Description and molecular characterisation of Xiphinema herakliense n. sp. (Nematoda: Longidoridae) from wild and cultivated olives in Crete

Xiphinema herakliense n. sp. from wild olive and cultivated trees in Vathy Rema and other localities, Heraklion province of Crete, Greece, is described based on morphological and molecular characters. The new species is characterised by having differentiation in the tubular part of the uterus, body length 4.1 (3.3-5.5) mm, odontostyle 149 (135-163) μm long, odontophore 85 (71-98) μm long, female tail dorsally convex-conoid with subdigitate peg, and presence of functional males. The polytomous identification code of the new species is: A4, B2 + 3, C5a, D6(5), E5, F4(5), G3, H2, I3, J5, K5, L2. The new species belongs to morphospecies Xiphinema non-americanum Group 5 characterised by having two equally developed female genital branches with uterine differentiation, presence of pseudo-Z-organ with small granular bodies plus small spines (in low number) and crystalloid bodies along uterus, and a short, rounded tail ending in a peg. Xiphinema herakliense n. sp. appears closely related to X. barense, X. horvatovicae, X. lanceolatum, X. loteni, X. sphaerocephalum and X. thorneanum, from which it may be differentiated morphologically and/or molecularly. The results of the phylogenetic analyses based on the sequences of the D2-D3 expansion regions of the 28S rRNA gene and ITS1 rRNA genes are also supportive of its specific status.

A population of Xiphinema barense from wild olive trees in Torre Pozzella, Brindisi province, southern Italy, is described using both morphological and molecular studies and compared with the description of the type specimens. The wild olive nematode population agrees very well with all morphometrics provided in the original description. However, detailed observations of the lumen of the tubular portion of the uterus in paratypes and specimens of the new population revealed a clear pseudo-Z-organ with small granules mixed with crystalloid bodies which were previously undetected. Photomicrographs of adult paratypes, which were lacking in the original description, and of specimens of the new population from wild olive trees are provided. The results of the phylogenetic analyses based on the sequences of the D2-D3 expansion regions of the 28S rRNA gene and ITS rRNA genes confirm the species differentiation and indicate the phylogenetic position of X. barense and its relationship with closely related species.

A new species of the genus Nothotylenchus, N. persicus n. sp. was collected around the roots of grapevine and is described and illustrated herein based on morphological and molecular studies. The new species is characterised by a body length of 776-900 μm, delicate stylet 5-6 μm long, six lines in the lateral field, post-vulval uterine sac short, 10-18 μm long, female tail elongate-conoid with pointed terminus, and bursa covering 40-45% of tail length. Morphologically, N. persicus n. sp. appears closer to four known species of the genus, namely: N. hexaglyphus, N. affinis, N. medians and N. taylori. The results of phylogenetic analyses based on sequences of D2-D3 expansion region of 28S rRNA gene confirmed the close molecular relationship between N. persicus n. sp. and other anguinids, but Nothotylenchus claded separately from Ditylenchus species.

Diploscapteroides persicus n. sp. is described and illustrated based on morphological, morphometric and molecular data. The new species is mainly characterised by female body length of 469-673 μm, female tail length of 51-99 μm, shape of gubernaculum and spicules and arrangement of male genital papillae. Also, the arrangement of male genital papillae of the new species shows the presence of precloacal papillae, a feature that has not been previously described for the genus. The new species is comparable with all valid species of the genus, namely D. boettgeri, D. brevicauda, D. chitinolabiatus, D. coroniger, D. dacchensis and D. flexuosus, from which the morphological comparisons of the new species with aforementioned species are discussed. Phylogenetic analyses using partial sequences of SSU and LSU rRNA genes were performed for the new species, being the first representative of the genus to be phylogenetically studied using both Bayesian inference (BI) and maximum likelihood (ML) methods, and revealed that D. persicus n. sp. formed a clade with a species of Cephaloboides.

Summary Helicotylenchus ciceri n. sp. and H. scoticus are described and illustrated based on morphological, morphometric and molecular characters. The new species is characterised by a conical and truncated lip region with five or six distinct annuli, stylet 32-37 μm long with anteriorly concave knobs, secretory-excretory pore posterior to the pharyngo-intestinal valve, dorsally convex-conoid tail with a terminal projection, phasmids 14 (7-20) annuli anterior to the level of anus, empty spermatheca and absence of males. Intraspecific variation of 16 populations of H. scoticus, collected from chickpea and lentil fields in Kermanshah province, western Iran, is discussed. The results of the phylogenetic analyses based on the sequences of the partial 18S rRNA, D2-D3 expansion segments of 28S rRNA and ITS rRNA genes are provided for the studied species, confirming their differences from each other and determining the position of them and their relationships with closely related species.

Xiphinema granatum n. sp. from pomegranate in Saveh, Markazi province, central Iran, is described and illustrated. The new species belongs to species group 8 of the genus Xiphinema sensu Loof & Luc, 1990. The new species is characterised by a body 3.5-4.1 mm long, 37-49 μm diam., odontostyle and odontophore 118-132 and 65-74 μm long, respectively, presence of sperm in the female genital tracts, uterus devoid of any Z-differentiation or spines, and abundant males with spicules 45-58 μm long. The polytomous identification codes of the new species are: A4, B4, C5a, D5, E56, F4, G23, H2, I3, J4, K2, L2. Beside morphological and morphometric data, molecular analyses of D2-D3 fragments of 28S rRNA gene placed the new species in close relationship with X. vuittenezi and X. italiae and some species belonging to Xiphinema morphospecies group 6, but clearly separated from X. index. In addition, an Iranian population of Longidorus pisi from the same locality is morphologically and molecularly characterised.

Aphelenchoides eldaricusn. sp. is described and illustrated from wood and bark samples of a dead Mondell pine in Kermanshah Province, western Iran. The new species has a body length of 597 (507-700) μm (females) and 662 (636-695) μm (males). The cuticle is weakly annulated with three lateral lines and the lip region is offset. The stylet is 9.6 (9-10) μm long with small basal swellings and the excretory pore is locatedcaone body diam. posterior to the metacorpus valve. The spicules are large with the dorsal limb 40 (38-42) μm long with a prominent rostrum and rounded apex. The female tail is conical, tapering gradually to an acute terminus with a simple ventral mucron. The male tail bears six (2 + 2 + 2) caudal papillae and a well-developed mucron. The new species belongs to the Group 2 category ofAphelenchoidesspecies and is close to four known species, namelyA. cibolensis,A. indicus,A. rutgersiandA. sacchari. Molecular analyses of the partial 18S and the D2-D3 expansion segments of the 28S rRNA gene confirmed its morphological status as a new species.

During a survey conducted on banana plantations in Sistan and Blouchestan province, south-east Iran, a new species of Tylenchulus was extracted from the soil and roots of banana plants. This species, named Tylenchulus musicola sp. n., is characterised by mature females having a swollen, hook-shaped body with a conical and elongate post-vulval portion ending in a round terminus, males having a weak stylet and a cylindrical and thick tail ending in a bluntly rounded and smooth terminus, and by second-stage juveniles having a slender body and a posterior body portion ending in a finely pointed or mucronate terminus. The results of glasshouse host tests indicated that the new species does not parasitise sugarcane ratoons or sour orange seedlings. Tylenchulus musicola sp. n. is distinguished from other known Tylenchulus species by the sequences of D2-D3 expansion segments of 28S rRNA and ITS rRNA genes. Phylogenetic relationships within Tylenchulus were reconstructed based on rRNA gene sequences using Bayesian inference. Diagnostic PCR-ITS-RFLP profiles are presented for T. musicola sp. n., T. furcus, T. graminis, T. palustris, T. semipenetrans and Trophotylenchulus floridensis. PCR with species-specific primers and genus-specific primer are tested and developed for rapid identification of five Tylenchulus species. An identification key to Tylenchulus species is provided.

Aphelenchoides salixaen. sp. was isolated from wood and bark samples of a dead willow in Kermanshah Province, western Iran. The nematodes were collected directly from wood samples and were successfully reared onBotrytis cinerea. The new species has a body length of 567 (507-620) μm (females) and 382 or 400 μm (males). The cuticle is weakly annulated with four lateral lines. Lip region offset. The stylet is 9.5 (9-10) μm long with small basal swellings. The excretory pore is locatedcaone body diam. posterior to the metacorpus valve. The spicules are short with the dorsal limb 14 or 15 μm long (n = 2), apex and rostrum rounded and only slight offset. The female tail is cylindrical with a broadly rounded terminus. The male tail is conical, bearing six (2 + 2 + 2) caudal papillae with a hamate mucron at terminus. The new species belongs to the Group 1 category ofAphelenchoidesspecies and is close to seven known species, namely:A. capsuloplanus,A. confusus,A. limberi,A. obtusicaudatus,A. obtusus,A. orientalisandA. rotundicaudatus. Molecular analyses of the partial 18S, D2-D3 expansion segments of the 28S rRNA genes and internal transcribed spacer (ITS) supported it as a new species and that the new species is closest toA.besseyi,A. fujianensisandA. ritzemabosiin dendrograms inferred using 18S and 28S D2-D3 genes.

A new species of the genusDitylenchus,D.persicusn. sp., was collected around the roots of grapevine and is described and illustrated herein based on morphological and molecular studies. The new species is characterised by a body length of 783 (635-928) μm and 689 (670-715)μm in female and male, respectively, delicate stylet 6.2 (5.0-7.0)μm long, six lines in the lateral field, median bulb of pharynx well developed, muscular with crescentic valve, basal pharyngeal bulb pyriform with 10 (8-13)μm long stem extending into intestine, post-vulval uterine sac relatively short, 16 (14-18)μm long, female and male tail elongate-conoid with finely rounded terminus, and bursa covering 65-80% of male tail length. Morphologically,D.persicusn. sp. appears closer to five known species of the genus, namely:D.arachis,D.caudatus,D.clarus,D.myceliophagusandD. nanus. A Gram-stain-negative, aerobic, rod-shaped strain (R2A-3T) was isolated from the toxin-producing dinoflagellate Centrodinium punctatum and identified as a novel genus and new species based on a polyphasic taxonomic approach. The optimum conditions for growth of the strain were at 25 °C, pH 8.0 and in the presence of 3 % (w/v) NaCl. Phylogenetic analyses based on 16S rRNA genes and 92 core genes sets revealed that strain R2A-3T belongs to the family Nevskiaceae in the class Gammaproteobacteria and represented an independent taxon separated from other genera. The 16S rRNA gene of strain R2A-3T showed the highest sequence similarity to Polycyclovorans algicola TG408T (95.2%), Fontimonas thermophila HA-01T (94.1%) and Sinimarinibacterium flocculans NH6-24T (93.2%), and less than 92.8 % similarity to other genera in the family Nevskiaceae . The genome length of strain R2A-3T was 3608892 bp with 65.2 mol% G+C content. Summed feature 8 (comprising C18 : 1  ω7c and/or C18 : 1  ω6c) was the major fatty acid (>10 %). Diphosphatidylglycerol, phosphatidylglycerol and phosphatidylethanolamine were detected as the major polar lipids. The major respiratory quinone was ubiquinone-8. According to its phylogenetic, phenotypic, chemotaxonomic and genomic features, strain R2A-3T represents a new species in the new genus of the family Nevskiaceae . It is recommended to name it Flagellatimonas centrodinii gen. nov., sp. nov. The type strain is R2A-3T (=KCTC 82469T=GDMCC 1.2523T).
