3‐O‐Acetyl‐11‐keto‐β‐boswellic acid ameliorates Chronic Unpredictable Mild Stress induced HPA axis dysregulation in relation with Glutamate/GABA aberration in depressive Rats

Baihe Jizihuang Tang (BHT) is a traditional Chinese medicine (TCM) prescription, which can also be used as a nutritional food with medicinal value. Herein, we aimed to clarify the antidepressive effects and molecular mechanism of BHT. Network pharmacological analysis; chronic unpredictable mild stress (CUMS) rat model assessment; behavioral tests; analysis of hippocampal neurotransmitter levels, hippocampal pathological structure, and hypothalamic-pituitary-adrenal (HPA) axis; western blot analysis; 16s RNA sequencing; ultraperformance liquid chromatography (UPLC)/mass spectrometry (MS); and high-performance liquid chromatography (HPLC)/ultraviolet (UV) analysis were used. We found 8 potentially active components and 12 targets from the database. KEGG analysis suggested that BHT significantly affected BDNF/tyrosine receptor kinase B levels, glutamate binding, synaptic transmission based on neurotransmitter signal, and the response to glucocorticoid signaling pathways. Consistently, 7 chemical components were identified using UPLC/quadrupole time-of-flight/MS; among them, regalosides A, B, C, and E were unique components of lily of TCM, and their content in BHT was significantly different: regaloside A > B > E > C. BHT could nourish hippocampal neurons, affect neurotransmitter metabolism, reduce HPA axis hyperactivity, improve deficits in hippocampal tissue structure, and change depressive behavior. Moreover, BHT regulated BDNF expression in the hippocampus and improved intestinal flora deficits in CUMS rats by changing the content of Bifidobacterium, Rothia, Glutamicibacter, and Lactobacillus at the genus level. Collectively, BHT attenuated CUMS-induced depression-like behavior by regulating BDNF and intestinal flora disorder through the brain-gut axis. Therefore, including BHT in the medication list may constitute a potential strategy for preventing depression.

Purpose: To investigate the antidepressant mechanism of action of Crassifoside H (CH) from the rhizomes of Curculigo glabrescens (Hypoxidaceae) in chronic unpredictable mild stress (CUMS)-induced rats.Methods: CUMS-induced rat depressant model was established. Behavioral tests, viz, sucrose preference test (SPT), open field test (OFT) and forced swimming test (FST) were applied to assess the antidepressant effect of CH. Enzyme linked immunosorbent assay (ELISA) was used to assess thelevels of corticosterone (CORT), TNF-α and IL-1β in serum. Protein expressions of TNF-α, IL-1β and NLRP3 in rat hippocampus were determined by Western blot.Results: Crassifoside H significantly ameliorated CUMS-induced depressant-like behavior as the serum CORT level of CUMS rats. CH remarkably decreased TNF-α and IL-1β levels in serum and hippocampus of CUMS rats. Moreover, Crassifoside H significantly inhibited NLRP3 activation inhippocampus.Conclusion: The findings demonstrate that Crassifoside H has antidepressant effect on CUMS rats. The mechanism of action of CH may be at least partly due to the improvement of hypothalamic-pituitaryadrenal (HPA) axis dysfunction by decreasing serum CORT. These findings suggest that Crassifoside H has a therapeutic potential for the management of depression. ABSTRAKStres kronis dapat menginduksi aktivasi dari hipotalamus-hipofisis-adrenal (HPA) axis secara terus menerus sehingga dapat menyebabkan gangguan pada proses folikulogenesis. Tingginya kadar glukokortikoid akibat stres kronis dapat menginduksi apoptosis sehingga mengganggu homeostasis didalam folikel. Adanya apoptosis sel granulosa dapat menyebabkan gangguan pada folikulogenesis dengan menurunkan komunikasi antara sel granulosa - oosit yang kemudian mempengaruhi suplai nutrisi dan faktor pematangan oosit sehingga mempengaruhi kualitas oosit. Membuktikan jumlah apoptosis sel granulosa lebih tinggi pada kelompok stres kronis. Subyek penelitian menggunakan Rattus norvegicus betina yang dibagi menjadi 2 kelompok, kontrol dan perlakuan. Kelompok perlakuan diberikan paparan stres kronis melalui metode Chronic Unpredictable Mild Stress (CUMS) selama 21 hari. Pengamatan apoptosis sel granulosa menggunakan teknik imunohistokimia dan dideteksi dengan Terminal Deoxynucleotidyl Transferase dUTP Nick End Labeling (TUNEL). Apoptosis sel granulosa antar kelompok dianalisis menggunakan uji independent t-test. Pada pengamatan apoptosis sel granulosa folikel antral menyatakan adanya perbedaan bermakna (p=0.000) apoptosis sel granulosa folikel antral antara kelompok kontrol dan perlakuan. Mean ± SD kelompok kontrol 1.12 ± 0.48, kelompok perlakuan 3.65 ± 1.37. Dapat disimpulkan stres kronis memengaruhi peningkatan apoptosis sel granulosa folikel antral pada Rattus norvegicus.  Kata kunci: stres kronis, CUMS, apoptosis sel granulosa, folikel antral
