Lead–radium dating of orange roughy (Hoplostethus atlanticus): validation of a centenarian life span

Life-span estimates for orange roughy ( Hoplostethus atlanticus ) range from ~20 years to well over 100 years. In this study, an improved lead–radium dating technique provided independent age estimates from sagittal otoliths. This technique used the known properties of radioactivity for lead-210 and radium-226 to determine the validity of fish age estimates. An improvement to lead–radium dating using mass spectrometry allowed the use of smaller samples than previously possible; therefore, an application was made to otolith cores, the first few years of otolith growth. This approach circumvented the use of whole otoliths and alleviated many of the assumptions that were necessary in previous lead–radium dating applications. Hence, it was possible to critically evaluate lead–radium dating as a tool in fish age validation. The measurement of lead–radium ratios for a series of age groups that consisted of otolith cores, grouped based on growth-zone counts from thin sections, showed a high degree of correlation to the expected lead–radium ingrowth curve. This finding provided support for age estimation procedures using thin otolith sectioning. As independent estimates of age, the results indicated that fish in the oldest age group were at least 93 years old, providing robust support for a centenarian life span.

Ages of orange roughy (Hoplostethus atlanticus) determined by two methods (counting annuli on the surface of whole and in longitudinally sectioned otoliths) were similar up to maturity. Beyond maturity, age estimates from sectioned otoliths exceeded those from whole otoliths. Maximum recorded age was 125 years for an individual 41 cm standard length (SL), and age at maturity was estimated to be 25 years (30–32 cm SL). These are consistent with ages estimated previously by radiometric methods. Results demonstrated a two-stage linear relationship between otolith weight and age that confirmed the two-stage otolith mass growth model previously used in radiometric ageing. However, in the radiometric analyses the reduction in otolith growth was arbitrarily estimated at 45% of the immature rate whereas annuli data demonstrated a reduction after maturity to 62% of the immature rate. The new estimates of otolith mass growth rate were incorporated into the radiometric data and ages recalculated, which reduced age estimates for 38–40 cm SL fish from 77–149 to 59–101 years. The radiometric data were also recalculated using only the percentage reduction in otolith growth after maturity, giving the radiometric age of 125 ± 9 years for the oldest fish.

Black cardinalfish (Epigonus telescopus, Apogonidae) is an important component of deepsea commercial fishing activity in the New Zealand region. It is estimated to live longer than 100 years on the basis of counts of unvalidated annual growth zones in otoliths. Age-validation procedures for long-lived fishes are often one of the following two techniques: (1) lead–radium disequilibria, which uses the natural decay of radium-226 into lead-210 as a natural clock; or (2) bomb radiocarbon (Δ14C) dating, which relies on the marine signal created by nuclear testing. The high estimated lifespan, as well as the large size of the otolith core region, make E. telescopus an excellent candidate for a combined application of these two independent age-validation techniques. The lead–radium dating using otolith cores indicated that growth-zone counts less than ~60 years were consistent with radiometric ages, whereas higher counts appeared to be under-estimates. There was 95% confidence that maximum age was at least 95 years. The validation indicated that fish aged over 60 years tended to be under-aged by up to 30%. The bomb radiocarbon levels in otolith cores supported age estimates up to ~40 years made from zone counts, and by inference from the zone counts validated with lead–radium dating, longevity exceeds 100 years.

Demersal reef fishes of the Indo-Pacific are under increasing pressure as a fisheries resource, yet many of the important life history characteristics required for suitable management are poorly known. The three fish species, eightbar grouper (Hyporthodus octofasciatus), ruby snapper (Etelis carbunculus) and the spangled emperor (Lethrinus nebulosus), are important components of fisheries and ecosystems throughout the Indo-Pacific. Despite their importance, age and growth information is incomplete. Age has been estimated for E. carbunculus and L. nebulosus, but validated age beyond the first few years is lacking and for H. octofasciatus no age estimates exist. Bomb radiocarbon dating can provide age estimates that are independent of growth-zone counting, but only if appropriate reference Δ14C chronologies exist. In this study, a series of Δ14C records from hermatypic corals was assembled to provide a basis for bomb radiocarbon dating in the western Indo-Pacific region. Results provided (1) valid age estimates for comparison to age estimates from two facilities investigating growth-zones in otolith thin sections; (2) support for age estimation protocols using otolith thin sections; and (3) the information necessary for further refinement of age estimation procedures. Estimates of longevity from bomb radiocarbon dating agree with some prior studies: H. octofasciatus, E. carbunculus and L. nebulosus all being long-lived species with life spans of at least 43, 35 and 28 years respectively.

Estimates of age and growth of yellowfin (Thunnus albacares) and bigeye (Thunnus obesus) tuna remain problematic because validation of growth zone deposition (opaque and translucent) has not been properly evaluated. Otolith growth structure (zone clarity) can be poorly defined for tropical tunas, but the use of bomb radiocarbon dating has validated age estimates to 16–18 years for yellowfin and bigeye tuna. Use of the radiocarbon decline period — defined by regional coral and otoliths — provided valid ages through ontogeny. Yellowfin tuna aged 2–18 years (n = 34, 1029–1810 mm FL) and bigeye tuna aged 3–17 years (n = 12, 1280–1750 mm FL) led to birth years that were coincident with the bomb radiocarbon decline. The results indicate there was no age reading bias for yellowfin tuna and that age estimates of previous studies were likely underestimated for both species.

Of 4657 snapper (Pagrus auratus) that were tagged, injected with oxytetracycline (OTC), and released in 1986-87, 1113 fish were recaptured after periods at liberty from 18 days to 3 years. An OTC mark was clearly visible in thin sections of all but four of a sample of 125 otoliths. In 90% of the remaining 121 otoliths the position of the OTC mark was correctly inferred from age readings (made without seeing the OTC mark) based on annual ring counts in otolith sections. It is concluded that the otolith rings are deposited annually, and estimates are made of error rates in age estimation-both in identifying annual rings and in interpreting the otolith edge. Further support for the ageing method is contributed by an analysis of the growth of tagged fish, and by year-to-year consistency in the relative strength of cohorts. The growth analysis showed a large (more than twofold) interyear variation in growth rates. Snapper (and other sparid) age estimates based on scale readings may be underestimates. It is recommended that age validation studies clearly distinguish between three separate factors: periodicity of marks, identification of marks, and edge interpretation.

Errors can play a major role for optimizing subsequent performance: Response conflict associated with (near) errors signals the need to recruit additional control resources to minimize future conflict. However, so far it remains open whether children and older adults also adjust their performance as a function of preceding response conflict. To examine the life span development of conflict detection and resolution, response conflict was elicited during a task-switching paradigm. Electrophysiological correlates of conflict detection for correct and incorrect responses and behavioral indices of post-error adjustments were assessed while participants in four age groups were asked to focus on either speed or accuracy. Despite difficulties in resolving response conflict, the ability to detect response conflict as indexed by the Ne/ERN component was expected to mature early and be preserved in older adults. As predicted, reliable Ne/ERN peaks were detected across age groups. However, only for adults Ne/ERN amplitudes associated with errors were larger compared to Nc/CRN amplitudes for correct trials under accuracy instructions, suggesting an ongoing maturation in the ability to differentiate levels of response conflict. Behavioral interference costs were considerable in both children and older adults. Performance for children and older adults deteriorated rather than improved following errors, in line with intact conflict detection, but impaired conflict resolution. Thus, participants in all age groups were able to detect response conflict, but only young adults successfully avoided subsequent conflict by up-regulating control.

SummaryCase histories suitable for statistical evaluation can be found even as far back as in the Corpus Hippocraticum. Such simple data as the patient’s age, body weight, size, the date of menarche, etc. are practically always included in the case records, and it is demonstrated that, when such data are recorded in a system of documentation suitable for mechanical sorting, it may enable us to draw conclusions of very great importance. Mechanical registration methods have revealed that, in the determination and recording of data as hitherto carried out. there has been a surprisingly large number of errors and a high degree of unreliability. This view has a considerable influence on modern clinical methods; it renders a more democratic relation between physician and patient necessary and makes clear the need for measures to enhance the reliability of diagnosis and treatment of pathological conditions. The author illustrates this view with reference to the mechanical falsification of the thesis of the proneness of early age groups to lupus vulgaris, furthermore with reference to the mechanical rationalization of modern routine diagnostic methods, to the constant surveillance of adverse effects on public health and to the protection against allergic reactions with the aid of recording systems of personal allergy and intolerance data with mechanical sorting and computer techniques.

Проведена статистическая обработка сформированной базы данных показателей неспецифической резистентности MS «Excel» с целью изучения динамики иммунитета по параметрам бактерицидной, лизоцимной и фагоцитарной активности у крупного рогатого скота черно-пестрой породы разных возрастов. Было установлено, что средние значения показателей лизоцимной и бактерицидной активности у телят в 6 мес были ниже (47,89% и 49,09%, соответственно), чем у животных в 3—4 года (53,75% и 52,81%). Выведено уравнение регрессии, описывающее взаимосвязь между средними показателями фагоцитарной активности и возрастом при высокой степени достоверности (R2=0,90). Изменения лизоцимной и бактерицидной активности в зависимости от возраста были недостоверными (R2=0,58 и R2=0,49, соответственно). Установлена высокая корреляция между бактерицидной и лизоцимной активностью. Отмечена динамика изменения неспецифического иммунитета у коров различных возрастных групп. Создание баз данных и постоянный мониторинг основных показателей естественной резистентности в зависимости от различных факторов (породы, возраста, стадий лактации, кормления, способов содержания), которые влияют на данные показатели, необходимо для оценки физиологического состояния организма животных и их устойчивости к болезням. Statistical processing of the generated MS "Excel" on indicators of non-specific resistance database was carried out with the aim of studying the dynamics of the development of immunity in terms of bactericidal, lysozyme and phagocytic activity in black-and-white cattle of different ages. It was found that the average values of indicators of lysozyme and bactericidal activity in calves at 6 months were lower (47.89% and 49.09%, respectively) than in animals at the age of 3—4 years (53.75% and 52.81%). The equation of regression was derived, it describes the relationship between the average indicators of phagocytic activity and the age with a high degree of reliability (R2=0.90). The changes in lysozyme and bactericidal activity depending on age were dubious (R2=0.58 and R2=0.49, respectively). The high correlation between bactericidal and lysozyme activity was established. It was noticed that the cows of different age groups experienced the dynamics of changes in nonspecific immunity. The creation of databases and constant monitoring of the main indicators of natural resistance, depending on various factors (breed, age, stages of lactation, feeding, methods of keeping) that affect these indicators, is necessary for assessing the physiological state of the animals’ body of and their resistance to diseases.
