Stabilities of uncomplemented and complemented M15 β-galactosidase (Escherichia coli) and the relationship to α-complementation

M15 β-galactosidase (Escherichia coli) is a mutant form of β-galactosidase having residues 11-41 deleted. It is an inactive dimer but can be complemented to the active tetrameric form by the addition of a peptide containing the deleted residues. The activities of uncomplemented and complemented M15 β-galactosidases decreased starting at 42°C-uncomplemented over a narrow temperature range, complemented over a broad range. This is because uncomplemented protein is a simple dimer while complemented is a mix of interacting oligomers at high temperatures. The effects of added components on stability and α-complementation are best explained by binding effects on equilibria between native forms and forms susceptible to inactivation. Mg2+ stabilized complemented protein but destabilized uncomplemented protein (10× less Mg2+ was needed for complemented protein). α-Complementation increased somewhat at low Mg2+ but decreased at high Mg2+. These effects can be explained by differential Mg2+ binding to the native and susceptible forms. The enhancement of both stability and α-complementation by Na+ can be explained by preferential binding of Na+ to the native forms of both the uncomplemented and complemented proteins. Low 2-mercaptoethanol concentrations stabilized uncomplemented M15 β-galactosidase, but high concentrations destabilized it. All concentrations destabilized complemented M15 β-galactosidase. α-Complementation was enhanced by 2-mercaptoethanol. Thus, there is a correlation between stability of the uncomplemented protein and α-complementation at low 2-mercaptoethanol owing to interactions with native forms. The lack of correlation at higher 2-mercaptoethanol probably results from precipitation by 2-mercaptoethanol. In contrast to irreversible thermal inactivation, differences in reversible stability in urea were small. This suggests that quaternary structure and Mg2+ and Na+ sites are lost at low urea concentrations and are unimportant at the urea concentrations that result in reversible denaturation. RESUMEN En el artículo se ofrece una reflexión en torno a la educación y valores. Tras una referencia a los diferentes sentidos en que se puede plantear el problema en función de la forma como se puede entender la relación entre los dos sustantivos “educación” y “valores”, la reflexión se centra en algunos problemas relacionados con los valores en cuanto contenidos de la educación. Primeramente se refiere al problema del criterio en función del cual determinar los valores de la educación, concluyendo en que el criterio no podía ser ni de carácter a priori, ni empírico, sino “sintético”. A continuación, se afronta el problema del principio, de la fuente de los valores, o la concreción del criterio de los valores de la educación, entendiendo que éstos deberían ser determinados a partir del sujeto de la educación. Se concluye con la referencia a una exigencia de los valores de la educación, la configuración de una totalidad unitaria e interactiva. After a reference to the different senses in which one can pose the problem in terms of how you can understand the relationship between the two nouns “education” and “values”, reflection focuses on some problems related to the values in the contents of education. First, it concerns the problem of the criterion against which to determine the values of education, concluding that the criterion could be neither a priori in nature, not empirical, but “synthetic”. Herein, the problem of principle is faced, the source of values, or the realization of the criterion of the values of education, understanding that these should be determined from the subject of education. Objectives: To study the relationship between serum estradiol and physical disorders after hysterectomy. Methods: A descriptive cross sectional study on 151 women after total hysterectomy from 12/2008 to 11/2010 at Hue Cental Hospial. Results: Serious disorders in total hysterectomy group include: physical disorders (34.78%), maternal disorders (71.74%), sexual disorders (58.70%). There were significant differences these disorders between total hysterectomy group and hysterectomy with/without accessiory remove group, as well as menopause group. Conclusion: Serum estradiol levels was inversely correlated with physical disorders, martenal disorders, sexual disorders and other disorders. Key words: hysterectomy, serum estradiol

Recently, numerous studies have found that particulate matter (PM) exposure is correlated with increased hospitalization and mortality from heart failure (HF). In addition to problems with circulation, HF patients often display high expression of cytokines in the failing heart. Thus, as a recurring heart problem, HF is thought to be a disorder characterized in part by the inflammatory response. In this review, we intend to discuss the relationship between PM exposure and HF that is based on inflammatory mechanism and to provide a comprehensive, updated evaluation of the related studies. Epidemiological studies on PM-induced heart diseases are focused on high concentrations of PM, high pollutant load exposure in winter, or susceptible groups with heart diseases, etc. Furthermore, it appears that the relationship between fine or ultrafine PM and HF is stronger than that between HF and coarse PM. However, fewer studies paid attention to PM components. As for experimental studies, it is worth noting that coarse PM may indirectly promote the inflammatory response in the heart through systematic circulation of cytokines produced primarily in the lungs, while ultrafine PM and its components can enter circulation and further induce inflammation directly in the heart. In terms of PM exposure and enhanced inflammation during the pathogenesis of HF, this article reviews the following mechanisms: hemodynamics, oxidative stress, Toll-like receptors (TLRs) and epigenetic regulation. However, many problems are still unsolved, and future work will be needed to clarify the complex biologic mechanisms and to identify the specific components of PM responsible for adverse effects on heart health.

Since 2013, there has been an increase (>23%) in naval traffic using maritime routes and ports on the coastal fringe of Santa Marta, Colombia. Of major concern, and described by several studies, is the relationship between maritime traffic and coastal contamination. This study proposed a maritime traffic indicator considering the simultaneous effects of several relevant measurements of water quality parameters to estimate the impact of naval activity. The approach involved developing a model including the number of vessels, hull length, and permanence time in berths. In addition, water quality variables, considering climatic seasons, were used to verify association with maritime traffic and touristic activities. The high concentrations of total coliforms (TC) and dissolved/dispersed petroleum hydrocarbons in chrysene equivalents (DDPH) reported by the International Marina of Santa Marta (SM) were affected by the local anthropic activities, including tourism, naval traffic, and urban wastewater discharges. Moreover, our results suggest the occurrence of multiple chemical impacts within Tayrona National Natural Park (PNNT) affecting conservation goals. The estimation of the maritime traffic indicator proposed in this study may be an easy and more complete tool for future studies evaluating the impact of naval activities on environmental quality.

Whereas the so-called ‘citizenship premium’ in the labour market has been widely studied, we know little about how naturalisation affects immigrants’ lives beyond work and income. Focusing on the Netherlands, this paper analyses the relationship between citizenship acquisition and immigrant residential mobility, in particular the propensity of immigrants to move away from areas with high concentrations of migrants. We draw on register data from Statistics Netherlands ( N = 234,912). We argue that possessing Dutch citizenship reduces spatial stratification by diminishing the risk of housing market discrimination, thereby facilitating mobility outside of migrant-concentrated areas. Our findings show that naturalised immigrants are 50% more likely to move out of concentrated neighbourhoods, all else constant. The effect of naturalisation is especially relevant for renting without housing benefits and for home ownership, and for mid-risk immigrants who earn around the median income and hold permanent jobs, whose applications face strong scrutiny from landlords, rental agencies and mortgage lenders.
