The study aimed to evaluate the suitability of Escherichia coli , enterococci, and Clostridium perfringens for assessing the microbiological quality of roof-harvested rainwater and assessing whether the concentrations of these faecal indicators can be used to predict the presence or absence of specific zoonotic bacterial or protozoan pathogens. From a total of 100 samples tested, 58%, 83%, and 46% of samples were found to be positive for, respectively, E. coli, enterococci, and Clostridium perfringens spores, as determined by traditional culture-based methods. Additionally, in the samples tested, 7%, 19%, 1%, 8%, 17%, and 15% were PCR positive for Aeromonas hydrophila lip, Campylobacter coli ceuE, Campylobacter jejuni mapA, Legionella pneumophila mip, Salmonella invA, and Giardia lamblia β-giardin genes, respectively. However, none of the samples was positive for E. coli O157 lipopolysaccharide, verocytotoxin 1, and verocytotoxin 2 and Cryptosporidium parvum oocyst wall protein genes. The presence or absence of these potential pathogens did not correlate with any of the faecal indicator bacterial concentrations as determined by a binary logistic regression model. The roof-harvested rainwater samples tested in this study appeared to be of poor microbiological quality, and no significant correlation was found between the concentration of faecal indicators and pathogenic microorganisms. The use of faecal indicator bacteria raises questions regarding their reliability in assessing the microbiological quality of water and particularly their poor correlation with pathogenic microorganisms. The presence of one or more zoonotic pathogens suggests that the microbiological analysis of water should be performed and that appropriate treatment measures should be undertaken, especially in tanks where the water is used for drinking.

The microbiological quality of 1,012 hot meals served on aircraft was studied in 1991 to 1994. The material included dishes prepared in 33 countries all over the world. The geometric means of aerobic colony counts and Escherichia coli were significantly lower in breakfasts (P < 0.05) than in main dishes. Pathogenic bacteria were found in 30 samples (3.0%), Bacillus cereus being the most common pathogen. The frequencies of B. cereus and Staphylococcus aureus did not differ between breakfasts and main dishes. Many of the samples exceeded the minimum acceptable microbiological standards recommended by the Association of European Airlines (AEA) for E. coli, S. aureus, B. cereus, Clostridium perfringens, and Salmonella; i.e., 8.2%, 0.6%, 0.7%, 0.7% and 0.3%, respectively. There were considerable differences in aerobic colony counts and E. coli counts between countries where the food was prepared.

The good installation, as well as commissioning plan, of a water network is a crucial step in reducing the risk of waterborne diseases. The aim of this study was to monitor the microbiological quality of water from a newly built pavilion before it commenced operation. Overall, 91 water samples were tested for coliforms, Escherichia coli, enterococci, Pseudomonas aeruginosa and Legionella at three different times: T0 (without any water treatment), T1 (after treatment with hydrogen peroxide and silver ions at initial concentration of 20 mg/L and after flushing of water for 20 min/day for seven successive days) and T2 (15 days later). Coliforms were detected in 47.3% of samples at T0, 36.3% at T1 and 4.4% at T2. E. coli was isolated in 4.4% of the samples only at T1, while enterococci appeared in 12.1% of the samples at T1 and in 2.2% at T2. P. aeruginosa was isolated in 50.5% of the samples at T0, 29.7% at T1 and 1.1% at T2. Legionella pneumophila serogroup 8 was isolated in 80.2% of the samples at T0, 36.3% at T1 and 2.2% at T2. Our results confirmed the need for a water safety plan in new hospital pavilions to prevent the risk of waterborne diseases.

Freshly prepared and pasteurized fruit juices sold by vendors in local market of Rawalpindi city from 10 locations were analyzed for the microbiological quality. Total viable count (TVC), total coliform, faecal coliform, molds and the presence of pathogenic microorganisms such as E. coli, Pseudomonas, Staphylococcus aureus, Salmonella, and fungi like Aspergillus, Pencillum, Rhizopus were determined .In open fruit juices available in city were highly contaminated with bacteria and fungi. E. coli, Salmonella, Staphylococcus, and Pseudomonas were isolated with different frequency. Aspergillus, Pencillum and Rhizopus were also found in juices especially Aspergillus was with high percentages. The pasteurized juices have less contamination as compared to the fresh juices samples. The number and type of microorganisms recovered from the freshly squeezed fruit juices made them unsafe for drinking. The results of this study demonstrate the unhygienic quality of popular types of market vended freshly squeezed fruit juices and their risk to the consumers. The WSP system serving Mèze and Poussan (French Mediterranean coast) was constructed in 1980 and enlarged and upgraded from 1994 to 1998. Water quality along the waste stabilisation pond to (WSP) system has been monitored over the years, thus allowing us to assess the influence of enlargement and upgrading works. A significant enhancement of the average microbiological quality of the effluent was observed, with respective E. coli and streptococci average abatements of 4.1 and 3.4 log. units. Former seasonal variations of microbiological removal have vanished. The contribution of the different ponds to the disinfection performance of the WSP system was analysed. A microbiological quality model was proposed to evaluate the die-off kinetics related to the different ponds and as a tool for the design and management of WSP systems. Though the relationships between die-off coefficients and environmental factors appeared somewhat frail, this modelling is considered a promising approach for the prediction of WSP microbiological performance.

Freshly squeezed or freshly prepared fruit juices sold by local market vendors in Dhaka city were analyzed for the microbiological quality. Total viable bacterial counts, fungal counts, total coliform, faecal coliform and the presence of pathogenic microorganisms such as E. coli, Bacillus cereus, Staphylococcus aureus, Salmonella, Streptococcus were analyzed by standard methods. The total viable count of samples ranged from 3.00×102 to 9.60×108 and fungal counts ranged from 1.00×102 to 8.05×104. Out of 114 freshly prepared fruit juices samples collected 113 samples (99%) showed the presence of coliform and E. coli. The other bacteria like B. cereus, Staphylococcus aureus, Salmonella, Streptococcus were found in 64.91%, 6.14%, 7.89% and (5.26%) of the tested samples. The number and type of microorganisms recovered from the freshly squeezed fruit juices made them unsafe for drinking. It was concluded that due to unhygienic fruit handling in the unsanitary environmental conditions under which the vendors operate the juices become contaminated with harmful bacteria. The results of this study demonstrate the unhygienic quality of popular types of market vended freshly squeezed fruit juices and their risk to the consumers. Introduction: Analyzing the health risk associated with the microbial contamination of seawater is necessary to ensure that there is not any threat to human or environment. The aim of this study was to evaluate the microbiological quality of Caspian sea water using indicator bacteria. Some Physicochemical parameters were studied to assed their association with the contamination level of Caspian sea water due to the important role in the growth organisms in the seawater. Materials and Methods: In this study, 100 samples were collected from the northeastern zone of the Caspian sea, Iran, from November 2017 to December 2018. Total coliform, Fecal coliform, E.coli, Fecal Streptococcus and Clostridium perfringens were the indicator bacteria and temperature, pH, electrical conductivity, chloride and turbidity were water physicochemical factors according to standard methods. Results: The finding showed that the mean of Total coliform, Fecal coliform, E.coli, Fecal Streptococcus, and Clostridium perfringens were 614.72 ± 516.13, 62.11 ± 235.30, 49.69 ± 188.24, 348.02 ± 490.01 and 3.04 ± 5.76 MPN/100 ml, respectively. Furthermore, the mean and SD of temperature, pH, electrical conductivity, chloride, and turbidity were 13.84 oC ± 5.62, 8.27 ± 0.45, 17.96 ± 6.56 µs/cm, 5776.95 ± 1996.38 mg/l and 27.48 ± 15.82 NTU, respectively. Statistical analysis showed that microbial qualities were affected significantly by physicochemical factors, but the roles of water temperature were more than others. Conclusion: The microbiological results revealed that there was a remarkable level of contamination in some areas of Caspian sea. The results suggested that Clostridium perfringens provide better health risk prediction than other analyzed indicator bacteria, particularly in the warm season.

Microbial quality of reclaimed water used for irrigation in two golf courses located in the southern Iberian Peninsula (Spain and Portugal) was evaluated. Bacterial indicators for faecal pollution (total and faecal coliforms, Escherichia coli and enterococci) were tested by membrane filtration using appropriate selective media. In addition, somatic E. coli bacteriophages, enteric viruses (entero-, hepatitis A and rota-) and Legionella pneumophila were also analysed. The results obtained showed that all wastewater treatment processes reduced adequately the number of indicator microorganisms although a significant correlation between pathogenic and indicator microorganisms tested was not found. L. pneumophila was detected by PCR but not confirmed by culture. Survival experiments of pathogenic microorganisms in aerosols and irrigated turf are conducted to determine the health hazards for the golf practice and to propose a microbial standard for wastewater used for irrigation of golf courses.

ABSTRACT In this study, the microbiological quality of roof-harvested rainwater was assessed by monitoring the concentrations of Escherichia coli, enterococci, Clostridium perfringens, and Bacteroides spp. in rainwater obtained from tanks in Southeast Queensland, Australia. Samples were also tested using real-time PCR (with SYBR Green I dye) for the presence of potential pathogenic microorganisms. Of the 27 rainwater samples tested, 17 (63%), 21 (78%), 13 (48%), and 24 (89%) were positive for E. coli, enterococci, C. perfringens, and Bacteroides spp., respectively. Of the 27 samples, 11 (41%), 7 (26%), 4 (15%), 3 (11%), and 1 (4%) were PCR positive for the Campylobacter coli ceuE gene, the Legionella pneumophila mip gene, the Aeromonas hydrophila lip gene, the Salmonella invA gene, and the Campylobacter jejuni mapA gene. Of the 21 samples tested, 4 (19%) were positive for the Giardia lamblia β-giardin gene. The binary logistic regression model indicated a positive correlation (P < 0.02) between the presence/absence of enterococci and A. hydrophila. In contrast, the presence/absence of the remaining potential pathogens did not correlate with traditional fecal indicators. The poor correlation between fecal indicators and potential pathogens suggested that fecal indicators may not be adequate to assess the microbiological quality of rainwater and consequent health risk.

AbstractIntroductionThe purpose of this study was to determine the microbiological quality of food fish and its safety for consumers.Material and MethodsThe study included 24 fish representing grass carp, bighead carp, Siberian sturgeon, and wels catfish. Specimens were collected in winter. Aerobic bacteria, psychrophilic, Enterobacteriaceae, Staphylococcus spp., and E. coli counts were made, and the presence of Salmonella spp., L. monocytogenes, S. aureus, and other coagulase-positive staphylococci was investigated.ResultsThe microbiological analysis showed a similar level of aerobic, psychrophilic, and Staphylococcus spp. contamination of the four fish species. The Enterobacteriaceae count was higher in the muscles of grass carp and bighead carp than S. sturgeon and wels catfish. No pathogenic bacteria such as Salmonella spp., E. coli, L. monocytogenes, Staphylococcus aureus, or other coagulase positive staphylococci were found in samples of the examined fish species.ConclusionThe fresh fish examined in this study were of good microbiological quality and there was no health risk for consumers.
