Saffron, which has many kinds of biological activities, has been widely used in medicine, cosmetics, food, and other fields of health promotion industries. Crocins are the main component of saffron (Crocus sativus L.). At present, most of the extraction methods for crocins require long time or special instruments to complete the process and some of them are not suitable for industrial production at present. In this article, homogenate extraction technology which is a convenient and efficient method was developed for crocins extraction from saffron. Firstly, the influences of extraction voltage, extraction time, ethanol concentration, and temperature on crocins yield were studied by single factor experiments; and then response surface methodology (RSM) was used to optimize levels of four variables based on the result of single factor experiments. Results showed that the optimum extraction process conditions for crocins were as follows: extraction voltage, 110 V; ethanol concentration, 70%; extraction temperature, 57°C; and extraction time, 40 s. Based on these conditions, the extraction yield of crocins can reach 22.76% which is higher than ultrasonic extraction method. Therefore, homogenate extraction is an effective way to extract crocins from saffron with higher extraction yield and shorter extraction time.

Response surface methodology (RSM) was employed to optimize the extraction conditions of tannins from banana peels. Based on the single-factor experiments, four independent variables including ethanol concentration, solid to liquid ratio, extraction time and extraction temperature were selected as affecting factors. According to central composite design (CCD) the four factor, five levels experiments were carried out with the tannin extraction yield as evaluation index. The results show that the optimized conditions are as follows: ethanol concentration of 75.44%, solid to liquid ratio of 1:35.34, extraction time of 7.94 h, extraction temperature of 62.85°C. Under the optimized conditions, the predicted tannin extraction yield reaches 58.55%, while the actual extraction yield is 57.42% with the relative error of 1.13%.

This current study focuses on the modelling and optimization of supercritical fluid extraction of Quercus infectoria galls oil. In this case, response surface methodology (RSM) and artificial neural network (ANN) were applied for the modelling and prediction of extraction yield of galls oil. A 17-run Box-Behnken Design (BBD) was employed to statistically optimize the process parameters of SC-CO2 extraction of Quercus infectoria galls at a condition as follows: pressure (5000, 6000, 7000 Psi), temperature (40, 50, 60°C) and extraction time (30, 45, 60 min). The maximum yield of the extracted oil is1.12 % and the optimum conditions are at an extraction pressure of 5574 Psi; extraction temperature of 75°C and extraction time of 54 min. Under the optimal conditions, the experimental results agree with the predicted values obtained through analysis of variance (ANOVA). This indicates a successful response surface methodology and highly satisfactory goodness of fit of the model used. The analysis of experimental design for process optimization results demonstrates that temperature and extraction time are the main parameters that influence the oil extraction of Quercus infectoria.

Background: Plantago ovata Forsk (Psyllium seed) is an annual plant of plantago genus. This plant widely grows in India and Iran. The Psyllium seed contains mucilage, protein, sugar, fat and tannins. Seed’s husk is a known source to extract hydrocolloid. Objective: The aim of this study was to optimize gum extraction from the Psyllium seed. Methods: The response surface methodology (RSM) was used to estimate the effect of independent variables on the extraction yield of Psyllium gum. The obtained experimental data were fitted to a second- order polynomial equation using multiple regression analysis and analyzed by appropriate statistical methods. A central composite design (CCD) was used for experimental design and analysis of the results to obtain the best possible combination of extraction temperature (X1= 60-80°C), extraction time (X2= 1-3 h), and the ratio of water to raw material (X3= 30-70%) for the maximum gum extraction. Results: Optimum extraction conditions were extraction temperature of 79°C, extraction time of 2.5 h, and the ratio of water to the raw material of 57%. The experimental extraction yield under optimal conditions was found to be 9 ± 0.25%, which was in agreement with the predicted value of 9.4%. Conclusion: This study showed that Psyllium gum can be used as a hydrocolloid source for pharmaceutical and food industry such as edible films for food packaging.

By using extraction yield, total polyphenolic content, antidiabetic activities (PTP-1B and α-glycosidase), and antioxidant activity (ABTS and DPPH) as indicated markers, the extraction conditions of the prescription Kursi Wufarikun Ziyabit (KWZ) were optimized by response surface methodology (RSM). Independent variables were ethanol concentration, extraction temperature, solid-to-solvent ratio, and extraction time. The result of RSM analysis showed that the four variables investigated have a significant effect (p<0.05) for Y1, Y2, Y3, Y4, and Y5 with R2 value of 0.9120, 0.9793, 0.9076, 0.9125, and 0.9709, respectively. Optimal conditions for the highest extraction yield of 39.28%, PTP-1B inhibition rate of 86.21%, α-glycosidase enzymes inhibition rate of 96.56%, and ABTS inhibition rate of 77.38% were derived at ethanol concentration 50.11%, extraction temperature 72.06°C, solid-to-solvent ratio 1 : 22.73 g/mL, and extraction time 2.93 h. On the basis of total polyphenol content of 48.44% in this optimal condition, the quantitative analysis of effective part of KWZ was characterized via UPLC method, 12 main components were identified by standard compounds, and all of them have shown good regression within the test ranges and the total content of them was 11.18%.

Objective: To optimize the extraction parameters for determining the highest yield of Total Flavonoids from Gardenia gummifera gum resin.Methods: In the present study, response surface methodology (RSM) with three level Box Behenken design (BBD) was performed to optimize extraction parameters for total flavonoids. Solvent concentration(A), extraction time (B) and extraction temperature(C) were considered for single factor experiment.Results: The highest flavonoid concentration was obtained with acetone 45.00% v/v, time 101.46 min, temperature 41.57 °C. The average experimental TFC under optimal conditions was 161.14 mg/g which was in close agreement with the predicted value of 163.42 mg/g.Conclusion: RSM-BBD is successfully applied in optimizing extraction conditions for extraction of total flavonoids from G. gummifera gum resin.

Abstract In this study, the ultrasound-assisted extraction (UAE) was used for extraction of pinocembrin from Flos populi. Based on the results of the single-factor experiment, four independent parameters, including ethanol concentration (40–80 %), extraction temperature (50–70 °C), extraction time (25–45 min) and electrical acoustic intensity (40.8–81.5 W/m2) were further investigated using response surface methodology (RSM) coupled with Box-Behnken design. The experimental data were fitted to the quadratic response surface model using multiple regression analysis with high adjusted determination coefficient value (R 2) of 0.9697. The highest yield (134.2 ± 1.53 mg/g) of pinocembrin was obtained under the optimal conditions (ethanol concentration of 68 %, extraction temperature of 69 °C, extraction time of 42 min and electrical acoustic intensity of 66.81 W/cm2), which agreed to the predicted value of 132.9 mg/g. Moreover, the comparison between the UAE and reflux extraction also showed the suitability of UAE for pinocembrin from Flos populi.

In this study, a simultaneous water extraction process for baicalin, wogonoside, and chlorogenic acid has been optimized. The effect of extraction temperature, extraction time, and liquid–solid ratio was scrutinized by single factor experiments and further analyzed by Box–Behnken design (BBD) approach using response surface methodology (RSM). The extraction yield of investigated compounds was determined by high performance liquid chromatography (HPLC). Single-factor experiments and response surface analysis results revealed that the optimized conditions are: Liquid to solid ratio 25:1 (mL/g), extraction temperature 93 °C, extraction time 2.4 h, and the extraction cycle two. Importantly, it has been noted that under the above conditions, concentrations of baicalin, wogonoside, and chlorogenic were 0.078, 0.031, and 0.013 mg/mL, respectively, and the overall desirability (OD) value was 0.76 which was higher than the non-optimized conditions and the deviation from the predicted OD value was only 2.44%. Conclusively, it has been suggested that the model was stable and feasible, and fit for extraction of baicalin, wogonoside, and chlorogenic acid from Scutellariae Radix and Lonicerae (L.) japonicae Flos.

In this study we define the optimal conditions for ultrasound-assisted extraction of bioactive polyphenols from S. raeseri aerial parts using response surface methodology. The influence of ethanol concentration (10–90%), extraction temperature (20–80 °C), extraction time (10–60 min), and solid-to-solvent ratio (1:10–1:50) on total phenolic content as well as on content of individual flavonoids, and hypolaetin and isoscutellarein derivatives was studied. For the experimental design, a central composite design was chosen. In the obtained extracts, the following ranges of targeted compounds were detected: total phenol from 19.32 to 47.23 mg GAE/g dw, HYP from 1.05 to 11.46 mg/g dw, ISC 1 from 0.68 to 10.68 mg/g dw, and ISC 2 from 0.74 to 15.56 mg/g dw. The optimal extraction conditions were set as: ethanol concentration of 65%, extraction time of 50 min, extraction temperature of 63 °C, and solid-to-solvent ratio of 1:40. Contents of TP, HYP, ISC 1, and ISC 2 in optimal extracts were 47.11 mg GAE/g dw, 11.73 mg/g dw, 9.54 mg/g dw, and 15.40 mg/g dw, respectively. Experimentally set values were in good agreement with those predicted by the response surface methodology model, indicating suitability of the used model, as well as the success of response surface methodology in optimizing the conditions of the extraction.

The current study reports the antioxidant activity of Ocimum basilicum. Central composite design (CCD) was used to study the effect of variables on the extraction using magnetic stirrer (MST). Three independent variables including temperature, extraction speed, and extraction time were studied that optimize particular responses of total phenolic content, ferric reducing antioxidant power (FRAP), and DPPH free radical scavenging activity. The effect of the extraction temperature on extraction process was investigated in the range from 26.3oC to 93.6oC, extraction time, from 1 to 30 h, and extraction speed, in the range from 263 to 936 rpm. The extraction yield was significantly (P < 0.05) affected by all the variables. Temperature and extraction speed were found to have a marked effect while the extraction time was found to have possible minor effects. Graphical optimization determined the optimum conditions for the extraction. The optimum condition predicted an extraction yield of 20.49g/100g at 75.33oC for five hours at 73 rpm. Optimum conditions were determined to obtain highest extraction yield. Results showed that water/seed ratio was the most significant parameter, followed by temperature and time. All quantitative modeling and response surface methodology recommended that extraction temperature and time were the most effective parameters of MST process. However, extraction time was found out to be an insignificant factor in MST extraction of antioxidant and total phenolic compounds of O. basilicum.

A Box-Behnken experimental design and response surface methodology (RSM) was employed to optimize the parameters of ultrasonic extraction of phenolics from longan seed based on the single-factor experiments. The effects of extraction temperature, methanol concentration and extraction time on the extraction yield of phenolics from longan seed were investigated. The major parameter influencing the extraction was the extraction temperature. In this experiment, the maximal extraction yield of phenolics was obtained with 5.931 ± 0.016% (n = 3) at extraction temperature 82°C, methanol concentration 50% and extraction time 41 min. Under optimal conditions, the activity predicted by the model agreed well with experimental data.
