breeding of parent consisted of 20 female and 21 male collected from area Punten, eight female and six male (Kepanjen), seven female and 12 male (Kediri), 27 female and 10 male (Sragen), 15 female and 11 male (Blitar). Larvae 10 days old reared for four moths. Then after spacing out 50% of total harvest, the offspring reared for 14 months for selection activity based on the sampling of 250 individual each population. Selection of broodstock candidates performed since 18 months age on male and female populations separately by selecting based on 10% of fish with best body weight. Candidates selected broodstocks were then maintained until mature. In oder to produce the next generation 150 pairs were sets and held for mass spawning. The results revealed that selection response were positive, 49.89 g (3.66%) for male and 168.47 (11.43%) for female. Heritability for body weight is 0.238 (male) and 0.505 (female).

The tryphine that coats the pollen grains of Raphanus is tapetally synthesized and is composed of a fibro-granular and a lipidic component. The fibro-granular material is proteinaceous and is secreted by cisternae of the endoplasmic reticulum. The lipidic component is derived, mainly, from degraded elaioplasts. The fibro-granular material is applied to the pollen exine first, followed by the lipidic mass. The tryphine condenses during the final stages of pollen maturation and dries down to form a thick, highly viscous coating. The major part of the condensation appears to result from dehydration. The tryphine, extracted from the pollen by a centrifugal method and mounted in a membrane, appears to be capable of penetrating the outer layers of a stigma of the same species and, if the pollen from which it was derived is incompatible with respect to the stigma, the stimulation of the production of the callosic reaction body in a manner similar to an incompatible pollen tube. It is proposed that, in Raphanus , substances responsible for the initiation of at least two stages in the self-incompatibility system are held in the tryphine.

The chief determining factor in spontaneous self-fertilization in white sweet clover (Melilotus alba) appears to be the distribution of pollen within the unopened flowers. This in turn depends on (1) length of stamens, (2) length of style, (3) amount of pollen, and (4) size of cavity within the upper part of the keel. When the flower structure and quantity of pollen is such as to insure the deposition of pollen grains on the stigma before the blossom is likely to be disturbed by insects, the plant will be normally self-fertilized. In the yellow flowered species of sweet clover (Melilotus officinalis) spontaneous self-fertilization is effectively prevented, except in certain varieties, by a characteristic of the stigma which makes it unreceptive. Scarification of the stigmatic surface appears to be necessary before fertilization can take place. Variation occurs also, as in M. alba, in the length of stamens and style, size of keel cavity, and the amount of pollen.There is a high correlation in M. alba between the percentage of flowers which are naturally self-pollinated and the percentage of flowers which produce pods when the plants are caged to exclude insects. Selection of plants which are normally self-fertilized can be made by examining the early flowers, thus obviating the necessity of bagging or caging plants which do not possess this character.A strain of M. alba which is normally self-fertilized was found to produce almost twice as much seed as another strain which is normally cross-pollinated under comparable conditions in the field.

Antibiotics have been widely used for a number of decades for human therapy and farming production. Since a high percentage of antibiotics are discharged from the human or animal body without degradation, this means that different habitats, from the human body to river water or soils, are polluted with antibiotics. In this situation, it is expected that the variable concentration of this type of microbial inhibitor present in different ecosystems may affect the structure and the productivity of the microbiota colonizing such habitats. This effect can occur at different levels, including changes in the overall structure of the population, selection of resistant organisms, or alterations in bacterial physiology. In this review, I discuss the available information on how the presence of antibiotics may alter the microbiota and the consequences of such alterations for human health and for the activity of microbiota from different habitats.

Nonomuraea gerenzanensis ATCC 39727 produces the glycopeptide antibiotic A40926, which is the natural precursor of the semi-synthetic, last-resort drug dalbavancin. To reduce the cost of dalbavancin production, it is mandatory to improve the productivity of the producing strain. Here, we report that the exposure of N. gerenzanensis wild-type population to sub-inhibitory concentrations of A40926 led to the isolation of differently resistant phenotypes to which a diverse A40926 productivity was associated. The most resistant population (G, grand colonies) represented at least the 20% of the colonies growing on 2 µg/mL of A40926. It showed a stable phenotype after sub-culturing and a homogeneous profile of self-resistance to A40926 in population analysis profile (PAP) experiments. The less resistant population (P, petit) was represented by slow-growing colonies to which a lower A40926 productivity was associated. At bioreactor scale, the G variant produced twice more than the wild-type (ca. 400 mg/L A40926 versus less than 200 mg/L, respectively), paving the way for a rational strain improvement based on the selection of increasingly self-resistant colonies.

Cultivated and wild almonds are self-incompatible and thus require outcrossing by insect pollinators to produce viable seed. In commercial production, considerable efforts are directed towards placement and selection of cultivars for cross-pollination. However, since honeybees do not distinguish between the different cultivars, stigmas are usually covered by a mixture of both compatible and incompatible pollen. Using in vitro assays, we demonstrated that pollen extracts promoted germination in self pollen with no inhibitory effects observed. Elemental analyses of pollen extracts showed that enhanced levels of Ca, Mg, K, Na, and P were eluted from the grains. From this, we raise the question of possible interaction between compatible and incompatible pollen, and speculate that incompatible pollen grains may support and enhance germination of adjacent compatible pollen.

This paper describes the cytochemistry and ultrastructure of the developing tapetum in Primula obconica, a plant with a heteromorphic, sporophytic self-incompatibility system. The tapetum is of the secretory type and cytochemical tests have shown that when it breaks down proteinaceous (esterase) and lipidic components are deposited on the developing pollen grains. Acid phosphatase, a marker of gametophytic enzyme activity, is confined to the cytoplasm and intine of the developing pollen. Ultrastructural studies show that prior to its dissolution the tapetum undergoes a number of changes. In the early stages of development the tapetum is rich in ribosomes and rough endoplasmic reticulum, but following the breakdown of the tapetal cell wall the main components of the cytoplasm are densely staining spherical bodies surrounded by ribosomes and orbicular bodies, which appear to be confined to the cell periphery. As the cells break down, rod-like fibrils can be seen amongst the degenerate organelles and within the bacular cavities of the pollen. On dehiscence the pollen has a lipidic coating in addition to the fibrillar material in the pollen wall and the remnants of the tapetum can be seen adhering to the fibrous layer of the anther wall. Thus the mature, binucleate pollen of P. obconica is demonstrated to carry wall materials of sporophytic origin.
