ABSTRACT Cross-inhibition by quorum-sensing pheromones betweenStaphylococcus aureus and Staphylococcus epidermidis was investigated using all known S. aureus agr pheromone subgroups. All S. aureus subgroups were sensitive towards the S. epidermidis pheromone, with the exception of the recently identified subgroup 4. The subgroup 4 pheromone was also the only S. aureus pheromone able to inhibit the S. epidermidis agr response. The close relation of subgroup 4 to subgroup 1 suggests that subgroup 4 might have evolved from subgroup 1 by mutation under the selective pressure of competition with S. epidermidis. The competition between S. aureus and S. epidermidis by means of quorum-sensing cross talk seems to be generally in favor of S. epidermidis, which might explain the predominance of S. epidermidis on the skin and in infections on indwelling medical devices.

ABSTRACT Recent studies highlight the abundance of commensal coagulase-negative staphylococci (CoNS) on healthy skin. Evidence suggests that CoNS actively shape the skin immunological and microbial milieu to resist colonization or infection by opportunistic pathogens, including methicillin-resistant Staphylococcus aureus (MRSA), in a variety of mechanisms collectively termed colonization resistance. One potential colonization resistance mechanism is the application of quorum sensing, also called the accessory gene regulator (agr) system, which is ubiquitous among staphylococci. Common and rare CoNS make autoinducing peptides (AIPs) that function as MRSA agr inhibitors, protecting the host from invasive infection. In a screen of CoNS spent media, we found that Staphylococcus simulans, a rare human skin colonizer and frequent livestock colonizer, released potent inhibitors of all classes of MRSA agr signaling. We identified three S. simulans agr classes and have shown intraspecies cross talk between noncognate S. simulans agr types for the first time. The S. simulans AIP-I structure was confirmed, and the novel AIP-II and AIP-III structures were solved via mass spectrometry. Synthetic S. simulans AIPs inhibited MRSA agr signaling with nanomolar potency. S. simulans in competition with MRSA reduced dermonecrotic and epicutaneous skin injury in murine models. The addition of synthetic AIP-I also effectively reduced MRSA dermonecrosis and epicutaneous skin injury in murine models. These results demonstrate potent anti-MRSA quorum sensing inhibition by a rare human skin commensal and suggest that cross talk between CoNS and MRSA may be important in maintaining healthy skin homeostasis and preventing MRSA skin damage during colonization or acute infection.

ABSTRACT Staphylococcus aureus is a prevalent cause of bacterial infections associated with indwelling medical devices. RNA III inhibiting peptide (RIP) is known to inhibit S. aureus pathogenesis by disrupting quorum-sensing mechanisms. RIP was tested in the present study for its ability to inhibit S. aureus biofilm formation in a rat Dacron graft model. The activity of RIP was synergistic with those of antibiotics for the complete prevention of drug-resistant S. aureus infections.

This study aimed to evaluate the bactericidal activity of common disinfectants used for surfaces and medical devices. Sodium hypochlorite (D1), disinfectant (D2) composed of N-(3-aminopropyl)-N-dodecylpropane-1,3-diamine, chloride de didecyldimethylammonium, and disinfectant (D3) composed of Didecyldimethylammonium chloride and Polyhexamethylene biguanide hydrochloride, were tested against 15 strains isolated from the hospital environment and four reference bacteria. The microdilution method was performed to assess antimicrobial activity. The susceptibility was evaluated by comparing the minimum inhibitory dilution with the dilution of disinfectant recommended by the manufacture. D1 and D2 were active against Staphylococcus epidermidis, Staphylococcus saprophyticus, Enterobacter cloacae, Escherichia coli, Pseudomonas fluorescens, Methicillin-resistant Staphylococcus aureus, Bacillus spp, Corynebacterium spp, Gram-positive bacillus, Escherichia coli ATCC 25922, Bacillus subtilis ATCC 3366, and Pseudomonas aeruginosa ATCC 27853 strains but not active against Micrococcus spp, and Staphylococcus aureus ATCC 29213. D3 was ineffective against Micrococcus spp, Bacillus Gram Positive, Staphylococcus epidermidis, and Escherichia coli ATCC 25922. Therefore, D1 and D2 can eliminate most pathogenic bacteria in hospitals, in comparison to D3. It is necessary to monitor the antibacterial activity of disinfectants against reference strains but also against those usually present on surfaces. The obtained results could have promising applications in controlling the emergence of nosocomial infections.

Тухайн ургамлын нийлбэр алкалоид, хлороформ, этилацетатын хандны куарум сенсинг механизмыг дарангуйлах (quorum sensing (QS) system) идэвх нь 30 мг/мл тунд 10.0 ± 0.0 мм, 15.5 ± 2.1 мм, 12.0 ± 4.24 мм; хлороформын хандны Staphylococcus aureus (S. aureus), Micrococcus luteus (M. luteus), Staphylococcus epidermidis (S. epidermidis) бактерийн эсрэг идэвх нь 10 мг/мл тундаа 11.4 мм, 13.12 мм, 15.16 мм байв. Харин тухайн ургамлын мутагенийг дарангуйлах идэвх нь петролейны эфир, хлороформ, этилацетат, н-бутанол, усан үлдэгдэл болон нийлбэр алкалоидод 8%, 62%, 46%, 7%, 44%, 59% байв. М. чоногоно ургамлын метанолын хандны нийлбэр фенолт нэгдлийн агууламж нь 14,1±0,3 мг/г, антиоксидант идэвх 3,6 ± 0,3 µг/мл (ABTS), 1,3 ± 0,1 µг/мл (DPPH) тус тус хэмжээтэй байв.

Zusammenfassung: Gegenstand und Ziel: Im Gegensatz zur Hündin liegen bei der Katze bisher wenige Studien über die genitale Keimflora geschlechtsgesunder Tiere vor. Ziel der Untersuchung war daher, physiologische Daten über die aerobe Vaginalflora bei dieser Spezies zu gewinnen. Material und Methoden: Für die vorliegende Studie standen 26 gesunde, anöstrische Katzen zur Verfügung, die zu einer Ovariohysterektomie vorgestellt wurden. Nach einer klinischen Untersuchung wurden von allen Probanden unter sterilen Bedingungen Vaginaltupfer entnommen. Ergebnisse: In allen Proben konnte ein Bakterienwachstum mit durchschnittlich zwei verschiedenen Bakterienspezies nachgewiesen werden. Die Gesamtkeimgehalte wurden bei 50% der Vaginaltupferproben als gering-, bei 15% als mittel- und bei 35% als hochgradig beurteilt. Vorherrschend waren Mischkulturen aus zwei bis vier verschiedenen Keimarten. Monokulturen wurden aus 38% der Tupferproben isoliert. Am häufigsten gelang der Nachweis von E. coli variatio haemolytica (E. coli var. haem.) (58%) und Staphylococcus epidermidis (42%). Als weitere Spezies wurden E. coli, α-, β-hämolysierende Streptokokken, anhämolysierende Streptokokken, aerobe Bazillen, Staphylococcus aureus, Staphylococcus intermedius, Pasteurella multocida sowie Klebsiellen isoliert. Auffällig ist die hohe Nachweisrate von E. coli var. haem. mit 35% in Mischkulturen und 23% in Reinkultur. Schlussfolgerungen: Die physiologische Mikroflora der felinen Vaginalschleimhaut differiert deutlich von der der anöstrischen Hündin. Besonders die Dominanz von E. coli var. haem. in 38% der Mischkulturen und 23% der Monokulturen bei der Katze ist hervorzuheben. Klinische Relevanz: Die vorliegenden Ergebnisse geben eine erste Grundlage für die Interpretation mikrobiologischer Befunde feliner Vaginaltupfer.
