From a statistical data bank containing only anonymous records, the records sometimes may be identified and then retrieved, as personal records, by on line dialogue. The risk mainly applies to statistical data sets representing populations, or samples with a high ratio n/N. On the other hand, access controls are unsatisfactory as a general means of protection for statistical data banks, which should be open to large user communities. A threat monitoring scheme is proposed, which will largely block the techniques for retrieval of complete records. If combined with additional measures (e.g., slight modifications of output), it may be expected to render, from a cost-benefit point of view, intrusion attempts by dialogue valueless, if not absolutely impossible. The bona fide user has to pay by some loss of information, but considerable flexibility in evaluation is retained. The proposal of controlled classification included in the scheme may also be useful for off line dialogue systems.

The effect of coating structure variations on flexographic print quality was studied using pilot-coated paperboard samples with different latex content and latex particle sizes. Two latexes, with particle sizes of 120 nm and 160 nm, were added at either 12 parts per hundred (pph) or 18 pph to the coating formulation. The samples were printed with full tone areas at print forces of 25 N and 50 N in a laboratory flexographic printing press using a waterbased ink. A high ratio of uncovered areas (UCAs) could be detected for the samples that contained 18 pph latex printed at a print force of 25 N. UCAs decreased with increased print force and with decreased amounts of latex in the coating formulation. The fraction of latex covered area on the coating surface was estimated to be 0.35–0.40 for the 12 pph, and 0.70–0.75 for the 18 pph samples. The ink penetration depth into the coating layer could be linked to the fraction of latex-free areas on the coating surface. Optical cross section microscopy indicated that a higher printing force did not increase the depth of penetrated ink to any greater extent. Higher printing force did increase contact between plate and substrate, leading to an improved distribution of the ink. This, in turn, increased print density and decreased UCAs. On closer inspection, the UCAs could be categorized as being induced by steep topographic changes. When appearing at other locations, they were more likely to be caused by poor wetting of the surface. To understand the wetting behavior of the coating surface, observed contact angles were compared with calculated contact angles on surfaces of mixed composition.

Capsules of different formulations were prepared by using a hydrophilic polymer, xanthan gum and a filler Ludipress. Metformin hydrochloride, which is an anti-diabetic agent, was used as a model drug here with the aim to formulate sustained release capsules. In the first 6 formulations, metformin hydrochloride and xanthan gum were used in different ratio. Later, Ludipress was added to the formulations in a percentage of 8% to 41%. The total procedure was carried out by physical mixing of the ingredients and filling in capsule shells of size ‘1’. As metformin hydrochloride is a highly water soluble drug, the dissolution test was done in 250 ml distilled water in a thermal shaker (Memmert) with a shaking speed of 50 rpm at 370C &plusmn 0.50C for 6 hours. After the dissolution, the data were treated with different kinetic models. The results found from the graphs and data show that the formulations follow the Higuchian release pattern as they showed correlation coefficients greater than 0.99 and the sustaining effect of the formulations was very high when the xanthan gum was used in a very high ratio with the drug. It was also investigated that the Ludipress extended the sustaining effect of the formulation to some extent. But after a certain period, Ludipress did not show any significant effect as the pores made by the xanthan gum network were already blocked. It is found here that when the metformin hydrochloride and the xanthan gum ratio was 1:1, showed a high percentage of drug release, i.e. 91.80% of drug was released after 6 hours. But With a xanthan gum and metformin hydrochloride ratio of 6:1, a very slow release of the drug was obtained. Only 66.68% of the drug was released after 6 hours. The percent loading in this case was 14%. Again, when Ludipress was used in high ratio, it was found to retard the release rate more prominently. Faecal enterococci ecology outside the host is of great relevance when using these organisms as indicators of water quality. As a complement to New Zealand epidemiological studies of bathing water quality and health risk, a study of the environmental occurrence of these organisms has been undertaken. Specific concerns over the use of enterococci derive from the unique situation in New Zealand which has few chlorinated sewage effluents, a high ratio of grazing animals to humans, and significant inputs of animal processing effluents into the environment. Human and animal faecal wastes are the main sources, with 106–107cfu/100ml found in human sewage. Analysis of domestic and feral animal faeces found enterococci in the range of 101–106cfu/g with considerable variation between species. The latter observations support the notion that a considerable proportion of the load in urban/rural catchments and waterways (typically 102–103 enterococci cfu/100ml) is derived from non-human sources. Previous studies of enterococci quiescence in marine/fresh waters indicate that they enter a non-growth phase, exposure to sunlight markedly reducing culturability on selective and non-selective media. Enterococci were also found to survive/multiply within specific non-faecal environments. Enterococci on degrading drift seaweed at recreational beaches exceeded seawater levels by 2–4 orders of magnitude, suggesting that expansion had occurred in this permissive environment with resultant potential to contaminate adjacent sand and water. These studies suggest that multiple sources, environmental persistence, and environmental expansion of enterococci within selected niches add considerable complexity to the interpretation of water quality data.

Abstract Background Whole-genome sequencing (WGS) is becoming increasingly useful to study the biology, epidemiology, and ecology of malaria parasites. Despite ease of sampling, DNA extracted from dried blood spots (DBS) has a high ratio of human DNA compared to parasite DNA, which poses a challenge for downstream genetic analyses. The effects of multiple methods for DNA extraction, digestion of methylated DNA, and amplification were evaluated on the quality and fidelity of WGS data recovered from DBS. Methods Low parasite density mock DBS samples were created, extracted either with Tween-Chelex or QIAamp, treated with or without McrBC, and amplified with one of three different amplification techniques (two sWGA primer sets and one rWGA). Extraction conditions were evaluated on performance of sequencing depth, percentiles of coverage, and expected SNP concordance. Results At 100 parasites/μL, Chelex-Tween-McrBC samples had higher coverage (5 × depth = 93% genome) than QIAamp extracted samples (5 × depth = 76% genome). The two evaluated sWGA primer sets showed minor differences in overall genome coverage and SNP concordance, with a newly proposed combination of 20 primers showing a modest improvement in coverage over those previously published. Conclusions Overall, Tween-Chelex extracted samples that were treated with McrBC digestion and are amplified using 6A10AD sWGA conditions had minimal dropout rate, higher percentages of coverage at higher depth, and more accurate SNP concordance than QiaAMP extracted samples. These findings extend the results of previously reported methods, making whole genome sequencing accessible to a larger number of low density samples that are commonly encountered in cross-sectional surveys.
