Interference of Plasma Fluorophores on the Red Blood Cell Zinc Protoporphyrin : Haemoglobin Ratio as Determined on a Haematofluorimeter

Direct measurement of the zinc protoporphyrin:haemoglobin ratio (ZPP: Hb ratio) in blood samples is performed by using a haematofluorimeter. Interference by non-specific fluorophores can be eliminated by removing the plasma and making the measurement on washed red blood cells (RBCs). After re-suspending RBCs in isotonic saline, haematofluorimeter readings for the ZPP: Hb ratios revealed higher stability in the course of time whereas a good relationship was found with results obtained by application of an extraction method. Separate reference ranges were established for adult male and female subjects. After washing, the mean values calculated for ZPP: Hb ratios of subjects belonging to the reference groups demonstrated a reduction of 0·04 μmol ZPP mol Hb, corresponding with approximately 30%. In the patients' group, application of washing resulted in a variable decrease of ZPP: Hb ratios.

This study was carried out on clinical cases of retained fetal membranes in crossbred cows presented at College Clinics and College dairy farm and from Villages in and around Mhow. The blood samples were collected from jugular vein just before 12 hr. postpartum and on 7th day postpartum. Haematological profile revealed that the mean values of haemoglobin, neutrophil and monocyte count after 12 hrs and 7th day postpartum were significantly lower and lymphocyte count was significantly higher in RFM cows (n=18) than normally calved cows (n=6). The differences in mean TLC, eosinophil and basophil counts were non-significant at 12 hrs postpartum, however on 7th day postpartum the TLC and eosinophil count were significantly higher and basophil count was non-significantly different in RFM cows than the normally calved cows.

The Crioulo breed of horses performs in one of the most physically demanding equestrian competitions, the Marcha de Resistência, which is a contest in which the horses run 750 km in 15 days. The study's aim was to characterize the metabolic responses during this period. We evaluated eleven Crioulo horses in the competition, specifically, two males and nine females. Blood samples were collected 24 hours before the contest and on the 4th, 9th, 11th, 14th and 15th days of competition. We evaluated CK, AST, LDH, glucose, lactate, urea, creatinine, sodium, potassium, chloride, magnesium, total calcium, ionized calcium, total protein, hematocrit and the white blood cell count. At the end of the competition, the mean values of serum AST were 1151±358 IU/ L the mean LDH values were 7418±1695 IU/L and CK was 13,867±3998UI /L. There was a significant increase in urea, creatinine and lactate (p<0.0001). A decrease in the mean values of chloride, sodium, potassium, and total and ionized calcium was observed (p≤0.0002). An evaluation of the total leukocytes and segmented neutrophils (p≤0.0002) revealed their increased values, and decreased values were observed for hematocrit, plasma protein and total lymphocytes (p≤0.0003). The values of glucose, on average, remained constant. Based on these data, we conclude that the Marcha de Resistência competition necessitated a high muscular demand and the depletion of energy and electrolytes, suggesting an inflammatory process in the animals evaluated.

ObjectiveTo define reference ranges for the 3% oxygen desaturation index (DI3) in healthy children under 12 years old during sleep.DesignObservational.SettingHome.SubjectsHealthy children aged 6 months to 12 years of age.InterventionNocturnal pulse oximetry at home. Parents documented sleep times. Visi-Download software (Stowood Scientific) analysed data with artefact and wake periods removed.Main outcome measuresThe following oximetry parameters used in the assessment of sleep-disordered breathing conditions were measured: 3% (DI3) and 4% (DI4) oxygen desaturation indices—the number of times per hour where the oxygen saturation falls by at least 3% or 4% from baseline, mean saturations (SAT50), minimum saturations (SATmin), delta index 12 s (DI12s), and percentage time with saturations below 92% and 90%.ResultsSeventy-nine children underwent nocturnal home pulse oximetry, from which there were 66 studies suitable for analysis. The median values for DI3 and DI4 were 2.58 (95% CI 1.96 to 3.10) and 0.92 (95% CI 0.73 to 1.15), respectively. The 95th and 97.5th centiles for DI3 were 6.43 and 7.06, respectively, which inform our cut-off value for normality. The mean values for SAT50 and SATmin were 97.57% (95% CI 97.38% to 97.76%) and 91.09% (95% CI 90.32% to 91.86%), respectively.ConclusionIn children aged 6 months to 12 years, we define normality of the 3% oxygen desaturation index as <7 using standalone, motion-resistant pulse oximeters with short averaging times.

At the moment the suitability of stored red blood cells (sRBC) for transfusion is checked by routine methods such as haemoglobin estimation and the level of haemolysis. These methods cannot characterize directly the quality of the membranes of sRBC. The aim of this work is to assess the quality of sRBC based on such criteria as the membrane’s stiffness and the size and the form of sRBC. Materials and Methods. We have investigated 5 series of dry cytosmears of the sRBC which had been kept in blood bank in a period from 1 to 35 days. After AFM imaging, in every specimen, 5 RBC were chosen at random; the diameter, the height, and the stiffness were measured on each of them. Results. The present study shows high increase of the mean values of YM and height of RBC after 35 days of storage and decrease of the mean values of their diameter. Conclusion. Statistically significant high increase of the mean values of YM indicates the decrease of the elasticity of the cells in the course of storing of the RBC. This parameter along with the morphological characteristics can be used as criterion for assessment of applicability of the sRBC for blood transfusion.

Blood donation allowed by cell separators can offer higher performance and higher yield to guarantee better quality and pureness of collected products. New systems for the collection of platelet concentrate (PC) and packed red blood cells (PRBC) are currently available. The aim of our work was to test the possibility of preparing PC routinely from normal apheresis donors in a minimum amount of time while providing a second product. Over a 3-month period we performed 40 procedures using the Hemonetics MCS3P blood cell separator and the Dideco Excel. The mean values of platelet yield were 2.8 x 1011 (range 1.4-4.1) with the MCS3P and 3.49 x 1011 (range 2.9-3.9) with the Excel, in a plasma volume of 240 ml and 215 ml respectively; the PRBC units were added with SAG-Mannitol allowing a storage time of 42 days. Collection times were 71’ and 48’ respectively. Donor tolerance was analogous to phateletapheresis or plasmapheresis.

Eight adult Soay rams (four control and four cranially sympathectomized by removal of the superior cervical ganglia) were exposed to alternating 16-week periods of short days of 8 h light: 16 h darkness (8L : 16D) and long days (16L : 8D) for more than 3 years, and the changes in the diameter of the testes were recorded. Once during short days and once during long days blood samples were collected hourly for 28 h, and the concentrations of melatonin, prolactin, FSH, LH, testosterone, tri-iodothyronine (T3), thyroxine (T4) and cortisol were measured by radioimmunoassay. In the control rams the testes were reduced in size during long days compared to short days, and the blood concentrations (24-h mean values) of FSH, LH and testosterone were decreased. The levels of prolactin were much increased, while there was no significant change in the mean levels of melatonin, T3, T4 and cortisol. During both photoperiods there was considerable hour-to-hour variation in all eight hormones indicative of episodic secretion, as well as a consistent variation related to the time of day which was most pronounced for melatonin and T3. There was a clear difference in the daily profile of plasma melatonin levels between short and long days. In the superior cervical ganglionectomized rams there were no significant changes in the size of the testes or in the hormone titres between short and long days. Compared to the controls the plasma levels of LH, FSH, testosterone and prolactin were in the intermediate range. Some consistent diurnal variation was evident in the levels of all the hormones measured, with a pattern similar to the controls for a few of the hormones (e.g. T3) but quite different for others (e.g. melatonin).

Abstract INTRODUCTION: Platelet along with red blood cell count is a part of complete blood cell count, one of the most frequent laboratory tests in medicine. Platelet distribution width, plateletcrit and mean platelet volume are three indices provided by hematological analyzers. There are few reports in literature regarding the correlation of these three parameters with red blood cell parameters. AIM: Aim of this study is to investigate the correlation between these platelets parameters and red cell parameters: hematocrit, mean corpuscular volume and red blood cell distribution width. METHODS: Three hundred and three healthy blood donor volunteers (176 men and 127 women, mean age 37,3 years) were included. None of them had any known hematological disease in the past. The parameters mentioned above were measured by the automated hematological analyzer Coulter®LH780. RESULTS: The mean values for platelets were: PCT: 0,25±0,11%, MPV: 8,11±1,94 fL and PDW: 15,89±2,74%. The mean values for their parallel red blood cell parameters were: HCT: 40,55±2,63%), MCV: 91±4,17 fL, RDW: 13,3±1,35% Statistical and regression analysis including the correlation coefficient between platelet and red cell parameters as well as Student’s t-test was carried out. CONCLUSIONS: There seems to be no significant correlation between HCT and PCT. MCV and MPV were not correlated significantly as well, indicating that red blood cell and platelet sizes are independent. But there is a statistically significant correlation between RDW and PDW (r: 0,68, p<0,01) demostrating that anisocytosis of red blood cells and platelets might occur simultaneously. Based on these observations, further more studies should be carried out for the correlation between platelets and red blood cell indices in certain disorders.

AimsThe presence of cold agglutinin in blood samples can cause a spontaneous agglutination of red blood cells (RBCs) when low temperature occurs. This phenomenon causes a spurious lowering of RBC count on the automated haematological analysers that are detected by incongruous values (≥370 g/L) of the mean cellular haemoglobi concentration (MCHC). A preheating at 37°C can remove the RBC agglutination generally resulting in a reliable count. It has been reported that the same result can be reached by using the optical reticulocyte (RET) channel of Sysmex analysers where the RBC count is not influenced by the presence of cold agglutinin. This study aims to evaluate these data in a larger population, with regard to environmental conditions on Sysmex analysers. We have also evaluated the influence of different thermal pretreatments on the RBC count.MethodsThis study was performed on 96 remnants of peripheral blood samples (48 with MCHC in normal range and 48 with MCHC>370 g/L) which have been analysed in different preanalytical conditions on the Sysmex analysers.ResultsA preheating of samples at 41°C for 1 min leads to a reversibility of the cold agglutination comparable to the one observed in the RET channel and yields better results compared with 37°C for 2 hours.ConclusionsNone of described procedures assure the complete cold agglutination reversibility in every case. Consequently, since the haematological analysers not yet provide reliable parameters to confirm the complete resolution of agglutination, further verification of RBC count accuracy needs to be performed.

ABSTRACT The Nelore breed is the second largest bovine breed in the world and has actively participated in the expansion of new Brazilian agricultural frontiers. In this context, the purpose of this study was to determine the hematological and biochemical reference intervals of healthy Nelore matrices raised under an extensive regime without supplementation along southwest of Piauí state. Blood samples were collected from fifty-five multiparous female of the Nelore breed. Biochemical and hematological parameters were analyzed using a parametric statistical method with 95% CI of reference limits. The average values of red blood cells, hemoglobin as well as hematimetric indices showed reference ranges similar to reference standards. The hematocrit as well as granulocytes and agranulocytes presented alterations typical of animals raised in environments with higher temperatures. Mineral, enzymatic, protein and metabolic profiles were similar to other bovine breeds but with a narrower range of values. However, lower mean values were observed for levels of ionized calcium, total protein and urea. Nelore females present slightly different biochemical and hematological profiles from other breeds, which might result from the environmental and nutritional management applied and the natural deficiency of nitrogen, phosphorus and calcium in the region's pastures.

This study was conducted to determine the levels of serum total cholesterol in Hair goats in different gender and age ranges. For this purpose, the blood samples from 120 Hair goats, young (£ 6 months) and adult (2-4 years), which were found to be clinically healthy, were taken from the vena jugularis. Triglyceride, total cholesterol, HDL and LDL cholesterol levels were measured in sera samples. The mean values in adult female goats, young female goats, adult male goats and young male goats were found. Triglyceride levels were found to be higher in male goats than in females. It was observed that the HDL values obtained from male kids were lower than the other ones. The LDL results obtained from the goats were close to each other and statistically insignificant. Thus, in this study, the data obtained is considered to contribute to the formation of reference values in the Hair goats.n
