Effect of blood platelets on heparin-induced clearing reaction

The relationship between the fall in optical density and the rise in glycerol concentration which occur during heparin-induced clearing was studied in platelet-rich and in platelet-poor post-heparin plasmas. This relationship as determined by the significance of the regression coefficients was highly significant with both plasmas. The effect of platelets on clearing was studied in three system containing either fasting plasma or post-heparin plasma or a mixture of both plasmas. In each system platelets inhibited clearing as measured by fall in optical density but did not inhibit lipolysis as measured by production of glycerol. Addition of platelet-poor fasting plasma to platelet-poor post-heparin plasma inhibited lipolysis but did not inhibit clearing. It is concluded that there may be at least two inhibitors against clearing, one associated with platelets and one present in plasma, which inhibit optical clearing and lipolysis, respectively. When fasting plasmas were incubated with substrate endogenous clearing occurred frequently, but sometimes a rise in optical density and a decrease in glycerol concentration were found, suggesting reversal of enzymatic processes.

The relationship between thickness of fat measured at the shoulder, last rib, and loin of live pigs and market weight (86.2 to 95.2 kg) was studied in 1596 pigs from the Lacombe and Yorkshire breeds and their reciprocal crosses. No significant breed or sex differences were found in the proportion of fat at the three sites. Breed and sex differences were evident in the average depth of fat. The regression coefficients for the mean of three fat measurements on market weight were.016 ±.003,.019 ±.007, and.017 ±.004 for Lacombe males, barrows, and females respectively;.039 ±.002,.036 ±.005, and.041 ±.001 for Yorkshire males, barrows and females respectively;.027 ±.005 and.029 ±.004 for barrows and females from the Lacombe male × Yorkshire female mating; and.023 ±.007 and.021 ±.009 for barrows and females from the Yorkshire male × Lacombe female mating. Comparisons between animals measured at a relatively constant weight should be made after the fat measurement is adjusted for variations in body weight. The results of this study indicate that separate corrections should be applied for each breed. Separate corrections for sexes within breeds do not appear warranted.

In linear regression modelling, the distortion of effects after marginalizing over variables of the conditioning set has been widely studied in several contexts. For Gaussian variables, the relationship between marginal and partial regression coefficients is well established and the issue is often addressed as a result of W. G. Cochran. Possible generalizations beyond the linear Gaussian case have been developed, nevertheless the case of discrete variables is still challenging, in particular in medical and social science settings. A multivariate regression framework is proposed for binary data with regression coefficients given by the logarithm of relative risks, and a multivariate Relative Risk formula is derived to define the relationship between marginal and conditional relative risks. The method is illustrated through the analysis of the morphine data in order to assess the effect of preoperative oral morphine administration on the postoperative pain relief.

Abstract BackgroundTriglyceride-glucose index (TyG index) has been regarded as a reliable alternative marker of insulin resistance. However, study on the relationship between TyG index and incident diabetes remains limited. This study aimed to investigate the association between TyG index and incident diabetes in a large cohort of Chinese population.MethodsThe present study was a retrospective cohort study using healthy screening programme data in China. A total of 201,298 subjects free of baseline diabetes were included who received a health check with all medical records from 2010 to 2016. TyG index was calculated as Ln[fasting triglyceride level (mg/dl) x fasting plasma glucose (mg/dl)/2]. Diagnosis of diabetes was based on fasting plasma glucose ≥ 7.00 mmol/L and/or self-reported diabetes. Cox proportion-hazard model was used to assess the relationship between TyG index at baseline and the risk of incident diabetes. It should be noted that the data was uploaded to the DATADRYAD website, and we only used this data for secondary analysis.ResultsDuring a mean follow-up of 3.12 years of 201,298 individuals aged ≥ 20 years old, 3389 subjects developed diabetes. After adjusting for age, sex, body mass index, systolic blood pressure, diastolic blood pressure, total cholesterol, low density lipoprotein cholesterol, alanine aminotransferase, aspartate aminotransferase, serum creatinine, smoking, drinking and family history of diabetes, multivariate cox hazards regression analysis indicated that TyG index was positive correlation with the risk of developing diabetes in Chinese population (HR, 3.34; 95% CI, 3.11 to 3.60). The risk of incident diabetes increased with increasing TyG index. Subjects with TyG index in the fourth quartile were 6.26 times more likely to develop diabetes than the lowest quartile (P trend < 0.001). Subgroup analysis showed the stronger association was observed in the population with age < 40, BMI (≥ 18.5, < 24 kg/m2), SBP < 140 mmHg or females (all P for interaction < 0.0001).Conclusions TyG index was independently correlated with the increased risk of diabetes in Chinese adults, suggesting that TyG index may be a useful marker for identifying individuals at high risk of developing diabetes.

A questionnaire survey was conducted to investigate eating behavior and the subjective symptoms of oral frailty, and to examine the relationship between them. A total of 744 subjects with ages over 65 years were included. The questionnaire comprised 18 question items indicating eating behavior and seven question items indicating oral frailty. All items were assessed according to 4 grades on a scale of 1 (not applicable) to 4 (applicable). The total score of oral frailty gradually increased with age. Regarding the scores for “eating recognition” and “eating habits”, no changes were observed, however the scores for “eating action” demonstrated a decreasing tendency with age and the scores of ≥ 85 years age group was significantly lower than the 65–69, 70–74, and 75–79 years age groups. As a result of multiple regression analysis, among the significant independent variable, the scores of “I do not chew foods well” under the category of “eating action” showed the highest standard partial regression coefficients for dependent variable of symptoms of oral frailty. The significant association was found between the eating behavior and subjective symptoms of oral frailty, and this study suggests that the good chewing habit might be an important criterion for the prevention of oral frailty.

An analysis has been carried out on the milk yield, milk composition and milking-rate records collected in one herd of dairy cows.It has been shown from the results of eighty-six heifers that for each pound per minute increase in inherent milking rate (i.e. peak flow) the standard lactation yield increases by 419 lb., the maximum daily yield by 0·9 lb., the lactation length by 8 days and the persistency index by 10 days, whilst the lactation solids-not-fat percentage decreases by 0·03 and the fat percentage by 0·04. All except the last coefficient were significant, although it was also shown that the relationship with solids-not-fat percentage was due to an interaction. Similarly, by the calculation of partial regression coefficients it was demonstrated that the relationship between milking rate and standard lactation yield was due to the effect of milking rate on maximum daily yield, lactation length and persistency.

Epidemiological studies suggest that a diet high in flavonoids protects against chronic diseases such as CVD and cancer. The objective of the present study was to evaluate the relationship between the intake of quercetin, kaempferol, isorhamnetin, apigenin and luteolin and their corresponding plasma concentrations, and further to explore whether these flavonoids can serve as biomarkers of their intake. Flavonoid intake and their plasma concentrations were analysed in ninety-two subjects consuming their habitual diet. Flavonoid intake was estimated with 7-d dietary records using available data on the flavonoid content of food. Plasma flavonoid concentrations were quantified by HPLC. In addition, we undertook a dietary intervention study to investigate plasma apigenin concentration after the consumption of celery leaf. The mean intake estimates of quercetin, kaempferol, isorhamnetin, apigenin and luteolin amounted to 13·58, 14·97, 12·31, 4·23 and 8·08 mg/d, respectively. The corresponding mean plasma concentrations were 80·23, 57·86, 39·94, 10·62 and 99·90 nmol/l. The mean 7 d intake of five flavonoids was positively correlated to their corresponding plasma concentrations, with correlation coefficients ranging from 0·33 to 0·51 (P < 0·05). In the dietary intervention study, the plasma apigenin concentration rose after celery leaf ingestion, and fell within 28 h to below the limit of detection (2·32 nmol/l). The present results suggest that quercetin, kaempferol, isorhamnetin, apigenin and luteolin are bioavailable from the diet. The levels of fasting plasma flavonoids seem to be suitable biomarkers of short-term intake. The combination of plasma flavonoids with their intake may prove useful when the possible health-protective effects of flavonoids are studied.
