Whether one collects the perivisceral fluid of a sea-urehin or of a worm, or the blood of a crustacean or a mollusc, the same phenomenon is always more or less distinctly to be observed. A kind of coagulation takes place, the fluid separating sooner or later into two portions, which have considerable superficial resemblance to the clot and serum of vertebrate blood. It is easy to watch the formation of the clot by placing a drop of fresh-drawn fluid upon a cover-glass and inverting this above a glass cell, of which the edge is oiled to prevent evaporation. The drop thus hangs freely and the coagulation can go on without interference.

As a powerful modeling tool, Gaussian process (GP) employs a Bayesian statistics approach and adopts a highly nonlinear regression technique for general scientific and engineering tasks. In the first step of constructing Gaussian process model is to estimate the best value of the hyperparameter which turned to be used in the second step where a satisfactory nonlinear model was fitted. In this paper, a modified Wolfe line search approach for hyperparameters estimation by maximizing the marginal likelihood based on conjugate gradient method is proposed. And then we analyze parameter correlation according to the value of hyperparameters to control the warpage which is a main defect for a thin shell structure part in injection molding.

AbstractCell plasticity is a crucial hallmark leading to cancer metastasis. Upregulation of Rho/ROCK pathway drives actomyosin contractility, protrusive forces and contributes to the occurrence of highly invasive amoeboid cells in tumors. Cancer stem cells are similarly associated with metastasis, but how these populations arise in tumors is not fully understood. Here we show that the novel oncogene RASSF1C drives mesenchymal to amoeboid transition and stem cell attributes in breast cancer cells. Mechanistically, RASSF1C activates Rho/ROCK via SRC mediated RhoGDI inhibition, resulting in generation of actomyosin contractility. Moreover, we demonstrate that amoeboid cells display the cancer stem cell markers CD133, ALDH1 and the pluripotent marker Nanog; are accompanied by higher invasive potential in vitro and in vivo; and employ extracellular vesicles to transfer the invasive phenotype to target cells and tissue. Importantly, the underlying RASSF1C driven biological processes concur to explain clinical data: namely, methylation of the RASSF1C promoter correlates with better survival in early stage breast cancer patients. Therefore, we propose the use of RASSF1 gene promoter methylation status as a biomarker for patient stratification.

The main subject of this paper is a detailed description of the reproductive organs of a planarian initially described by Dendy as Geoplana triangulata. Five unusual features are observed in the reproductive system: 1. The vas deferens consists of a series of wide convoluted branching tubes extending from the region of the mouth to the anterior end of the seminal vesicle. 2. The penis is very small and inconspicuous. 3. The atrium masculinum is provided with three pairs of muscular gland-organs or adenodactyli. 4. The paired ovaries are situated one on each side of the seminal vesicle, not in the region of the brain as is usual. 5. Each ovary is a long fusiform body enclosing more than one true ovary or germarium, as well as specialized parovarian and amoeboid cells which are probably nutritive, and are associated with the internal opening of the oviduct. The writer refers Geoplana triangulata Dendy to the genus Artioposthia owing to the presence of adenodactyli in the atrium masculinum. Each adenodactylus encloses a glandular reservoir from which a ciliated duct leads to the atrial cavity. The actual function of the adenodactyli is obscure, but the very small size of the penis and the fact that the adenodactyli are extrusible suggests the possibility of these latter performing the function of a penis.

Naegleria fowleri is a pathogenic amoeboflagellate that can be evoked to transform from amoebae to flagellates by subculture to nonnutrient buffer. More than half of the amoebae of strains KUL, nN68, and Lovell became enflagellated 300 min after subculture to amoeba-saline, whereas no amoebae of strains NF66, NF69, and HB4 did. N. fowleri nN68 enflagellated best when grown at 32 or 37 degrees C and subcultured to amoeba-saline at 37 or 42 degrees C. Amoebae from the stationary phase of growth enflagellated more readily than did actively growing amoebae. Incubation in expended culture medium from stationary-phase cultures enhanced the capability of growing amoebae to enflagellate after subculture to amoebasaline. Enflagellation was more extensive when the population density in amoebasaline did not exceed 2 x 10(5) amoebae per ml. Cycloheximide at 1 microgram/ml and actinomycin D at 25 micrograms/ml inhibited growth of N. fowleri nN68. Cycloheximide at 0.5 microgram/ml and actinomycin D at 25 micrograms/ml completely prevented enflagellation when added at time zero. Cycloheximide at 0.5 microgram/ml, added 120 to 300 min after initiation of enflagellation, prevented further differentiation and caused existing flagellates to revert to amoeboid cells. Similarly, actinomycin D at 25 micrograms/ml, added 90 to 300 min after initiation of enflagellation, retarded differentiation and caused flagellates to revert. Radiolabeled precursors were incorporated into macromolecules during differentiation in nonnutrient buffer. Enflagellation of N. fowleri is a suitable model for studying regulation of a eucaryotic protist.

ABSTRACTCell guidance by anchored molecules, or haptotaxis, is crucial in development, immunology and cancer. Adhesive haptotaxis, or guidance by adhesion molecules, is well established for mesenchymal cells such as fibroblasts, whereas its existence remains unreported for amoeboid cells that require less or no adhesion in order to migrate. We show that, in vitro, amoeboid human T lymphocytes develop adhesive haptotaxis mediated by densities of integrin ligands expressed by high endothelial venules. Moreover, lymphocytes orient towards increasing adhesion with VLA-4 integrins (also known as integrin α4β1), like all mesenchymal cells, but towards decreasing adhesion with LFA-1 integrins (also known as integrin αLβ4), which has not previously been observed. This counterintuitive ‘reverse haptotaxis’ cannot be explained by existing mechanisms of mesenchymal haptotaxis involving either competitive anchoring of cell edges under tension or differential integrin-activated growth of lamellipodia, because they both favor orientation towards increasing adhesion. The mechanisms and functions of amoeboid adhesive haptotaxis remain unclear; however, multidirectional integrin-mediated haptotaxis might operate around transmigration ports on endothelia, stromal cells in lymph nodes, and inflamed tissue where integrin ligands are spatially modulated.
