Abstract. Organic farming plays a considerable role in Austrian agriculture. Therefore it seems to be worthwhile to make suggestions about a more ecological breeding goal for cattle breeds. Because of the fact that Simmental is the most frequent dual purpose cattle breed in Austria, this breed is chosen to show the impact of selection under an ecological total merit index. With model calculations the effect on selection response in milk production beef production and functional traits under selection with the current economic total merit index and more ecological index variants, e.g. with higher economic values for fitness and functional traits, is shown. As a basis for future decisions the efficiency of more or less ecological indices is compared. The results indicate that an increase of the current economic values of functional traits of about 50 percent does not present a great risk, expected selection responses for milk production traits are still high.

German Black Pied cattle (DSN) is an endangered population of about 2,550 dual-purpose cattle in Germany. Having a milk yield of about 2,500 kg less than the predominant dairy breed Holstein, the preservation of DSN is supported by the German government and the EU. The identification of the genomic loci affecting milk production in DSN can provide a basis for selection decisions for genetic improvement of DSN in order to increase market chances through the improvement of milk yield. A genome-wide association analysis of 30 milk traits was conducted in different lactation periods and numbers. Association using multiple linear regression models in R was performed on 1,490 DSN cattle genotyped with BovineSNP50 SNP-chip. 41 significant and 20 suggestive SNPs affecting milk production traits in DSN were identified, as well as 15 additional SNPs for protein content which are less reliable due to high inflation. The most significant effects on milk yield in DSN were detected on chromosomes 1, 6, and 20. The region on chromosome 6 was located nearby the casein gene cluster and the corresponding haplotype overlapped the CSN3 gene (casein kappa). Associations for fat and protein yield and content were also detected. High correlation between traits of the same lactation period or number led to some SNPs being significant for multiple investigated traits. Half of all identified SNPs have been reported in other studies, previously. 15 SNPs were associated with the same traits in other breeds. The other associated SNPs have been reported previously for traits such as exterior, health, meat and carcass, production, and reproduction traits. No association could be detected between DGAT1 and other known milk genes with milk production traits despite the close relationship between DSN and Holstein. The results of this study confirmed that many SNPs identified in other breeds as associated with milk traits also affect milk traits in dual-purpose DSN cattle and can be used for further genetic analysis to identify genes and causal variants that affect milk production in DSN cattle.

In addition to the main components of the somatotrophic axis (GH/GHR/IGF-I/IGF-IR), great importance in the control of growth and development is also attached to the Janus kinase 2 (JAK2) pathway. Induced by the GH/GHR complex, JAK2 activates signal transducer and activator of transcription 5 (STAT5), and in consequence, may be involved in the regulation of expression of insulin-like growth factor I (IGF-I) in the mammary gland. Silent mutation (rs110298451) has been identified within exon 20 using polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP). A total of 904 individuals of four dairy or dual-purpose breeds (Polish Holstein-Friesian, Montbeliarde, Simmental and Jersey) were genotyped. A genotypic imbalance in the populations was observed. In the case of dual-purpose breeds (Montbeliarde and Simmental), the frequencies of both alleles were almost equal. In contrary, the JAK2G allele was predominant in the Polish Holstein-Friesian breed while JAK2A allele in Jersey. A pronounced relationship between JAK2/RsaI polymorphism and milk production traits was found where, irrespective of breed and lactation order, the GG genotype was significantly associated with higher milk, protein and fat yields, as compared to the AA genotype. Heterozygous individuals were generally characterised by intermediate values of the analysed milk traits. It can be argued that the JAK2 gene polymorphism is a potential marker for milk production traits. However, due to the fact that rs110298451 SNP does not directly affect amino acid sequence, other association studies involving missense mutation should also be performed.

The aim of the present study was to screen the genotype profile of bovine DGAT1 gene among Gir and Kankrej cattle in Gujarat, India. Total number of 200 Gir cattle and 100 Kankrej cattle were evaluated for CfrI-RFLP based genotyping of DGAT1 gene. We observed that only two genotypes (KK and KA) exist among the studied population with the alleles frequency of 0.925 (n=170) and 0.075 (n=30), respectively for K and A in Gir cattle as well as 0.895 (n=79) and 0.105 (n=21) respectively in Kankrej cattle. The overall calculated allele frequency for K and A was 0.915 and 0.085, respectively in both zebu (Bos indicus) cattle. Association of genotypes with milk production traits revealed that KA had significant (P<0.05) effect on total milk yield, as compared to KK in Gir and Kankrej cattle breed. Similarly, KK genotype had significant (P<0.05) effect on fat percentage, as compared to KA in Gir and Kankrej.

The study was carried out on 19 ewes of local Awassi sheep and 12ewes local Arabi sheep in the Al-kafeel sheep station Karbala, to determine the effect of breed and some non-genetic factors such as (sex of the lamb, type of birth, age and weight of ewes at birth) on daily and total milk production and lactation period and some of milk components (fat, protein and lactose). The results showed that a significant effect (P <0.05) of the breed on milk production traits where Awassi sheep recorded the highest mean (0.91 kg , 101.63 kg , 104.86 day) compared to the Arabi sheep she was means (0.77 kg , 88.15 kg , 99.15 day) respectively. As well as in proportions of milk components with mean( 5.1 , 4.90 , 5.51) % respectively compared to the Arabi sheep (4.70 . 4.20 . 4.89) ewes with male lambs also exceeded superior ewes with female lambs in daily and total milk production and the lactation period the sex of the lamb did not affect the proportions of milk components the weight of the ewes had a significant effect (P <0.05) in milk production attributes with superior weight of ewes on lower ewes and did not affect the proportions of milk ingredients except for lactose. The type of birth and the age of the ewes did not have a significant effect in all the studied traits except for the superiority (P<0.05) of young ewes on age ewes in the fat percentage of milk.

This Research Communication describes the association between genetic variation within the prolactin (PRL) gene and the milk production traits of Italian Mediterranean river buffalo (Bufala mediterranea Italiana). High resolution melting (HRM) techniques were developed for genotyping 465 buffaloes. The association of genetic polymorphism with milk production traits was performed and subsequently the effects of parity and calving season were evaluated. Single nucleotide polymorphisms (SNPs) were identified at exons 2 and 5 and at introns 1 and 2. All the SNPs were in Hardy–Weinberg equilibrium, and statistical analysis showed that the polymorphism of intron1 was significantly (P < 0·05) associated with milk yield, milk protein content and peak milk yield. The average contribution of the intron1 genotype (r2intron1) to total phenotypic variance in milk production traits was 0·09, and the TT genotype showed lower values than CC and CT genotypes. A nonsynonymous SNP was identified in exon 2, which resulted in an amino acid change from arginine to cysteine. Moreover, the polymorphism of exon 2 was associated significantly with milk fat content (P < 0·05), and the buffaloes with TT genotype showed higher total fat content than the buffaloes with CT genotype. These findings provide evidence that polymorphisms of the buffalo PRL gene are associated with milk production traits and PRL can be used as a candidate gene for marker-assisted selection in Italian Mediterranean river buffalo breeding.

Abstract Our objective was to determine the content of the bioactive protein osteopontin (OPN) in bovine milk and identify factors influencing its concentration. OPN is expressed in many tissues and body fluids, with by far the highest concentrations in milk. OPN plays a role in immunological and developmental processes and it has been associated with several milk production traits and lactation persistency in cows. In the present study, we report the development of an enzyme linked immunosorbent assay (ELISA) for measurement of OPN in bovine milk. The method was used to determine the concentration of OPN in milk from 661 individual Danish Holstein cows. The median OPN level was determined to 21.9 mg/l with a pronounced level of individual variation ranging from 0.4 mg/l to 67.8 mg/l. Breeding for increased OPN in cow's milk is of significant interest, however, the heritability of OPN in milk was found to be relatively low, with an estimated value of 0.19 in the current dataset. The variation explained by the herd was also found to be low suggesting that OPN levels are not affected by farm management or feeding. Interestingly, the concentration of OPN was found to increase with days in milk and to decrease with parity.
