Salivary Cortisol Responses and Session Ratings of Perceived Exertion to a Rugby Match and Fatigue Test

This study compared the effects of an official rugby match and a fatigue test on the salivary cortisol responses of 13 rugby players. We also examined the relationship between this cortisol response and session ratings of perceived exertion (session-RPE). We collected saliva before and after the match and fatigue test and assessed physical effort intensity via session-RPE using a CR-10 scale. We measured cortisol concentration by enzyme-linked immunosorbent assays. Results were greater session-RPE and cortisol concentrations for the rugby match, compared with the fatigue test. There was a significant difference between cortisol concentrations obtained pre- and postmatch ( p < .022) and significant correlations between cortisol response and session-RPE sampling in both the rugby match ( r = .81; p < .001) and fatigue test ( r = .91; p < .001). This study provides evidence of greater perceived effort and higher cortisol concentrations in actual competition versus a fatigue test. Our data further support session-RPE as a relatively inexpensive close correlate of a stress biomarker (cortisol response). Thus, session-RPE can be used by coaches as a valid indication of training loads and adequate recovery time after exertion.

Abstract To investigate the effect of hypophyseal transection (HST) on GH secretagogue activity of the non-peptidyl GH secretagogue L-692,585 in the conscious pig, male castrated swine were randomly assigned to either a hypophyseal stalk transection group (HST; n=3) or to a sham-operated control group (SOC; n=3). Treatments administered were L-692,585 (100 γg/kg), human GH-releasing factor(1–29)NH2 (GRF; 20 γg/kg) or L-692,585 (100 γg/kg) + GRF (20 γg/kg) on days −7 to −3 before surgery and days +3 to +8 after surgery. To evaluate the integrity of the pituitary gland, the animals were challenged with corticotropin-releasing hormone (CRH; 150 γg) or GnRH (150 ng/kg) both before and after surgery. Blood was collected from −60 to +180 min post treatment and assayed for GH, cortisol and LH. Before surgery, no significant difference (P>0·05) in peak GH response (ng/ml) was present between the two groups (SOC vs HST) in response to L-692,585 (101 ± 12 vs 71 ± 9) or L-692,585 + GRF (171 ± 21 vs 174 ± 21). Only two out of three SOC vs three out of three HST pigs responded to GRF (13 ± 2 vs 25 ± 3) resulting in a significant difference between groups. Following surgery, significant differences were present in peak GH response (ng/ml) between SOC and HST groups following L-692,585 (79 ± 6 vs 13·8 ± 1·0); however, the response to L-692,585 + GRF was similar (115 ± 8 vs 94 ± 7). All animals responded to GRF; however, a significant difference was present between groups due to the magnitude of the responses. Whereas the cortisol responses (ng/ml) to L-692,585 in the SOC and HST groups were similar before surgery, a significant difference was present after surgery (44·4 ± 6·4 vs 14·6 ± 2·1). No significant difference was noted between the HST and SOC groups in response to CRH or GnRH either before or after surgery. These results indicated that L-692,585 induced an immediate GH response in the intact animal in contrast to GRF where the GH release was variable. L-692,585 also stimulated an immediate increase in cortisol levels. Transection of the hypophyseal stalk dramatically decreased but did not ablate the GH or cortisol response to L-692,585. Co-administration of L-692,585 + GRF induced an immediate GH response of similar magnitude in the intact and HST animal. We conclude that L-692,585 has a direct but limited action at the level of the pituitary and that an intact hypophyseal stalk is required for a maximal GH and cortisol response. L-692,585 acts with GRF at the level of the pituitary to induce a maximal GH response. These findings suggest that L-692,585 stimulates GH secretion by acting in combination with GRF and interrupting the inhibitory tone of somatostatin on the somatotroph. Alteration in cortisol response to acute social stressors has been hypothesized to mediate childhood adversities (CA) and increased morbidity in adulthood. However, the evidence supporting an association between CA and cortisol response to social stressors is inconclusive. The present review addressed this issue by reviewing the literature on CA and cortisol response to acute social stressors, with a focus on studies with adolescents or adults, using the Childhood Trauma Questionnaire (CTQ) to assess CA, and examining salivary cortisol response to the Trier Social Stress Test (TSST). Systematic searches of relevant articles in PsycINFO, PubMed, Web of Science and ScienceDirect in February and March 2020 identified 12 articles including 1196 participants with mean ages ranging from 15.3 to 52.3 yrs. across studies. CTQ scores were significantly associated with cortisol response in 2 studies. In addition, the physical abuse and emotional neglect subscales were associated with cortisol response respectively in 2 separate studies. The lack of association between CA and cortisol response calls for more longitudinal studies, and the use of formal records of maltreatment or informant reports in future research to complement information collected by retrospective measures. In addition, increased attention to biological mechanisms other than that associated with the regulation of cortisol in explaining the connection between CA and psychiatry morbidity is warranted.

The purpose of this study was to examine the effects of anticipated task duration on ratings of perceived exertion during treadmill running. Male subjects.(N = 15) completed two separate runs on a motor-driven treadmill at 85% V02 max. During one trial, subjects ran for a period of 20 minutes, while for a second trial, subjects were led to believe that they would be running for 30 minutes. In each case, the trials were terminated at the 20-minute mark. Ratings of perceived exertion, heart rates, respiratory rates, and ventilatory minute volumes were collected across each trial. Results supported the supposition that the anticipation of continued performance mediated ratings of effort expenditure. This effect was obtained only during moderate work levels and was in contrast to research examining mental fatigue.

Two studies were conducted to assess whether indoor video-assisted cycling influenced a person's quality of exercise (subjectively and quantitatively), compared to indoor cycling alone. In the first study 12 recreationally active subjects completed an initial test of VO2max, and three randomized trials of cycling at 70% VO2max (35 min.) watching a commercial cycling tape (cycle video), a test pattern displayed on the ergometer screen (blank video), or no video. Subjects' ratings of perceived exertion (RPE) and Affect were recorded, and heart rate and oxygen consumption (VO2) were measured during testing. The second study required 12 different subjects first to complete an assessment of VO2max and then two randomized trials (cycle video and no video) on a cycle ergometer where they freely set the intensity of their own exercise. Measurements of VO2, heart rate, blood lactate, power output, RPE, and Affect were recorded during testing. Results of Exp. 1 indicated that subjects' perceived effort equally between the two conditions, yet reported significantly ( p <.05) higher affect at 25 and 35 min. of cycling during the cycle video condition than no video condition. Results of Exp. 2 indicated that despite similar levels of blood lactate, subjects exercised at a significantly higher intensity during the cycle video condition compared to no video condition, with a higher VO2 and heart rate. The data support the use of indoor exercise videos to improve the exercise experience and also to increase the physiological demands of indoor exercise.

Koalas (Phascolarctos cinereus) are one of Australia’s most charismatic native small marsupial species. Unfortunately, populations of koalas are rapidly declining throughout Australia as they continue to face increasing pressure from a changing ecosystem. All wildlife species to some degree will use their hypothalamic–pituitary–adrenal (HPA) axis in response to stress. Depending on the duration of activation, the stress response can lead to either acute or chronic side effects and is modulated through the neuroendocrine stress system with the release of catecholamines and glucocorticoids (e.g., cortisol). It is well known that rehabilitation sanctuaries are inherently stressful for all animals, in particular for rescued wild koalas, as it is an unfamiliar environment where the animals cannot predict or control what will happen to them. In this pilot study, we set out to quantify faecal and fur cortisol metabolites in wild rescued koalas undergoing wildlife rehabilitation. Absolute levels of acute and chronic stress were indexed non-invasively, with faecal samples taken to evaluate acute stress, and fur samples taken to evaluate chronic stress. Sampling occurred sporadically over four months (the start of September 2018 to the end of December 2018), and was performed on three rescued koalas (Maree, Tai, and Solstice) being held at the rehabilitation centre. Results of this study show that between the three koalas, the highest recorded faecal cortisol result was 241 ng/g, and the lowest recorded faecal cortisol result was 4 ng/g, whereas the highest recorded fur cortisol result was 1.75 ng/g, and the lowest recorded fur cortisol result was 0.10 ng/g. Statistically, there was a significant difference between all three koalas and their faecal cortisol responses, as well as their fur cortisol responses. Statistically for Maree and Solstice, there was a significant difference in their faecal cortisol response between days when a stressor was recorded, and days when a stressor was not recorded. However, statistically for Tai, this was not the case, as there was no significant difference in his faecal cortisol response between days when a stressor was recorded, and days when a stressor was not recorded. In summary, the hypothesis that faecal glucocorticoids and fur glucocorticoids between koalas will differ based on individual responses to stressors was true as a whole, but individually, this hypothesis was true for Maree and Solstice, but untrue for Tai. The use of biological samples such as faeces and fur to obtain readings of glucocorticoids is a method of measuring absolute levels of physiological stress that is still evolving for koalas, and there is no current glucocorticoid baseline with which to compare the results of this study; although, measuring faecal and fur glucocorticoids is the first step in understanding how koalas undergoing wildlife rehabilitation respond to stressors.

Adults are biologically responsive to context, and their responses to particular situations may differ across cultures. However, are preschoolers’ biological systems also responsive to situational contexts and cultures? Here we show that children’s neurobiological stress responses, as indexed by salivary cortisol, are activated and responsive to psychosocial stressors relevant to their socio-cultural emphases. By examining cortisol changes across different contexts among 138 preschoolers living in the United States, China and Japan, we found that an achievement-related stressor elicited an increased cortisol response among Chinese preschoolers, whereas interpersonal-related stressors elicited an increased cortisol response among Japanese preschoolers. Contrastingly, US preschoolers showed decreased cortisol responses after these stressors, but consistently showed higher levels of anticipatory responses to separation at the beginning of each session. Our findings suggest that children’s neurobiological stress systems may be a critical biological mechanism allowing societal-level cultural phenomena to be embodied in individual-level responses, even amongst preschoolers.

The short Synacthen (tetracosactrin) test (SST) is an established method of assessing adrenocortical reserve, and is increasingly replacing the insulin tolerance test (ITT) for the assessment of the hypothalamic-pituitary-adrenal (HPA) axis. However, there is no consensus on how the test is performed, and various time points and routes of administration are used. The present study was done, first, to determine whether there was any difference in cortisol responses when Synacthen was administered intramuscularly compared with intravenously and, secondly, to compare cortisol responses at 30 and 60 min. We found no significant difference between the two routes of administration. However, cortisol responses at 60 min were significantly higher than at 30 min ( P < 0·05). Previous validations for the use of the SST in place of the ITT have used cortisol responses 30 min after Synacthen. We conclude that where the SST is used to assess the HPA axis, cortisol response at 30 min after intravenous Synacthen should be used.

Background: Functional Fitness Training (FFT) is a new exercise modality prioritizing functional multi-joint movements executed at high intensity as a circuit. Objective: To examine the impacts of introducing rest intervals in a FFT workout compared to “rounds for time” (RFT) FFT. Materials and Methods: Participants were 25 resistance-trained adults who completed two FFT workouts 1 week apart. The study design was crossover such that in a given session half the participants completed the standard and the other half the adapted FFT (FFTadapted). The workouts consisted of the same exercises (circuit of four rounds of exercises), but one (FFTadapted) included preset rest intervals (three sets of 1 min after each completed round). Before and after the workouts, countermovement jump ability and blood lactate were measured. Heart rate (HR) and ratings of perceived exertion (RPE) were measured post-exercise. Results: For both the standard and adapted protocols, mean HR was 90% age-predicted maximum. Final RPE was also similar for both workouts (~15–15.5) and indicated a “hard” work intensity. Both FFTs took the same time to complete (~13 min). Furthermore, no significant differences were observed in jump ability between FFTs. In contrast, lactate (15.11 ± 3.64 vs. 13.48 ± 3.64 mmol·L−1, p < 0.05), measured 3 min post-exercise, was significantly lower in FFTadapted. Conclusions: In FFTadapted, there was a significant reduction in RPE and blood lactate concentrations after exercise, while there were no significant differences in either HR or jumping ability, compared to a FFT workout in RFT methodology.

Abstract Salivary cortisol increases in response to stressors, including physical exertion and psychological stress associated with sports competition. In addition, stress may induce change in brain-derived neurotrophic factor (BDNF). However, there are still no data available to compare the salivary BDNF level in sedentary male individuals and elite team-sport male athletes, regularly involved in activities that require elevated attention and concentration. This information could contribute to the advance of understanding of the effect of regular exercise on the salivary level of BDNF, the pre-to-post change in salivary BDNF during exercise, and the association between salivary cortisol and salivary BDNF responses to physical exercise. Therefore, this study aimed to compare the concentration of salivary cortisol and BDNF, before and after exercise, in sedentary individuals and elite male basketball players. The sedentary group (23.0 ± 4.2 yrs) performed a high-intensity exercise protocol and the basketball players (18.6 ± 0.5 yrs) participated in three official basketball matches. Saliva samples were tested for cortisol and BDNF using ELISA. A significant increase in salivary cortisol from pre- to post-match was observed only for the basketball players (p < 0.05). Basketball players also presented a higher salivary BDNF concentration for both resting (pre) and post-physical exercise (p < 0.05); however, no change in pre- to post-exercise salivary BDNF was observed for either group (p > 0.05). Elevated BDNF in athletes may be associated to their repeated exposure to stressful competition situations. The current findings also suggest that different mechanisms might be involved in salivary cortisol and BDNF responses during physical exercise.
